GPR40 signaling in agouti-related peptide neurons mediates fat preference.

AIMS: Fat preference is mediated by fatty acid receptors in the oral, intestinal, and central nervous systems, but their central nervous system roles remain unclear. Here, we investigated how GPR40, a medium- and long-chain fatty acid receptor, regulates fat preference via agouti-related peptide (AgRP) neurons in the hypothalamic arcuate nucleus (ARC). MATERIALS AND METHODS: AgRP neuron-specific Gpr40 knockout mice were generated to investigate the role of GPR40 in dietary fat preference. Behavioral tests were conducted to assess dietary preferences, and metabolic analyses were performed afte

Developing a functional non-animal CNS stress model utilizing long-term potentiation with human iPSC-cortical neurons to evaluate therapeutics.

Cortisol, the main stress hormone of the hypothalamic-pituitary-adrenal (HPA) axis, has been hypothesized to cause considerable detriment to cognitive function in both a time and concentration-dependent manner. However, there is a current lack of functional in vitro models available to evaluate the stress condition. Long-term potentiation (LTP) has served as a quantitative correlate for memory and learning through in vitro neuronal network responses to electrical stimuli, where a high-frequency stimulation (HFS) protocol on microelectrode arrays (MEAs) evaluates synaptic integrity related to h

The NLRP3-P2X7 Axis and Cytokine Crosstalk in Alzheimer's Disease: Mechanisms, Implications, and Therapeutic Opportunities.

Alzheimer's disease (AD) is the primary cause of dementia in elderly individuals, characterized by progressive memory loss, cognitive decline, and impaired daily functioning. Pathologically, AD is associated with the accumulation of amyloid-β (Aβ) plaques, tau tangles, mitochondrial dysfunction, and chronic neuroinflammation. The activation of the NOD-like receptor pyrin domain- containing 3 (NLRP3) inflammasome by Aβ clusters triggers microglial activation, leading to a cascade of inflammatory responses. Similarly, tau tangles stimulate neuronal and glial cells, further amplifying NLRP3 activ

Forecasting daily bathtub-drowning mortality in Japan: a comparative analysis of statistical, machine learning, and deep learning approaches.

BACKGROUND: Japan reports the highest global mortality rate from drowning among older adults, predominantly owing to bathtub-related incidents. Despite sustained public health interventions, this mortality has increased over several decades. Timely warnings advising older adults to avoid unsupervised bathing during high-risk conditions may mitigate this issue; however, no nationwide forecasting model currently exists. METHODS: We integrated death certificate records from 1995-2020 (99,930 bathtub-drowning deaths) with meteorological, temporal, and demographic data across all 47 prefectures (4

Immunopathogenesis of polymorphic light eruption, role of cytokines, effector and regulatory T-cells and its therapeutic implications.

Polymorphic light eruption (PMLE) is mediated by a type IVc delayed-type hypersensitivity (DTH) reaction to ultraviolet radiation (UVR) induced neoantigens. While UVR normally exerts an immunosuppressive effect with increased regulatory T-cells (Tregs) and an accentuated T helper 2 (Th2) response, PMLE shows a heightened Th1 response. Existent data in PMLE suggests a Th1-skewed immune response with reduced Treg numbers and function, lack of Langerhans cells activity, and increased CD8+ resident memory T-cells. Keratinocytes (KCs) contribute to inflammation by releasing interleukin (IL)-1β, Vas

Research on Deep Learning-Based Human-Robot Static/Dynamic Gesture-Driven Control Framework.

For human-robot gesture-driven control, this paper proposes a deep learning-based approach that employs both static and dynamic gestures to drive and control robots for object-grasping and delivery tasks. The method utilizes two-dimensional Convolutional Neural Networks (2D-CNNs) for static gesture recognition and a hybrid architecture combining three-dimensional Convolutional Neural Networks (3D-CNNs) and Long Short-Term Memory networks (3D-CNN+LSTM) for dynamic gesture recognition. Results on a custom gesture dataset demonstrate validation accuracies of 95.38% for static gestures and 93.18%

Comparison of deep LSTM and machine learning models for predicting compressive strength of fly ash/slag-based geopolymer concrete.

