OCTA and microperimetry in Behçet's retinal vasculitis: subgroup analysis, structure-function correlation, and 12-month follow-up evaluation.

BACKGROUND: In Behçet's syndrome (BS)-associated retinal vasculitis, optical coherence tomography (OCT) angiography (OCTA) and microperimetry allow noninvasive assessment of retinal microvascular integrity and macular function, but integrated structure-function analyses across all retinal vascular plexuses during inactive disease remain limited. This study evaluated age- and sex-related differences, structure-function associations, and 12-month changes in inactive retinal vasculitis. METHODS: This observational study included 14 patients (23 eyes; mean age 40.6 ± 11.7 years) with inactive BS-

The EH-binding protein EHBP1 operates in a ciliary functional module affected by INPP5E dysfunction.

Inositol polyphosphate-5-phosphatase E (INPP5E) encodes the ciliary protein INPP5E, which plays an important role in regulating the phospholipid membrane makeup of the primary cilium. Here, we utilize proximity labeled proteomics of INPP5E to broaden the number of known functional modules that work in close proximity to the protein. In doing so, we identified the EH-binding protein EHBP1 as a ciliary protein that localizes to the basal body and ciliary compartment of the primary cilium in human-derived fibroblasts and RPE cells. Additionally, we show that EHBP1 localizes to the rudimentary out

A deep learning system for detecting systemic lupus erythematosus from retinal images.

Systemic lupus erythematosus (SLE) is a serious autoimmune disorder predominantly affecting women. However, screening for SLE and related complications poses significant challenges globally, due to complex diagnostic criteria and public unawareness. Since SLE-related retinal involvement could provide insights into disease activity and severity, we develop a deep learning system (DeepSLE) to detect SLE and its retinal and kidney complications from retinal images. In multi-ethnic validation datasets comprising 247,718 images from China and UK, DeepSLE achieves areas under the receiver operating

Retinal Autophagy for Sustaining Retinal Integrity as a Proof of Concept for Age-Related Macular Degeneration.

Current evidence indicates that most types of autophagy represent a pivot in promoting retinal integrity. In healthy conditions, autophagy acts on multiple pathways, which are fundamental for the biochemistry and the fine structure of the retina. Autophagy is essential in granting visual processes. On the other hand, autophagy dysfunction characterizes several retinal disorders. This is mostly evident in age-related macular degeneration (AMD), which represents the most common degenerative disease leading to blindness. The involvement of autophagy in AMD is documented in vitro and in vivo exper

Retinotopic degeneration of the retina and optic tracts in autosomal dominant Alzheimer's disease.

INTRODUCTION: We investigated the correlation between retinal thickness and optic tract integrity in subjects with autosomal dominant Alzheimer's disease (ADAD) causing mutations. METHODS: Retinal thicknesses and diffusion tensor images (DTI) were obtained using optical coherence tomography and magnetic resonance imaging, respectively. The association between retinal thickness and DTI measures was adjusted for age, sex, retinotopy, and correlation between eyes. RESULTS: Optic tract mean diffusivity and axial diffusivity were negatively correlated with retinotopically defined ganglion cell in

Acute retinal necrosis: pathophysiological aspects, diagnosis, and treatment.

Acute retinal necrosis (ARN) is a devastating viral infection commonly associated with varicella zoster virus (VZV) and herpes simplex virus types 1 and 2 (HSV-1, HSV-2). Typically, ARN affects individuals without immune status disorders between the ages of 50-70. In two-thirds of the cases, one-eye involvement is observed and the inflammation can be presented as panuveitis. The most characteristic clinical manifestations are vitreitis, occlusion of the retinal arterioles, and peripheral necrotizing retinitis. Retinitis presents with the appearance of deep, multifocal, yellowish-white foci, ty

Gene regulatory networks controlling temporal patterning, neurogenesis, and cell-fate specification in mammalian retina.

Gene regulatory networks (GRNs), consisting of transcription factors and their target sites, control neurogenesis and cell-fate specification in the developing central nervous system. In this study, we use integrated single-cell RNA and single-cell ATAC sequencing (scATAC-seq) analysis in developing mouse and human retina to identify multiple interconnected, evolutionarily conserved GRNs composed of cell-type-specific transcription factors that both activate genes within their own network and inhibit genes in other networks. These GRNs control temporal patterning in primary progenitors, regula

[Prevalence, morphology and therapy of toxoplasmosis chorioretinitis in AIDS].

Toxoplasmosis-retinochorioiditis is the second most frequent opportunistic infection of the eye among our series of AIDS patients. Between 1985 and 1990 we diagnosed 7 cases in 261 AIDS patients (Walter Reed classification 6); prevalence = 2.7%). The incidence has been increasing over the years. In four cases, toxoplasmosis was restricted to the eye, in three cases, ocular disease occurred combined with toxoplasmosis of the central nervous system. Since serological findings are not very reliable in AIDS-patients, the most important element in the differential diagnosis against retinitis of dif

Dominantly inherited retinitis pigmentosa. Ultrastructure and biochemical analysis.

A 66-year-old white man had dominant retinitis pigmentosa. He developed progressive restriction of his visual field, night blindness, pallor of the optic discs, pigmentary retinopathy and posterior subcapsular cataracts. Postmortem examination of the eyes included electron microscopy and biochemical analysis of cyclic nucleotides and interphotoreceptor retinoid-binding protein (IRBP). Except for the fovea and periphery, the retina showed extensive gliosis and neuronal loss with loss of photoreceptor cells. The choriocapillaris was variably occluded in the regions of absent retinal pigment epit

In Vitro retinal ganglion cell differentiation and enrichment under the scope: do subtypes matter?

Retinal ganglion cells (RGCs) play a pivotal part transmitting visual data to the brain. Yet, damaged RGCs are unable to maintain and regrow axons and connectivity, as in the common blinding disease glaucoma. Thus, the idea of rescuing and replacing damaged RGCs holds immense therapeutic potential. In recent years pluripotent stem cells cultured in both 2D and 3D (retinal organoid) environments have generated RGCs from healthy- and patient-derived cells. These models can be used to study normal retinal physiology and compare it to the diseased retina. Although the effects of glaucomatous injur

Human RGR Gene and Associated Features of Age-Related Macular Degeneration in Models of Retina-Choriocapillaris Atrophy.

Age-related macular degeneration (AMD) is a progressive eye disease and the most common cause of blindness among the elderly. AMD is characterized by early atrophy of the choriocapillaris and retinal pigment epithelium (RPE). Although AMD is a multifactorial disease with many environmental and genetic risk factors, a hallmark of the disease is the origination of extracellular deposits, or drusen, between the RPE and Bruch membrane. Human retinal G-protein-coupled receptor (RGR) gene generates an exon-skipping splice variant of RGR-opsin (RGR-d; NP_001012740) that is a persistent component of s
