Visual gamma stimulation induces 40 Hz neural oscillations in the human hippocampus and alters phase synchrony and lag.

Nonpharmaceutical approaches based on gamma entrainment using sensory stimuli (GENUS) have shown promise in reducing Alzheimer's disease pathology in mouse models. While human studies remain limited, GENUS has been shown to alleviate aspects of neurodegeneration in patients with Alzheimer's disease. In this study, we analyze intracranial EEG data from 490 contacts across eleven patients with refractory epilepsy in response to three visual stimulation conditions. We find that 40 Hz visual stimulation successfully entrains neural activity beyond early visual areas, including the hippocampus and

Targeting brain connectivity in Alzheimer's disease with repurposed drugs.

Neuroimaging studies have highlighted both hyperconnectivity and hypoconnectivity across the Alzheimer's disease (AD) continuum, alongside task-induced activity changes. These alterations may reflect compensatory mechanisms or network breakdowns. While connectivity-based measures are not yet established as clinical biomarkers, they hold promises for evaluating therapeutic efficacy and informing the design of targeted interventions. Based on these insights, this review explores the potential of off-label FDA-approved drug repositioning as a cost-effective strategy to identify therapeutic approa

Temporal Dynamics of Beta Power Related to the Stun Effect in Parkinson's Disease Patients after Deep Brain Stimulation Surgery.

BACKGROUND: Deep brain stimulation (DBS) of the subthalamic nucleus (STN) is an effective treatment for patients with advanced Parkinson's disease (PD). Early programming of DBS may be complicated by the postoperative "stun effect," which also may impact local field potentials (LFPs). METHODS: Percept PC neurostimulators (Medtronic) were used to continuously record LFPs from the STN in 32 PD patients at home during the first 2 months after DBS surgery. Average beta peak power ("beta power") per day was analyzed using change-point analysis. RESULTS: Median beta power across patients showed an

EEG as a neural measure of hypoxia-related impairment.

Ambient oxygen decreases with increasing altitude, which poses a primary threat to aviators known as hypoxic hypoxia. Decades of research have shown that hypoxia impairs cognition, but the neurophysiological bases for these effects remain poorly understood. Recent advances in neuroscience have permitted non-invasive observation of neural activity under controlled hypoxia exposures and have begun to uncover how the brain responds to hypoxia. Electroencephalography (EEG) in particular has been used to explore how electrical activity produced by networks of cortical neurons changes under hypoxia.

Dual GSK3β/SIRT1 modulators for Alzheimer's: mechanisms, drug discovery and future perspectives.

Alzheimer's disease (AD) remains without effective disease-modifying therapies, in part due to the limited efficacy of single-target approaches. Dual modulation of glycogen synthase kinase-3β (GSK3β), a key driver of tau hyperphosphorylation and amyloid-β (Aβ) production, and sirtuin-1 (SIRT1), a neuroprotective NAD+-dependent deacetylase, has emerged as a promising therapeutic strategy. This review explores the mechanistic rationale for concurrently inhibiting GSK3β and activating SIRT1 to disrupt AD's pathological cascade while enhancing endogenous neuroprotective pathways. Natural compounds

Delayed Onset of Spinal Anesthesia in a Chronic Opioid User Undergoing Cesarean Section.

Delayed onset of spinal anesthesia can pose challenges during cesarean section (CS) in patients with a history of chronic opioid use. We present a case of delayed onset of spinal anesthesia in a patient with chronic opioid use undergoing CS, highlighting the clinical implications and management strategies. Delayed onset of spinal anesthesia in chronic opioid users can result from neurophysiological adaptations and pharmacokinetic interactions in the central nervous system. Previous studies have suggested chronic opioid users display a delayed onset and shorter duration of action of neuraxial b

Screening of Aβ and phosphorylated tau status in the cerebrospinal fluid through machine learning analysis of portable electroencephalography data.

Diagnosing Alzheimer's disease (AD) through pathological markers is typically costly and invasive. This study aims to find a noninvasive, cost-effective method using portable electroencephalography (EEG) to detect changes in AD-related biomarkers in cerebrospinal fluid (CSF). A total of 102 patients, both with and without AD-related biomarker changes (amyloid beta and phosphorylated tau), were recorded using a 2-minute resting-state portable EEG. A machine-learning algorithm then analyzed the EEG data to identify these biomarker changes. The results showed that the machine learning model could

Motor neuron diseases are not exclusively motor; the SSR paradigm.

Motor Neuron Diseases (MNDs), familial and sporadic, are progressive neurodegenerative disorders that, for an extended period in the past, were considered purely motor disorders. During the course of the disease, however, some patients exhibit concomitant non-motor signs; thus, MNDs are currently perceived as multisystem disorders. Assessment of non-motor symptoms is usually performed clinically, although laboratory tests can also be routinely used to objectively evaluate these symptoms. Sympathetic Skin Response (SSR) is an example of a neurophysiological test that has been used in cases of A

Microsurgical reconstruction of lingual nerve in 130 patients without nerve grafting.

Lingual Nerve Injury (LNI) typically results in loss of sensation and pain, which can significantly impact social life and cause personal discomfort. Microsurgical reconstruction of the lingual nerve (LN) has been shown to alleviate these symptoms, leading to satisfactory functional recovery. Between January 2009 and December 2023, 149 patients underwent direct microneurorrhaphy of the LN, with 130 patients meeting our inclusion criteria. All patients were evaluated preoperatively and one year postoperatively using clinical and neurophysiological assessments (Trigeminal Sensitivity Test) to ev

Gap Junctions May Have A Computational Function In The Cerebellum: A Hypothesis.

