Smoking and environmental tobacco smoke exposure: implications in ocular disorders.

PURPOSE: This article aims to gather and review the available knowledge on several implications of smoking and environmental tobacco smoke (ETS) exposure in ocular disorders and provides pathomechanistic insights where applicable. MATERIALS AND METHODS: PubMed and Scopus databases were searched for relevant studies on the association of smoking and ETS exposure with various ocular disorders. Studies with different evidence levels, e.g., in-vivo, case-control, cohort, and meta-analysis, were included. RESULTS: Smoking is an established, modifiable risk factor in several ocular diseases, inclu

Prenatal and postnatal mercury exposure, breastfeeding and neurodevelopment during the first 5 years.

OBJECTIVE: We evaluated the association between infant hair-Hg and Gesell schedules (GS). BACKGROUND: Longitudinal assessment of prenatal and postnatal Hg exposure during the first 60 months. METHODS: We used hair-Hg as a marker of postnatal Hg exposure (inorganic and methyl-Hg from breast milk, and ethyl-Hg from thimerosal) and GS measured at 6, 36, and 60 months. RESULTS: Hair-Hg at 6 months responded to events related to Hg exposure and breastfeeding. However, most neurodevelopment delays observed at 6 months were overcome with infant growth; at 60 months 87% of children showed adequate

Do Genetic Variants Modify the Effect of Smoking on Risk of Preeclampsia in Pregnancy?

OBJECTIVE: Maternal smoking is associated with as much as a 50% reduced risk of preeclampsia, despite increasing risk of other poor pregnancy outcomes that often co-occur with preeclampsia, such as preterm birth and fetal growth restriction. Researchers have long sought to understand whether this perplexing association is biologically based, or a result of noncausal mechanisms. We examined whether smoking-response genes modify the smoking-preeclampsia association to investigate potential biological explanations. STUDY DESIGN: We conducted a nested case-control study within the Norwegian Mothe

Associations between Prenatal Exposure to Perchlorate, Thiocyanate, and Nitrate and Child Neurodevelopment in the MARBLES Study.

Associations between prenatal exposure to perchlorate, thiocyanate, and nitrate and autism spectrum disorder (ASD) remain largely unexplored. We investigated these associations in the MARBLES cohort, a prospective cohort with an elevated familial likelihood of ASD. A total of 310 mothers provided 1916 urine samples during pregnancy, and their children were classified at age 3 years as typically developing (TD), ASD, or non-TD (developmental concerns not meeting ASD diagnostic criteria). We estimated relative risks for ASD and non-TD compared with TD using Poisson regression and assessed mixtur

Developmental bisphenol A exposure alters adipose metabolism and lipid remodeling in female mouse offspring.

Bisphenol A (BPA), an endocrine-disrupting chemical widely detected in human populations, has been implicated in metabolic disorders following early-life exposure. However, the long-term effects of developmental BPA exposure on adipose metabolism under post-weaning dietary challenge remain incompletely understood. Pregnant C57BL/6J mice received BPA in drinking water at a maternal nominal target exposure of 50 μg/kg/day from gestational day 6 to postnatal day 21. After weaning, offspring were fed a normal diet or high-fat diet and evaluated at PND22, PND36, and PND91. Body weight, adipose morp

Fetal Fentanyl Syndrome: An Opportunity to Quantify the Risk in Prospective Cohorts of Patients With Substance Use Disorder.

The increased availability of fentanyl in the United States over the past decade extends to women of reproductive age, raising concern about maternal and fetal risks when used in pregnancy. A 2023 case series of 10 infants exposed to fentanyl in utero proposed a "fetal fentanyl syndrome" (FFS), characterized by distinctive craniofacial features, limb anomalies, and genitourinary defects. However, as with any case series, concerns about selective case ascertainment, uncontrolled confounding, imprecise timing of exposure, phenotype heterogeneity, and lack of a denominator preclude causal inferen

Sun Protection Behaviours and Photosensitivity in Japanese Children: Toyama Adjunct Study of Japan Environment and Children's Study (JECS).

Children have up to three3 times the annual sun exposure of adults. This cross-sectional study evaluated sun protection behaviorbehaviour, related factors, and the incidence of photosensitivity in Japanese children. A self-administered questionnaire administered to mothers of second2nd-grade children as part of the Toyama Adjunct Study of the Japan Environment and Children's Study was used and analyzedanalysed (n=1,691). The results showed that 67.5 % of children always or sometimes used sunscreen when going out for a long time, while 73.2 % used at least one method of sun protection when swim

Early Life Microplastic Exposure Impairs Mouse Incisor Enamel Formation.

Microplastics (MPs) are pervasive environmental contaminants with growing evidence of systemic toxicity; however, their effects on tooth development during early life remain poorly understood. In the present study, we investigated whether early-life exposure to polystyrene microplastics (PS-MPs) affects tooth eruption, root formation, and enamel development in mice. Pregnant mice were administered 1, 4, and 10 μm PS-MPs (10 μg/kg/day) from gestation through lactation, and offspring continued exposure post-weaning. Mandibles were analyzed at postnatal days 14, 25, and 42 using micro-computed to

Prenatal exposure to polycyclic aromatic hydrocarbons, reduced hippocampal subfield volumes, and word reading.

