Spinal muscular atrophy (SMA) is a severe neuromuscular disease characterized by loss of spinal alpha-motor neurons, resulting in the paralysis of skeletal muscle. SMA is caused by deficiency of survival motor neuron (SMN) protein levels. Recent evidence has highlighted an axon-specific role for SMN protein, raising the possibility that axon degeneration may be an early event in SMA pathogenesis. The Wallerian degeneration slow (Wld(s)) gene is a spontaneous dominant mutation in mice that delays axon degeneration by approximately 2-3 weeks. We set out to examine the effect of Wld(s) on the phe…

Spinal muscular atrophy is an autosomal recessive neurodegenerative disease characterised by degeneration of spinal cord motor neurons, atrophy of skeletal muscles, and generalised weakness. It is caused by homozygous disruption of the survival motor neuron 1 (SMN1) gene by deletion, conversion, or mutation. Although no medical treatment is available, investigations have elucidated possible mechanisms underlying the molecular pathogenesis of the disease. Treatment strategies have been developed to use the unique genomic structure of the SMN1 gene region. Several candidate treatment agents have…

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative motor neuron disease. The etiology is likely multifactorial, involving both genetic and environmental factors.
METHOD: Literature review.
FINDINGS: Motor neuron death is believed to arise from mutations in superoxide dismutase 1. Abnormal neurofilament metabolism, glutamate transporter dysfunction, and altered responses to growth factors may play a role. These discoveries have highlighted possible interventions, several of which have subsequently undergone human clinical trials. These include studies of antiox

Trauma to the spinal cord in males may result in both erectile dysfunction and infertility. Reflex erections may be preserved in upper motor neuron lesions and psychogenic erections in lower motor neuron or incomplete lesions. There are numerous reports of abnormalities of testicular biopsies that may relate to increased scrotal temperature and other factors. Techniques to restore erections include intracavernous injections of vasoactive substances, vacuum tumescence constriction therapy and penile prostheses. All have potential adverse affects. When reflex function in the sacral cord is prese

Simultaneous electromyographic (EMG) recordings from the bladder detrusor muscle and the inferior rectus abdominis muscle were made in six normal subjects, in four patients with lower motor neuron bladder disease and in two patients with an upper motor neuron type of bladder lesion. Results of the study demonstrate that the bladder electrodes do not record remote muscle activity from the abdominal muscles so that any increased detrusor electrical activity with abdominal contraction must have some other explanation such as a possible abdominal-detrusor reflex or the production of increased intr

Expression of calbindin D28K (CB) and parvalbumin (PV) distinguishes matrix and core type thalamic neurons, respectively, in primates. Whether the nice correspondence of intracellular calcium binding proteins and matrix/core neuron types translates to mouse remains unknown. Sixty reconstructed 3D mediodorsal thalamic nuclear neurons (MDCalb1 neurons), defined by calb1, the gene encoding CB in mouse MD, were mapped onto Allen Mouse Brain Common Coordinate Framework (CCFv3). Most MDCalb1 neurons (54/60) were isocortex-projecting ones, possessing collateral connections with caudate putamen (CP).

Rett syndrome (RTT) is a progressive neurological disorder primarily caused by mutations in the X-linked gene methyl-CpG-binding protein 2 (MECP2). The heterozygous female brain consists of mosaic of neurons containing both wild-type MeCP2 (MeCP2+) and mutant MeCP2 (MeCP2-). Three-dimensional morphological analysis was performed on individually genotyped layer V pyramidal neurons in the primary motor cortex of heterozygous (Mecp2(+/-) ) and wild-type (Mecp2(+/+) ) female mice ( > 6 mo.) from the Mecp2(tm1.1Jae) line. Comparing basal dendrite morphology, soma and nuclear size of MeCP2+ to MeCP2

Activation of esophageal mechanosensors excites neurons in and near the central nucleus of the solitary tract (NSTc). In turn, NSTc neurons coordinate the relaxation of the stomach [i.e., the receptive relaxation reflex (RRR)] by modulating the output of vagal efferent neurons of the dorsal motor nucleus of the vagus (DMN). The NSTc area contains neurons with diverse neurochemical phenotypes, including a large population of catecholaminergic and nitrergic neurons. The aim of the present study was to determine whether either one of these prominent neuronal phenotypes was involved in the RRR. Im

