Validation of body composition parameters extracted via deep learning-based segmentation from routine computed tomographies.

Sarcopenia and body composition metrics are strongly associated with patient outcomes. In this study, we developed and validated a flexible, open-access pipeline integrating available deep learning-based segmentation models with pre- and postprocessing steps to extract body composition measures from routine computed tomography (CT) scans. In 337 surgical oncology patients, total skeletal muscle tissue (SMtotal), psoas muscle tissue (SMpsoas), visceral adipose tissue (VAT), and subcutaneous adipose tissue (SAT) were quantified both manually and using the pipeline. Automated and manual measureme…

Senotherapeutic approach to age-related endocrine diseases.

Senescent cells progressively accumulate in the endocrine glands and their target tissue during the biological aging process. Senescence leads to hormonal imbalances contributing to various age-related endocrine diseases (AREDs). Cellular senescence, characterized by irreversible cell-cycle arrest, becomes more prevalent in advanced age, and the senescent cells release pro-inflammatory and pro-fibrotic factors, exacerbating endocrine dysregulation. Senescence-associated secretory phenotype (SASP) contributes to the pathogenesis of AREDs such as metabolic syndrome, sarcopenia, osteoporosis, and…

Phase angle: a novel application of bioelectrical impedance technology in osteoarthritis screening and diagnosis.

Bioelectrical impedance technology (EBI) offers a non-invasive, cost-effective method for body composition assessment, showing promise in diagnosing and managing musculoskeletal disorders, particularly osteoarthritis (OA). OA is a major global health concern, notably affecting the knee and hip joints. Conventional imaging techniques like X-rays and MRI have limitations in early detection, as they cannot capture microscopic cartilage changes. Phase angle (PhA), an essential EBI parameter, is widely applied in evaluating sarcopenia, tumors, and body fluids, and is increasingly valuable in OA res…

Intermuscular adipose tissue and muscle function in patients on maintenance hemodialysis.

Sarcopenia is common in advanced chronic kidney disease (CKD), contributing to poor outcomes. Intermuscular adipose tissue (IMAT) may worsen muscle function through metabolic and inflammatory pathways. We conducted a cross-sectional study comparing maintenance hemodialysis (MHD) patients and controls to assess IMAT and its associations. IMAT was measured via calf muscle MRI, and body composition (BMI, fat mass index), metabolic markers (hs-CRP, TNF-α, IL-6, insulin resistance), and circulating cell-free mitochondrial DNA (ccf-mtDNA) were assessed. Muscle function was evaluated with handgrip st…

Cigarette smoking inhibits myoblast regeneration by promoting proteasomal degradation of NPAT protein and hindering cell cycle progression.

Cigarette smoking (CS) causes skeletal muscle dysfunction, leading to sarcopenia and worse prognosis of patients with diverse systemic diseases. Here, we found that CS exposure prevented C2C12 myoblasts proliferation in a dose-dependent manner. Immunoblotting assays verified that CS exposure promoted the expression of cell cycle suppressor protein p21. Furthermore, CS exposure significantly inhibited replication-dependent (RD) histone transcription and caused S phase arrest in the cell cycle during C2C12 proliferation. Mechanistically, CS deregulated the expression levels of Nuclear Protein At…

Durable Muscle Extracellular Matrix Engineered with Adhesive Phenolic Moieties for Effective Skeletal Muscle Regeneration in Muscle Atrophy.

Muscle atrophy detrimentally impacts health and exacerbates physical disability, leading to increased mortality. In particular, sarcopenia, aging-related degenerative muscle loss, necessitates urgent remedies. Current approaches for treating muscle atrophy include exercise and nutrition, while drug exploration remains in its early stages. Cell therapy, focusing on satellite cells, faces significant challenge due to poor engraftment, safety issue, and high cost. Cell-free approach using extracellular matrix (ECM) shows a regenerative potential, but a lack of mechanical and adhesive properties h…

Predicting falls with ultrasound, physical parameters or fall-risk questions among older adults: A prospective cohort study.

BACKGROUND: Falls are a significant issue among older adults, leading to morbidity and mortality. Screening for fall risk in the ED is crucial but challenging due to time limitations and patient conditions. Sarcopenia, characterized by muscle loss, is associated with increased fall risk, and ultrasound has been proposed as a non-invasive tool to measure muscle mass in this context. METHODS: This prospective cohort study enrolled 174 older adults from an urban teaching hospital's EDOU, assessing muscle mass via POCUS, grip strength, Timed Up and Go (TUG) test, and fall risk using the STEADI to…

The influence of resistance training on adipokines in post-menopausal women: A brief review.

The onset of menopause elicits changes in body composition that negatively influence adipokine levels. Consequently, various health risk factors (e.g., cardiovascular disease, osteoporosis, physical inactivity, obesity, arterial hypertension, hypercholesterolemia, sarcopenia) are influenced by adipokines due to changes in body composition after menopause. Thus, improvements in body composition are considered the primary influencer of adipokines. Though several therapeutic interventions (e.g., medication, diet, meditation, exercise) are employed to target changes in body composition, resistance…

Tumor cell anabolism and host tissue catabolism-energetic inefficiency during cancer cachexia.

Cancer-associated cachexia (CC) is a pathological condition characterized by sarcopenia, adipose tissue depletion, and progressive weight loss. CC is driven by multiple factors such as anorexia, excessive catabolism, elevated energy expenditure by growing tumor mass, and inflammatory mediators released by cancer cells and surrounding tissues. In addition, endocrine system, systemic metabolism, and central nervous system (CNS) perturbations in combination with cachexia mediators elicit exponential elevation in catabolism and reduced anabolism in skeletal muscle, adipose tissue, and cardiac musc…

Speculation on the pathophysiology of musculoskeletal injury with COVID-19 infection.

Coronavirus disease 2019 (COVID-19) primarily affects the respiratory tract, but also many other organs and tissues, leading to different pathological pictures, such as those of the musculoskeletal tissues. The present study should be considered as a speculation on the relationship between COVID-19 infection and some frequent musculoskeletal pathologies, in particular sarcopenia, bone loss/osteoporosis (OP) and fracture risk and osteoarthritis (OA), to hypothesize how the virus acts on these pathologies and consequently on the tissue regeneration/healing potential. The study focuses in particu…

Camphene Attenuates Skeletal Muscle Atrophy by Regulating Oxidative Stress and Lipid Metabolism in Rats.

Sarcopenia- or cachexia-related muscle atrophy is due to imbalanced energy metabolism and oxidative stress-induced muscle dysfunction. Monoterpenes play biological and pharmacological reactive oxygen species (ROS) scavenging roles. Hence, we explored the effects of camphene, a bicyclic monoterpene, on skeletal muscle atrophy in vitro and in vivo. We treated L6 myoblast cells with camphene and then examined the ROS-related oxidative stress using Mito TrackerTM Red FM and anti-8-oxoguanine antibody staining. To investigate lipid metabolism, we performed real-time polymerase chain reactions, holo…
