Anesthetic Considerations for Maternal-Fetal Interventions.

Advances in diagnosing and managing fetal anomalies have enabled the rapid evolution of maternal-fetal surgery (MFS). These surgeries are broadly categorized as minimally invasive, open, or ex utero intrapartum treatment (EXIT) procedures. A multidisciplinary team is required for these techniques with the anesthesia provider playing a pivotal role. A unique aspect of MFS is the need to manage both mother and fetus. Successful delivery of anesthesia requires knowledge of the physiologic changes of pregnancy, fetal physiology, maintenance of uteroplacental blood flow, and mitigating risk of gene

Quantifying assembly processes of dissolved organic matter pools in eutrophication using high-resolution mass spectrometry and ecological models.

Dissolved organic matter (DOM) represents a large, dynamic pool of carbon, playing a crucial role in eutrophic aquatic ecosystems through its continuous transport and transformation. However, the assembly mechanisms of DOM under different eutrophic conditions remain elusive, hindering the understanding of carbon dynamics and the prediction of carbon fate. Here we collected 72 lake water samples during two sampling events in Chaohu Lake, the fifth largest freshwater lake in China, and performed high-resolution mass spectrometry (HRMS) and ecological null modeling to quantify the assembly proces

The threat of microplastics to human kidney health: Mechanisms of nephrotoxicity and future research directions.

Following the inadequacy of global plastic pollution control measures, microplastic (MP) pollution is posing new challenges to human health. In recent years, MPs have been detected in various human tissues, including their first identification in human kidneys in 2023. MPs can reach the kidneys through inhalation, oral ingestion, and intravascular injection, and they can be excreted via urine. Based on the latest research, this article reviews the nephrotoxicity of MPs and proposes a filtration-reabsorption-translocation hypothesis regarding the potential renal excretion mechanism of MPs. Shor

In-silico identification of novel Cis-aconitate decarboxylase inhibitors as potential anti-inflammatory agents using molecular docking and dynamics.

Cis-aconitate decarboxylase (CAD), also known as ACOD1 or IRG1, catalyzes the conversion of cis-aconitate to itaconate, playing a pivotal role in innate immunity and inflammatory diseases. Although CAD is a key enzyme in the pathophysiology of inflammatory diseases, no specific inhibitors for CAD currently exist. In this study, we screened 86,326 compounds similar to CAD ligand from the Zinc database to identify potential CAD inhibitors using molecular docking with Molegro Virtual Docker and AutoDock-Vina, followed by molecular dynamics (MD) simulations. The top candidates were further assesse

Aprocitentan in hypertension management: clinical efficacy, safety, and future prospects.

Hypertension (HTN) is a prevalent medical condition characterized by systolic blood pressure ≥ 130 mm Hg and diastolic blood pressure ≥ 80 mm Hg. In 2010, the global prevalence of HTN was 31.1%, with higher rates in men and low- and middle-income countries (LMICs). The etiology of primary HTN involves neurohumoral, renal, metabolic, genetic, and environmental factors, with the kidneys playing a significant role in long-term blood pressure regulation. Endothelin-1 (ET-1), a potent vasoconstrictor, contributes to HTN by affecting salt-water balance and promoting vascular remodeling. HTN often pr

Necessity for further exploration of both endogenous and exogenous DNases.

Deoxyribonucleases (DNases) are essential enzymes involved in DNA metabolism, playing critical roles in replication, repair, recombination, and the degradation of apoptotic DNA. They are primarily classified into two families: DNase I and DNase II, each exhibiting distinct catalytic properties and tissue-specific functions. Dysregulated DNase activity is implicated in various diseases. This review synthesizes current knowledge on the biochemical properties, gene structures, tissue distributions, and clinical significance of DNases. It explores their roles in disease mechanisms, the impact of g

Co-targeting of VEGFR2 and PD-L1 promotes survival and vasculature normalization in pleural mesothelioma.