Canonical parasympathetic innervation of tissues/organs is accomplished by two in-series neurons, often called pre- and postganglionic neurons. Such two-neuron in-series organization has recently been challenged by claiming direct vagal preganglionic innervation of Brunner's glands (BGs) in the duodenal bulb, as well as the spleen, and the liver. To resolve this controversial issue, we labeled vagal preganglionic neurons in the dorsal motor nucleus of the vagus (DMV) by injecting a Cre-dependent AAV construct to express tdTomato in ChAT-IRES-Cre transgenic mice and analyzed the distribution of

The ventromedial prefrontal cortex regulates both emotional and physiological processes. The infralimbic cortex (IL), a prefrontal subregion in rodents, integrates behavioral, neuroendocrine, and autonomic responses to stress. However, the organization of cortical inputs to brainstem nuclei that regulate homeostatic responses are not well defined. We hypothesized that IL projections differentially target pre-ganglionic parasympathetic neurons and adrenergic/noradrenergic nuclei. To quantify IL projections to autonomic brainstem nuclei in male rats, we utilized viral-mediated gene transfer to e

Cognitive impairment is a common non-motor complication of Parkinson's disease (PD). Glucocerebrosidase gene (GBA1) variants are found in 10-15% of PD cases and are numerically the most important risk factor for PD and dementia with Lewy bodies. Accumulation of α-synuclein and tau pathology is thought to underlie cognitive impairment in PD and likely involves cholinergic as well as dopaminergic neurons. Neural crest stem cells were isolated from both PD patients with the common heterozygous N370S GBA1 mutation and normal subjects without GBA1 mutations. The stem cells were used to generate a c

Ex vivo, wild-type cortical neurons formed synapses, ganglia and aggregates at 96 h, whereas SPG21-/- neurons exhibited attenuated growth with markedly less axonal branches. Additionally, epidermal growth factor markedly promoted the growth and mat …

NURR1 is an essential transcription factor for the differentiation, maturation, and maintenance of midbrain dopaminergic neurons (DA neurons) as it has been demonstrated using knock-out mice. DA neurons of the substantia nigra pars compacta degenerate in Parkinson's disease (PD) and mutations in the Nurr1 gene have been associated with this human disease. Thus, the study of NURR1 actions in vivo is fundamental to understand the mechanisms of neuron generation and degeneration in the dopaminergic system. Here, we present and discuss findings indicating that NURR1 is a valuable molecular tool fo

BACKGROUND: The potential neuroprotective effects of dexmedetomidine against ketamine-induced neurotoxicity remain inconclusive. This study aims to investigate the influence of dexmedetomidine on ketamine-induced neuronal apoptosis and neurodevelopmental toxicity.
METHODS: In vitro experiments employed concentrations of 0.1 uM for dexmedetomidine and 50 uM for ketamine individually as well as their combination. Changes in apoptotic proteins and dendritic development in neurons were assessed after a 6-h exposure to the drugs with evaluations conducted 24 hs' post-treatment. In vivo experiments

Deficiencies in the complex I (CI; NADH-ubiquinone oxidoreductase) of the respiratory chain are frequent causes of mitochondrial diseases and have been associated with other neurodegenerative disorders, such as Parkinson's disease. The NADH-ubiquinone oxidoreductase 1 alpha subcomplex subunit 5 (NDUFA5) is a nuclear-encoded structural subunit of CI, located in the peripheral arm. We inactivated Ndufa5 in mice by the gene-trap methodology and found that this protein is required for embryonic survival. Therefore, we have created a conditional Ndufa5 knockout (KO) allele by introducing a rescuing

Motor activity of guinea-pig ileum in vitro was monitored by recording isometric tension. Superfusion of the serosal surface with vinblastine (22 microM) increased the motility of ileum but failed to do so in the presence of local anaesthetics and also in preparations depolarised with K+-Tyrode. Atropine (6 microM) or pentolinium (186 microM) reduced the motor response of vinblastine but higher concentration of atropine (60 microM) or pentolinium (930 microM) did not further attenuate the response. Strychnine (58 microM) reversibly blocked the atropine-resistant motor response elicited by vinb

Postural and locomotor dysfunction represent axial symptoms of Parkinson's disease (PD), which remain poorly treated by medication and deep brain stimulation. Whilst non-invasive neuromodulation of the vestibular system via the vestibular nucleus complex (VNC) offers a novel therapeutic avenue, the underlying circuits are still poorly characterized. Here we show that the mouse VNC feeds extensive Vglut2-defined projections into striato-thalamo-subthalamic and caudal medulla motor hubs and receives substantial input from the sensorimotor cortex. Optogenetic activation of excitatory VNC neurons
