HSD3B1 links ileal steroid metabolism to bile acid regulation in patients with prostate cancer.

Androgen deprivation therapy (ADT), a cornerstone of advanced prostate cancer treatment, effectively suppresses androgen signaling but frequently induces systemic metabolic dysregulation. Here, we delineate an unrecognized intestinal steroid/bile acid regulatory axis that mechanistically links androgen suppression to extratumoral metabolic aberrations. HSD3B1 is the most common inherited link to prostate cancer mortality and mediates its effects by regulating steroid metabolism. Integrated metabolomic profiling of patients undergoing ADT revealed a rapid genotype-associated reduction in circul

Association of formaldehyde with sex steroid hormones in US children and adolescents (NHANES 2013-2016).

Experimental studies suggest that formaldehyde may disrupt reproductive and endocrine function, but human evidence remains limited. We examined the association between formaldehyde exposure, measured as hemoglobin adducts, and serum sex steroid hormones among children (6-11 years) and adolescents (12-19 years) participating in NHANES 2013-2016. Multivariable regression models were used to evaluate associations between formaldehyde and total testosterone (TT), estradiol (E2), sex hormone-binding globulin (SHBG), free testosterone, free estradiol (FE2), and the TT/E2 ratio. False discovery rate

Associations of Allopregnanolone and Related Steroid Hormones with Prenatal Psychosocial Distress in the Healthy Start Cohort.

Psychosocial distress is associated with adverse perinatal outcomes, yet its biological correlates remain incompletely understood. We evaluated associations of prenatal psychosocial distress with allopregnanolone (ALLO) and related steroids, and assessed race/ethnicity as a potential effect modifier, using data from 237 participants from the Healthy Start Study. We selected participants based on high distress (Edinburgh Perinatal Depression Scale [EPDS] ≥13 or EPDS-3A ≥7; n = 57) or low distress (EPDS <4 and EPDS-3A <2; n = 180). We quantified ALLO, progesterone, pregnanolone, cortisol, and co…

Steroid Biosynthesis Pathway Counteracts Iron Overload-Induced Ferroptosis in Mouse Granulosa Cells.

Iron overload is a recognized risk factor for female reproductive dysfunction, yet the underlying cellular and molecular mechanisms remain incompletely understood. In this study, the effects of iron overload on ovarian granulosa cells were investigated, and a protective role of the steroid biosynthesis pathway against ferroptosis was identified. A mouse model of ovarian iron overload was established by daily gavage of ferric citrate (FC, 120 mg/kg for 40 days). Iron-overloaded female mice exhibited disrupted estrous cycles, reduced serum estradiol levels, impaired antral follicle development,

Aspirin or non-steroidal anti-inflammatory drug initiation and subsequent bladder cancer evaluation.

BACKGROUND: Whether aspirin or non-aspirin non-steroidal anti-inflammatory drugs (NSAIDs) initiation may unmask unknown bladder cancers (BCs) by hematuria remains controversial. OBJECTIVES: To evaluate unmasking, we compared cystoscopy rates, BC prevalence at cystoscopy, and invasive stage prevalence at diagnosis in aspirin or NSAID initiators with never-users. METHODS: Using the Danish population-based registries, we identified all new aspirin or NSAID users in 2005-2023 and a matched never-using comparison cohort. Second, we identified all first-time cystoscopy recipients in 2005-2022 with

Exploring the potential efficacy of Ziziphus spina-christi on restoring steroid sensitivity in severe asthma.

OBJECTIVE: To determine whether Ziziphus spina-christi adjuvant restores steroid hyporesponsiveness in severe asthma by strengthening the gut epithelial barrier and inhibiting the STING sensing pathway. METHODS: Chemical profiling of Z. spina-christi extract was performed using liquid chromatography mass spectrometry (LC/MS-MS). Steroid-hyporesponsive asthma was induced with house dust mite (HDM) and cyclic-di-GMP (c-di-GMP). Mice were treated with dexamethasone, Z. spina-christi, or combination therapy. The study assessed gut histopathology, epithelial tight junction proteins, lung inflammat

The impact of steroid-impregnated polypropylene mesh on foreign body reaction in an experimental rat study.

PURPOSE: Foreign body reaction is a tissue response in which excessive reactions may lead to complications such as chronic inflammation, mesh contraction, and impaired tissue integration. The present study aimed to evaluate the effects of steroid-impregnated polypropylene mesh on foreign body reaction and fibrosis in a rat abdominal wall model. METHODS: Forty adults female Wistar rats were randomly assigned into four groups (n = 10 each): polypropylene mesh in the acute period (group A), steroid-impregnated polypropylene mesh in the acute period (group AS), polypropylene mesh in the chronic p

Steroid-Sensitive Nephrotic Syndrome and Anti-Nephrin Antibodies: Pathogenic Insights and Therapeutic Opportunities.

Steroid-sensitive nephrotic syndrome (SSNS) encompasses a heterogeneous spectrum of podocytopathies, ranging from classical T-cell-mediated forms to those with an autoimmune component. Anti-nephrin autoantibodies have recently emerged as serological markers in a subset of immune-mediated podocytopathies, particularly in minimal change disease (MCD), primary FSGS, and immunoresponsive SSNS/SDNS. Although they correlate with disease activity and remission in selected cases, their sensitivity remains variable and assay standardization is still lacking. Rituximab shows the greatest efficacy in ste

Rapid Remission of Steroid-Refractory IgA Nephropathy With Targeted-Release Budesonide: A Case Report.