Pleural mesothelioma (PM) is an aggressive cancer caused by asbestos exposure, with limited treatment options and poor prognosis, highlighting the need for more effective therapies. Combining immune checkpoint blockade with anti-angiogenic therapy has shown potential in other cancers. Our study investigated the combined inhibition of PD-L1 and VEGFR2 in a mouse model of PM. Using C57BL/6 mice with subcutaneous AE17 mesothelioma tumors, we assessed the effects of anti-PD-L1 therapy with induction, concomitant, or consolidation anti-VEGFR2 treatment. Mice received intraperitoneal doses every thr

The Histidine Kinase VraS Interacts with Vancomycin and Penicillins Through its Membrane-Anchored N-terminal Domain.

Multidrug-resistant Staphylococcus aureus (S. aureus) is a major global health threat, with the VraTSR three-component system playing a key role in conferring resistance to cell-wall active antibiotics, through regulation of the cell wall stress stimulon. The molecular signals sensed by VraTSR remain unknown. We investigated interactions of the membrane histidine kinase VraS with β-lactams and glycopeptides. Photo-crosslinking assays with a vancomycin-derived and an ampicillin-derived photoprobe revealed direct interaction of these two classes of antibiotics with full-length VraS. Saturation t

Irreducible Isolated Radial Head Dislocation in a Child Due to Annular Ligament Interposition: A Case Report.

The annular ligament of the elbow is essential for its stability, playing a key role in both the proximal radioulnar and humeroradial joints, as well as supporting surrounding muscles and ligaments. Radial head dislocation is rare in children, and when isolated they can be challenging to reduce and may require surgical intervention. An 11-year-old boy presented with an anteromedial dislocation of the radial head after a fall. Initial, closed reduction was attempted and failed, requiring surgical intervention, where we found a rupture of the annular ligament and interposition, which was repaire

TNFR1 Suppression by XPro1595 Reduces Peripheral Neuropathies Associated with Perineural Invasion in Female Mice.

Perineural invasion (PNI), defined by cancer spreading or invading into the nerve, links to severe pain, recurrence, and poor prognosis. PNI contributes to nerve damage, Schwann cell activation, and sensory neuron dysfunction. Soluble tumor necrosis factor α (solTNFα) binds to TNFR1 to drive inflammation and nerve injury, playing a key role in cancer progression and pain. This study, using a mouse sciatic nerve PNI model, explored whether blocking solTNFα-TNFR1 signaling via TNFR1 knockout or pharmacological inhibition by XPro1595 could reduce PNI-associated pain. Data showed that XPro1595, bu

High-density Multielectrode Array Recordings of Retinal Waves Using An Electrophysiology Platform.

Spontaneous retinal waves are a hallmark of retinal network activity during development, playing a crucial role in the formation of the visual system by influencing the refinement of axons, permeability of vasculature, and overall maturation of neural circuits. These waves are commonly studied in ex vivo retinal preparations using multielectrode arrays (MEAs), which enable electrophysiological recordings of large populations of retinal ganglion cell (RGC) activity. MEA-based electrophysiology has become a powerful tool due to its ease of use to rapidly collect high-throughput data, thus making

Two novel SH3TC2 mutations predispose to Charcot-Marie-Tooth disease type 4C by mistargeting away from TFRC.

Charcot-Marie-Tooth disease type 4C (CMT4C) is an autosomal recessive form of demyelinating neuropathy caused by the biallelic pathogenic mutations in the SH3TC2 gene and characterized by progressive scoliosis, muscular atrophy, distal weakness, and reduced nerve conduction velocity. Here, we report two novel SH3TC2 mutations (c.452dupT and c.731 + 1G > T) from a proband with typical clinical manifestations of CMT4C. Splicing assay reveals the SH3TC2 c.731 + 1G > T mutation leads to a 58-nucleotide (nt) deletion from the downstream of exon 6 causing a frameshift and resulting in an early termi
