Urinary free-cortisol levels (micrograms per day) were measured by radioimmunoassay at 2-week intervals during the course of hospitalization in the following patient groups: posttraumatic stress disorder (PTSD); major depressive disorder; bipolar I, manic; paranoid schizophrenia; and undifferentiated schizophrenia. The mean cortisol level during hospitalization was significantly lower in PTSD (33.3 +/- 3.2) than in major depressive disorder (49.6 +/- 5.9), bipolar I, manic (62.7 +/- 6.7), and undifferentiated schizophrenia (50.1 +/- 8.9), but was similar to that in paranoid schizophrenia (37.5

Sixty-five alcoholic patients admitted for detoxification had blood pressure, withdrawal symptoms, plasma cortisol (PC) and plasma aldosterone (PA) levels, plasma renin activity (PRA), and serum dopamine beta-hydroxylase (DBH) levels measured on the first and fourth days after admission. On the morning after admission blood pressure was elevated (greater than 140/90) in 32 patients (49%) and was 160/95 mmHg or more in 21 (32%). PRA was initially elevated in 41 patients, PA levels in 14, and 13 patients had raised PC levels. By the fourth day, blood pressure and biochemical measures had fallen

BACKGROUND: Excessive salt intake is a major contributor to hypertension; however, the role of sympathetic nerve activity (SNA) remains controversial. Most existing evidence relies on indirect markers or single-time-point measurements. Therefore, we directly and continuously recorded SNA to test the hypothesis that SNA contributes to the initiation, progression, or both phases of salt-induced hypertension.
METHODS: We continuously recorded renal and lumbar SNA, arterial pressure, and heart rate for 4 weeks in conscious sham-operated and uninephrectomized rats. After a control period on a norm

Myocardial ischemia (MI) often leads to sympathetic overexcitation, though the underlying mechanisms are not fully understood. This work was to elucidate the role of P2Y6 receptors in the superior cervical ganglion (SCG) in mediating sympathetic hyperactivity after MI. In vivo, a rat model of MI was established by mild ligation of the left anterior descending coronary artery. These rats exhibited ST-segment elevation, elevated blood pressure, increased heart rate, enhanced sympathetic nerve discharge, raised serum norepinephrine levels, and structural damage to myocardial tissue. These changes

We have shown that single-pulse transcranial magnetic stimulation (TMS) of the dorsolateral prefrontal cortex (dlPFC) inhibits muscle sympathetic nerve activity. However, this was likely due to arousal caused by the TMS pulses themselves, rather than altering the underlying neuronal circuitry. In extension, we have aimed to explore the effects of single-pulse TMS on skin sympathetic nerve activity (SSNA), which is more sensitive to arousal. It was hypothesised that TMS-evoked arousal would increase SSNA but would not generate de novo bursts from the dlPFC. Microneurographic recordings were tak

Systemic inflammation engages autonomic reflexes that modulate immune responses, yet the peripheral sensors and central circuits responsible for recruiting sympathetic anti-inflammatory pathways remain incompletely defined. Here, we identify the carotid bodies as critical afferent sensors required for full engagement of a splanchnic sympathetic reflex during endotoxemia. Using in vivo splanchnic sympathetic nerve activity recordings, cytokine measurements, and brainstem neuronal mapping, we show that intraperitoneal injection of lipopolysaccharide (LPS) activates splanchnic sympathetic outflow

BACKGROUND: Congestive heart failure (CHF) is characterized by the activation of neurohumoral drive concomitant with avid fluid retention. Renal denervation alleviates this fluid retention. SGLT2 (sodium-glucose cotransporter 2) inhibitors have shown remarkable improvement in patients with cardiovascular diseases. We have recently demonstrated a relationship between enhanced renal sympathetic nerve activity and SGLT2 expression as well as function during CHF; however, the precise molecular mechanisms involved in the expression and translocation of SGLT2 and associated scaffolding proteins to t

This study tested the hypothesis that neural and vascular α2-adrenergic mechanisms contribute to sympathetic baroreflex regulation of human blood pressure. Muscle sympathetic nerve activity (MSNA; peroneal microneurography) was measured, and sympathetic action potentials (APs) were extracted from the filtered MSNA neurogram (continuous wavelet transform) in eight participants (5 females; 28 ± 7 yr) during a baseline (BSL) condition and a dexmedetomidine infusion (DEX; α2-adrenergic receptor agonist; 10-min loading dose at 0.225 µg/kg; maintenance dose: 0.1-0.5 µg/kg/h). Vascular conductance increases following a single bout of dynamic exercise, whereas sympathetic nerve traffic commonly appears unchanged. Discordance between vascular and sympathetic responses may reflect modulation in vasoconstrictor responsiveness. The primary study objective was to evaluate sympathetic neurovascular transduction following cycling exercise. The secondary objective was to explore mechanisms contributing to altered sympathetic neurovascular transduction by manipulating limb vascular conductance using local heating of the foot. We hypothesized that sympathetic neurovascular tra

