Chronic fenfluramine administration: some cerebral effects.

Human cerebral function was monitored electrophysiologically during sleep over a period of months before, during, and after the intake of fenfluramine, 40-120 mg/day. Effects included dose-related reduction of paradoxical sleep, increase of intra-sleep restlessness, and changes in E.E.G. slow-wave sleep. It is hypothesized that weight loss may be associated with increase of the last. Grinding of teeth (bruxism) also was noted.Long-term studies make it possible to demonstrate changing central effects with time, including tolerance phenomena. Withdrawal abnormalities are related to the time take

Personalized assessment of high-definition slow-oscillatory transcranial direct current stimulation (so-tDCS) in older adults with sleep and subjective cognitive complaints: A study protocol for a randomized clinical trial.

BACKGROUND: Subjective cognitive decline (SCD) is increasingly recognized as an early and potentially modifiable stage in the trajectory of neurodegenerative diseases. In older adults with SCD, untreated sleep disturbances may accelerate the neurodegenerative process, potentially via mechanisms involving amyloid-beta accumulation, and contribute to more rapid cognitive deterioration. Currently, non-pharmacological treatment options that concurrently manage SCD and sleep disturbances remain scarce. High-definition slow-oscillatory transcranial direct current stimulation (so-tDCS) is an emerging

Longitudinal Sleep Patterns and Risk of Type 2 Diabetes Mellitus.

AIMS: Evidence relating sleep characteristics to type 2 diabetes mellitus (T2DM) has largely been based on single assessments. We examined whether transitions in a multidimensional healthy sleep score between two assessments were associated with incident T2DM and explored the relationship of the triglyceride-glucose (TyG) index to this association. MATERIALS AND METHODS: We conducted a landmark analysis among 60,249 UK Biobank participants with sleep assessed at visits 1 and 2. Visit 2 was defined as time zero for follow-up. Sleep pattern transitions were classified according to poor, interme

Association of Stress and Neighborhood Social Context With Actigraphy-Measured and Self-Reported Adolescent Sleep Outcomes.

PURPOSE: Chronic stress and unfavorable neighborhood environments may increase adolescents' risk for poor sleep. Few studies have examined whether neighborhood social environments moderate associations of adolescents' perceptions of stress with actigraphy-measured and self-reported sleep outcomes. METHODS: In a cross-sectional study of adolescents aged 15-18 (n = 163) years, perceived stress (10-item Perceived Stress Scale) and perceived neighborhood collective efficacy and safety were assessed via survey. Over 14 days, actigraphy measured nightly sleep duration and timing, while ecological m

Association Between Gastrointestinal Symptoms and Sleep Habits in Children with Metabolic Dysfunction-Associated Steatotic Liver Disease: A Cross-Sectional Study.

OBJECTIVE: Reduced sleep quality in children has been associated with obesity and fatty liver disease. However, there are no studies evaluating the impact of gastrointestinal symptoms on sleep quality in children with metabolic dysfunction-associated steatotic liver disease (MASLD). The aim of this cross-sectional study was to investigate the prevalence of gastrointestinal symptoms and sleep disturbances in children with MASLD and to examine the association of gastrointestinal symptoms with sleep disturbances. METHODS: Anthropometric and biochemical examinations were performed on children age

Stability of commonly used sleep questionnaires in people with type 2 diabetes mellitus treated in primary care.

INTRODUCTION: Sleep is increasingly recognized as a modifiable factor influencing metabolic and cardiovascular outcomes in type 2 diabetes mellitus (T2DM). Reliable sleep assessments are essential in primary care to assess sleep in T2DM. However, the stability of self-rated sleep measures in this group remains unclear. AIM: This study evaluated the temporal stability of the Epworth Sleepiness Scale (ESS), Pittsburgh Sleep Quality Index (PSQI), and STOP-BANG questionnaire in T2DM patients. MATERIAL AND METHODS: This prospective observational study included 53 individuals with T2DM referred fr

Screening of sleep assisting drug candidates with a Drosophila model.