Reading difficulties and exposure to air pollution are both disproportionately high among youth living in economically disadvantaged contexts. Critically, variance in reading skills in youth living in higher socioeconomic status (SES) contexts largely derives from genetic factors, whereas environmental factors explain more of the variance in reading skills among youth living in lower SES contexts. Although reading research has focused closely on the psychosocial environment, little focus has been paid to the effects of the chemical environment. In this study, we measured prenatal exposure to a

Intrauterine exposure to omeprazole increases the risk of teeth morphological anomalies in the offspring of a murine model.

Conditions experienced in early life have long-lasting effects on offspring health. Despite this, little is known about how maternal exposure to drugs during pregnancy affects offspring teeth morphogenesis. In humans, omeprazole is a common drug used to mitigate Gastroesophageal Reflux Disease. Importantly, omeprazole is a non-specific proton-pump inhibitor, which may inhibit the proton pumps expressed in the developing tooth germ. To date, however, the effects of intrauterine life exposure to omeprazole on offspring tooth development remain unknown. In this study, we addressed this gap in a m…

Traffic-Related Air Pollution and Child BMI-A Study of Prenatal Exposure to Nitrogen Oxides and Body Mass Index in Children at the Age of Four Years in Malmö, Sweden.

Traffic-related air pollution could be a danger to the health of children. Earlier studies have linked prenatal exposure to an increased risk of a range of diseases and negative health outcomes, including overweight and obesity. Presently, a knowledge gap exists in investigating the risk of overweight and obesity among children exposed to lower levels of air pollution in utero. This study aimed to investigate the relationship between prenatal traffic-related air pollution (nitrogen dioxides (NOx) and traffic density) and childhood overweight and obesity in Malmö, Sweden. A cohort, based on att

Does the average drug exposure in pregnant women affect pregnancy outcome? A comparison of two approaches to estimate the baseline risks of adverse pregnancy outcome.

PURPOSE: The results of observational cohort studies on drug effects on pregnancy outcome may depend among others on suitable comparison cohorts. The aim of this investigation was to compare two distinct definitions of maternal exposure status for comparison cohorts. METHODS: We performed an observational cohort study of prospectively ascertained pregnant women who spontaneously contacted the Teratology Information Service (TIS) Berlin for drug risk consultation. The only exclusion criteria were exposures to established teratogens and/or fetotoxicants. Pregnancy outcomes of 3250 women with th…

In utero exposure to valproic acid and autism--a current review of clinical and animal studies.

Valproic acid (VPA) is both an anti-convulsant and a mood stabilizer. Clinical studies over the past 40 years have shown that exposure to VPA in utero is associated with birth defects, cognitive deficits, and increased risk of autism. Two recent FDA warnings related to use of VPA in pregnancy emphasize the need to reevaluate its use clinically during child-bearing years. The emerging clinical evidence showing a link between VPA exposure and both cognitive function and risk of autism brings to the forefront the importance of understanding how VPA exposure influences neurodevelopment. In the pas

The role of gut microbiota in fetal methylmercury exposure: Insights from a pilot study.

PURPOSE: The mechanisms by which gut microbiota contribute to methylmercury metabolism remain unclear. Among a cohort of pregnant mothers, the objectives of our pilot study were to determine (1) associations between gut microbiota and mercury concentrations in biomarkers (stool, hair and cord blood) and (2) the contributions of gut microbial mercury methylation/demethylation to stool methylmercury. METHODS: Pregnant women (36-39 weeks gestation, n=17) donated hair and stool specimens, and cord blood was collected for a subset (n=7). The diversity of gut microbiota was determined using 16S rRN

Tracking of [14C]Polystyrene Nanoplastics in Pregnant Mice.

Micro- and nanoplastics (MNP) are increasingly reported in the reproductive and maternal organs and tissues, yet their translocation and accumulation during pregnancy remain poorly quantified - often the material used as MNP is heavily modified. Herein, we have used unmodified polystyrene (PS) nanoparticles of three sizes (33 ± 11, 246 ± 81, and 1052 ± 189 nm) with covalent 14C backbone labels to track biodistribution in late-gestation mice after intranasal (IN) or intravenous (IV) administration. Tissue collection after administrations revealed exposure route-dependent nanoplastic accumulatio

Risk and burden of adverse birth outcomes associated with flood exposure in China: A 4.4 million mother-infant cohort study.

The risks and attributable burden of adverse birth outcomes from flooding remain unclear. Here, we analyzed a nationwide cohort of over 4.4 million mother-infant pairs in China, matching a spatiotemporal flood database with multiple buffer zones centered on maternal delivery hospitals to assess the health risks of multi-window flood exposure before and during pregnancy. We found that flood exposure during the three months preconception and the first trimester significantly increased the risks of preterm birth (PTB), low birth weight (LBW), small for gestational age (SGA), and term low birth we

High-temperature exposure and risk of spontaneous abortion during early pregnancy: a case-control study in Nanjing, China.

As one of the most common complications of early pregnancy, spontaneous abortion is associated with environmental factors, but reports estimating the effect of ambient temperature on spontaneous abortion are still inconclusive. Herein, a case-control study (1002 cases and 2004 controls) in Nanjing, China, from 2017 to 2021 was conducted to evaluate the association between temperature exposure and the risk of spontaneous abortion by using distributed lag nonlinear model (DLNM). As a result, daily mean temperature exposure and early spontaneous abortion showed a nonlinear relationship in 14-day