In the cerebellum, granule cells make parallel fibre contact on (and excite) Golgi cells and Golgi cells inhibit granule cells, forming an open feedback loop. Parallel fibres excite Golgi cells synaptically, each making a single contact. Golgi cells inhibit granule cells in a structure called a glomerulus almost exclusively by GABA spillover acting through extrasynaptic GABAA receptors. Golgi cells are connected dendritically by gap junctions. It has long been suspected that feedback contributes to homeostatic regulation of parallel fibre signals activity, causing the fraction of the populatio

Spatiotemporal consistency analysis of cerebral small vessel disease: an rs-fMRI study.

INTRODUCTION: Cerebral small vessel disease (SVD) affects older adults, but traditional approaches have limited the understanding of the neural mechanisms of SVD. This study aimed to explore the effects of SVD on brain regions and its association with cognitive decline using the four-dimensional (spatiotemporal) consistency of local neural activity (FOCA) method. METHODS: Magnetic resonance imaging data from 42 patients with SVD and 38 healthy controls (HCs) were analyzed using the FOCA values. A two-sample t test was performed to compare the differences in FOCA values in the brain between th

Neural signatures of natural behaviour in socializing macaques.

Our understanding of the neurobiology of primate behaviour largely derives from artificial tasks in highly controlled laboratory settings, overlooking most natural behaviours that primate brains evolved to produce1-3. How primates navigate the multidimensional social relationships that structure daily life4 and shape survival and reproductive success5 remains largely unclear at the single-neuron level. Here we combine ethological analysis, computer vision and wireless recording technologies to identify neural signatures of natural behaviour in unrestrained, socially interacting pairs of rhesus

A multi-omics Mendelian randomization study identifies new therapeutic targets for alcohol use disorder and problem drinking.

Integrating proteomic and transcriptomic data with genetic architectures of problematic alcohol use and alcohol consumption behaviours can advance our understanding and help identify therapeutic targets. We conducted systematic screens using genome-wise association study data from ~3,500 cortical proteins (N = 722) and ~6,100 genes in 8 canonical brain cell types (N = 192) with 4 alcohol-related outcomes (N ≤ 537,349), identifying 217 cortical proteins and 255 cell-type genes associated with these behaviours, with 36 proteins and 37 cell-type genes being new. Although there was limited overlap

An Open-Source 3D-Printed Recording Stage with Customizable Chambers for Ex Vivo Experiments.

Much of what has been discovered concerning neurophysiological mechanisms can be credited to ex vivo biomedical experiments. Beyond these discoveries, ex vivo research techniques have enhanced the global understanding of human physiology and pathology in almost every biomedical specialty. Naturally, ex vivo experiments are among the most desired methods of research, particularly in the field of neuroscience. Ex vivo experiment platforms may be purchased commercially. However, their substantial cost and sometimes limited availability can render them inaccessible to many research labs. Moreover,

Experimental Evidence on Age-related Differential Outcomes Associated With Substance Abuse.

Growing evidence indicates that adolescent substance abuse is now an alarming concern that imposes a considerable socio-economic burden on societies. On the other hand, numerous studies have shown that due to specific neurophysiological features, the brain is more vulnerable to the adverse effects of psychoactive drugs at an early age. Unfortunately, these negative effects are not limited to the period of drug use, but can persistently affect the brain's responsiveness to future exposures to the same or other types of drug. For researchers to develop pharmacological strategies for managing sub

The translational inhibitor and amnestic agent emetine also suppresses ongoing hippocampal neural activity similarly to other blockers of protein synthesis.

The consolidation of memory is thought to ultimately depend on the synthesis of new proteins, since translational inhibitors such as anisomycin and cycloheximide adversely affect the permanence of long-term memory. However, when applied directly in brain, these agents also profoundly suppress neural activity to an extent that is directly correlated to the degree of protein synthesis inhibition caused. Given that neural activity itself is likely to help mediate consolidation, this finding is a serious criticism of the strict de novo protein hypothesis of memory. Here, we test the neurophysiolog

Rapid-Onset Obsessive-Compulsive Disorder With Hallucinations in a Post-seizure Four-Year-Old Male.

Rapid-onset obsessive-compulsive disorder (OCD) has been classically described in the context of infectious and autoimmune stressors, most famously PANDAS (pediatric autoimmune neuropsychiatric disorders associated with streptococcal infections) and then PANS (pediatric autoimmune neuropsychiatric syndrome). PANS itself, however, specifically excludes neurological and medical disorders, including seizures, from the diagnostic criteria. Changes in affect, such as depression/anxiety and new-onset psychosis, have been previously described in the post-seizure period but often self-resolve. To the

Mechanisms and multimodal treatment of neuropathic pain associated with degenerative cervical spondylosis: Current status and progress.

Degenerative cervical spondylosis is a common spinal disorder in the elderly. During its progression, structural changes such as intervertebral disc degeneration, vertebral osteophyte formation, and facet joint degeneration directly impact neural structures, resulting in neuropathic pain. Moreover, the release of inflammatory mediators can further exacerbate pain, and increased neuronal excitability is considered one of the key pathological mechanisms of neuropathic pain. Current diagnostic and assessment methods for neuropathic pain associated with degenerative cervical spondylosis include cl

Immediate neural impact and incomplete compensation after semantic hub disconnection.

The human brain extracts meaning using an extensive neural system for semantic knowledge. Whether broadly distributed systems depend on or can compensate after losing a highly interconnected hub is controversial. We report intracranial recordings from two patients during a speech prediction task, obtained minutes before and after neurosurgical treatment requiring disconnection of the left anterior temporal lobe (ATL), a candidate semantic knowledge hub. Informed by modern diaschisis and predictive coding frameworks, we tested hypotheses ranging from solely neural network disruption to complete