In the production of geopolymer concrete (GPC), using ground granulated blast furnace slag (GGBFS) and fly ash (FA) can reduce the carbon dioxide footprint and decrease the amount of waste materials released into the environment. Finding the compressive strength (fc) of GPC through experiments is time-consuming and costly; thus, applying artificial intelligence models can expedite this process. This study aims to compare the performance of deep Long Short-Term Memory (LSTM) and the machine learning (ML)-based algorithms in predicting the fc of FA/GGBFS-based GPC. Artificial neural networks (AN

Incremental learning for acute lymphoblastic leukemia classification based on hybrid deep learning using blood smear image.

The prevalent type of blood cancer is called leukemia, which is caused by the irregular production of immature malignant cells in the bone marrow. This dangerous condition weakens the immune system, making the body susceptible to infections, and can lead to death if not treated quickly. Thus, immediate treatments are necessary to detect leukemia at the initial stage to control abnormal cell growth. Leukemia detection from microscopic images of blood smears of malignant leukemia cells is a time-consuming and tedious task. Thus, a Tangent Sand Cat Swarm Optimization-Long Short-Term Memory-LeNet

Fading suppression method based on redundant data within the spatial resolution and deep learning for a Φ-OTDR system.

Weak light intensity positions induced by interference fading adversely affect the sensing performance of phase-sensitive optical time-domain reflectometry (Φ-OTDR). Most effective fading suppression methods rely on frequency or phase modulation of the light source, which requires complex hardware modifications. To solve the above issue, this paper proposes a novel multi-channel data synthesizing method based on deep neural network (MDS-DNN) to reduce the impact of interference fading on the signal-to-noise ratio (SNR) of Φ-OTDR. The proposed algorithm can work efficiently without any modifica

Multifunctional nanoparticle-mediated targeting of metabolic reprogramming and DNA damage response pathways to treat drug-resistant triple-negative breast cancer.

Multi-drug resistance and immunosuppressive triple-negative breast cancer (TNBC) is triggered by the Warburg effect, which promotes homologous recombination repair (HRR) and upregulates expression of P-glycoprotein (P-gp), in turn preventing DNA damage from chemotherapy and creating an immunosuppressive microenvironment. It is therefore of clinical relevance to develop an effective delivery system that targets metabolic reprogramming and DNA damage response pathways for the treatment of drug-resistant TNBC. Herein, a P-gp-inhibiting and GSH-responsive multifunctional drug carrier targeting int

Changed detection: No relationship between working memory and media usage in Covid-era and contemporary young adults.

A decade ago, the widespread adoption of smartphones prompted research assessing the effects of media multitasking on cognitive performance. Early findings showed that high media multitasking imposed a significant cost to attention and executive function. Here, we revisited whether the harmful effects of media multitasking persist in digital native undergraduates, in whom nearly all media usage involves multitasking. We investigated the relationship between media usage and working memory (WM) performance. Across six experiments, we conducted online and in-person testing, using simultaneous or

A one year longitudinal study of cortical myelination changes following pediatric mild traumatic brain injury.

The impact of pediatric mild traumatic brain injury (pmTBI) on cortical (i.e., grey matter) myelination is not yet understood, especially for interactions with neurodevelopment. The current study examined the impact of pmTBI on cortical myelination relative to healthy controls (HC) by estimating myelin content using the T1w/T2w ratio method. Data were obtained from pmTBI (N = 217) participants at approximately 7 days (Visit 1 [V1]), 4 months (Visit 2 [V2]), and 1 year (Visit 3 [V3]) post-injury, with equivalent sampling points for age and sex-matched HC (N = 180). Clinical results suggested on