IgA nephropathy (IgAN) is the most common primary glomerulonephritis worldwide and a major cause of progressive chronic kidney disease. Systemic corticosteroids are commonly used in high-risk patients but are frequently limited by inadequate response and significant adverse effects. We present the case of a 46-year-old woman with biopsy-confirmed IgAN who developed persistent microscopic hematuria and nephritic-range proteinuria. After six months of treatment with high-dose systemic prednisone, renin-angiotensin-aldosterone system blockade, and a sodium-glucose cotransporter-2 inhibitor, there…

Treatment of Steroid-Induced Osteonecrosis of the Distal Femoral Condyle Using Structural Bone Grafting.

BACKGROUND: Steroid-induced osteonecrosis of the distal femoral condyle is an uncommon condition that predominantly affects young patients receiving high-dose corticosteroid therapy. Joint-preserving treatment options for precollapse lesions remain limited, particularly in cases with extensive subchondral involvement. The purpose of this study was to evaluate the clinical and radiological outcomes of structural bone grafting (SBG) using a bicortical iliac bone graft (autograft or allograft) for precollapse osteonecrosis of the distal femoral condyle. METHODS: This retrospective case series in

Effectiveness of epidural steroid injections in the treatment of lumbar radiculopathy: a systematic review and meta-analysis.

INTRODUCTION: Epidural steroid injection (ESI) techniques have demonstrated effectiveness in improving short- and intermediate-term pain and disability outcomes; however, their efficacy may vary depending on the specific approach used. This meta-analysis aims to compare disability and pain outcomes among different ESI techniques - transforaminal (TFESI), interlaminar (ILESI), caudal (CESI), and combined approaches - in patients with lumbar radiculopathy. METHODS: The authors searched PubMed/MEDLINE, Scopus, and Cochrane from inception to December 16, 2025, in accordance with PRISMA guidelines

Steroid-responsive meningitis-arteritis in a dog presented with marked cardiac dysfunction and a hypercoagulable state.

Canine steroid-responsive meningitis-arteritis (SRMA) is an immune-mediated inflammatory disorder targeting the leptomeninges and associated vasculature with systemic inflammation and occasionally with mild systolic dysfunction and cardiac arrhythmias. Hypercoagulability has not been previously documented in dogs with SRMA, despite increases in inflammatory markers. This case report describes a 6-month-old female Australian heeler dog with acute progressive pyrexia and cervical hyperesthesia. Notable findings included marked decreased cardiac contractility and hypercoagulability determined via

Rethinking Steroid-Induced Rosacea: Why Vascular Laser Therapy Deserves an Earlier Role.

Steroid-induced rosacea (SIR) is traditionally managed through withdrawal of the offending corticosteroid, trigger avoidance, barrier-supportive care, and topical or systemic anti-inflammatory therapy. Although these measures remain essential, persistent erythema, flushing, and telangiectasia may continue to dominate after the acute rebound has stabilized. This editorial argues for earlier vascular assessment and, in selected clinically stabilized patients with a vascular-predominant phenotype, consideration of vascular-directed treatment rather than automatic deferral until prolonged medical-

Steroid profiling in congenital adrenal hyperplasia: comparing immunoassays and LC-MS/MS accuracy.

OBJECTIVES: Congenital adrenal hyperplasia (CAH) is an autosomal recessive disorder caused by defects in enzymes responsible for cortisol synthesis in the adrenal cortex. Accurate steroid hormone measurement is essential for diagnosis and monitoring, but not all metabolites can be reliably quantified by ELISA. Immunoassays and liquid chromatography-tandem mass spectrometry (LC-MS/MS) are commonly used for steroid profiling. This study aimed to compare steroid hormone levels measured by immunoassay and LC-MS/MS and to evaluate their correlation with clinical control in CAH patients. METHODS: F

Expanded Steroid Profiling Identifies Novel Newborn Screening Markers for Congenital Adrenal Hyperplasia.

CONTEXT: Simultaneous measurement of multiple steroids in dried bloodspots facilitates accurate newborn screening (NBS) for classic congenital adrenal hyperplasia (CAH), although false-positive (FP) tests are encountered. Some babies with milder forms of CAH, who may benefit from early detection and treatment, can be missed by screening. OBJECTIVE: This work aims to evaluate a method for expanded steroid profiling in NBS bloodspots to identify novel sensitive and specific markers that will inform improvements for screening for CAH, and to identify the metabolic routes to excessive androgen sy…

Steroid-responsive Encephalopathy Associated with Autoimmune Thyroiditis: A Case Fulfilling Definite Graus Criteria.

Steroid-responsive encephalopathy associated with autoimmune thyroiditis (SREAT), also known as Hashimoto encephalopathy, is a rare immune-mediated neurological syndrome with an incidence of 2.1 per 100,000 and a 4:1 female predominance. Neurological manifestations are independent of thyroid status, occurring in 30-40% of euthyroid patients, and include cognitive impairment, confusion, seizures, and psychiatric symptoms. Diagnosis is challenging in the elderly, where symptoms overlap with dementia or delirium, with only 51 reported cases in patients aged ≥65 years. We describe a 70-year-old wo