Lately, Drosophila has been favored as a model in sleep and circadian rhythm research due to its conserved mechanism and easily manageable operation. These studies have revealed the sophisticated parameters in whole-day sleep profiles of Drosophila, drawing connections between Drosophila sleep and human sleep. In this study, we tested several sleep deprivation protocols (mechanical shakes and light interruptions) on Drosophila and delineated their influences on Drosophila sleep. We applied a daytime light-deprivation protocol (DD) mimicking jet-lag to screen drugs that alleviate sleep deprivat

Subjective and objective features of sleep disorders in patients with acute ischemic or haemorrhagic stroke: It is not only sleep apnoea which is important.

BACKGROUND: More than half of stroke patients present with a sleep-related breathing disorder including both central and obstructive forms of sleep apnoea. A cerebral infarction in different brain areas can disrupt sleep regulating pathways and cause insomnia, hypersomnia, circadian rhythm disturbances and other sleep disorders. Therefore, there is a need of objective data about various sleep disorders arising after ischemic or haemorrhagic stroke in order to implement practical recommendations how to suspect, diagnose and treat these conditions. Our medical hypothesis is that non-breathing sl

Sleep latency versus shuteye latency: Prevalence, predictors and relation to insomnia symptoms in a representative sample of adults.

Shuteye latency (SEL) refers to the time spent performing activities in bed before attempting sleep. This study investigates (a) the prevalence, duration and predictors of SEL, (b) its association with insomnia symptoms (sleep onset latency [SOL], sleep quality and fatigue), and (c) the activities engaged in during SEL. A representative sample of 584 adults (18-96 years old) participated in an online survey. Respondents reported their SEL on weekday nights (Sunday to Thursday) and weekend nights (Friday and Saturday), and activities during SEL. One in five adults tried to sleep immediately at

Professional shift-work drivers who adopt prophylactic naps can reduce the risk of car accidents during night work.

STUDY OBJECTIVES: Night work can be dangerous because both circadian sleep propensity (process C) and sleep pressure due to the prolonged wakefulness (process S) contribute to the reduction of vigilance levels. As naps are a countermeasure to sleepiness, this study evaluates the role they play in preventing sleep-related accidents in Italian shift-working police drivers. DESIGN/SETTING/PARTICIPANTS: The study concerns highway car accidents that occurred to Italian shift-working police drivers; it was performed in 2 steps: a retrospective analysis of the overall number of accidents that occurr

Cognitive behavioral therapy for insomnia combined with eszopiclone for the treatment of sleep disorder patients transferred out of the intensive care unit: A single-centred retrospective observational study.

Patients transferred out of the intensive care unit (ICU) are always impaired by sleep disorders. Cognitive behavioral therapy for insomnia (CBT-I) and eszopiclone are 2 commonly prescribed strategies for insomnia. In the current study, the effect of the combined application of the 2 methods on sleep disorders in ICU transferred out patients was assessed.Twenty-nine insomnia patients receiving combined treatment of CBT-I and eszopiclone and a corresponding number of patients treated with eszopiclone were collected. The incidence of discomfort experiences in ICU was recorded. Polysomnogram (PSG

Understanding mental and physical fatigue complaints in those with depression and insomnia.

Fatigue is a concern for both people with insomnia and with depression, yet it remains poorly understood. Participants (N = 62) included those meeting Diagnostic and Statistical Manual of Mental Disorders (4th ed., text revision) criteria for insomnia and major depressive disorder (MDD). Multiple regression examined sleep, mood, activity, and cognitive factors as predictors of mental and physical fatigue. Only the cognitive factors (i.e., unhelpful beliefs about sleep and symptom-focused rumination) were predictive of both physical and mental fatigue. Beliefs about not being able to function a

A comparison of sleep disturbances and sleep apnea in patients on hemodialysis and chronic peritoneal dialysis.

Studies have shown that sleep disorders are common among dialysis patients; however, few studies have compared the prevalence of different sleep disorders in patients on peritoneal dialysis (PD) and hemodialysis (HD). We used questionnaires to assess the prevalence of common sleep disorders in dialysis patients. We compared the prevalence of sleep apnea (SA) risk, restless legs syndrome (RLS), insomnia, and excessive daytime sleepiness (EDS), as well as sleep quality, in both groups. Of the 227 patients who were enrolled in the study, the total number of patients on HD was 188 (82%), while the