AIMS: One important contributor to the pathogenesis and perpetuation of HFpEF is unfavorable pulse wave reflection, frequently caused by increased arterial stiffness, due to aging and longstanding hypertension. One possible path to address pathological wave reflections is reducing sympathetic overactivity via renal denervation (RDN) with the aim to enhance arterial compliance. We sought to non-invasively quantify pulse wave reflections changes in patients undergoing RDN.
METHODS: Patients who underwent RDN for resistant hypertension were classified as patients with HFpEF and patients without

Stress has profound effects on health, yet how it damages tissues remains poorly understood. Here, we show that acute stress triggers rapid hair loss and initiates autoimmunity. Under stress, hyperactivated sympathetic nerves release excessive norepinephrine, causing necrosis in rapidly dividing hair follicle transit-amplifying cells (HF-TACs) while sparing most hair follicle stem cells (HFSCs). This differential sensitivity stems from differences in cell death pathways, metabolic strategies, and calcium homeostasis, which render HF-TACs more susceptible to norepinephrine-induced calcium surge

BACKGROUND: Cardiac stereotactic radiation therapy is a potential treatment for refractory ventricular tachycardia. We previously reported that carbon ion irradiation, with a higher dose concentration than x-rays, restored gap junction proteins and denervated excess sympathetic nerves in a rabbit heart model. However, histologic analysis is limited to endpoint evaluations, requiring in vivo functional assessments.
OBJECTIVE: This study aimed to evaluate the effects of heavy ion therapy on mitochondrial and sympathetic nerve function in a rabbit model of hypercholesterolemia (HC), showing symp

PURPOSE: Vasovagal syncope is thought to be mediated by a progressive fall in cardiac output secondary to venous pooling of blood in the splanchnic circulation. How and when this occurs before syncope has not been determined.
METHODS: A total of 20 patients who became hypotensive during head-up tilt (age 40.9 ± 3.4 years; 10 females) were divided into two groups-the glyceryl trinitrate (GTN) group (n = 12) and the vasovagal syncope (VVS) group (n = 8) - on the basis of whether or not nitroglycerine provocation was required. They were compared with a control group (age 38.6 ± 3.3; 8 females; n

We investigated second-messenger signalling components linked to the stimulation of Gq protein-coupled receptors (e.g. thromboxane A2 and bradykinin B2 receptors) on the sensory endings of thin fibre muscle afferents in the chronic mechanoreflex sensitization in rats with myocardial infarction-induced heart failure with reduced ejection fraction (HF-rEF). We hypothesized that injection of either the inositol 1,4,5-trisphosphate (IP3) receptor antagonist xestospongin C (5 µg) or the PKCε translocation inhibitor PKCe141 (45 µg) into the arterial supply of the hindlimb would reduce the increase i

BACKGROUND: A growing body of evidence suggests that oxidative stress plays a role in the pathophysiology of hypertension. However, the involvement of the reactive oxygen species (ROS) in the commissural nucleus of the solitary tract (commNTS) in development the of hypertension remains unclear.
METHOD: We evaluated the hemodynamic and sympathetic responses to acute inhibition of NADPH oxidase in the commNTS in renovascular hypertensive rats. Under anesthesia, male Holtzman rats were implanted with a silver clip around the left renal artery to induce 2-kidney 1-clip (2K1C) hypertension. After

Both voluntary rebreathing (RB) of expired air and voluntary apneas (VA) elicit changes in arterial carbon dioxide and oxygen (CO2 and O2) chemostimuli. These chemostimuli elicit synergistic increases in cerebral blood flow (CBF) and sympathetic nervous system activation, with the latter increasing systemic blood pressure. The extent that simultaneous and inverse changes in arterial CO2 and O2 and associated increases in blood pressure affect the CBF responses during RB versus VAs are unclear. We instrumented 21 healthy participants with a finometer (beat-by-beat mean arterial blood pressure;

BACKGROUND: Initial orthostatic hypotension (IOH) is defined by a large drop in blood pressure (BP) within 15 s of standing. IOH often presents during an active stand, but not with a passive tilt, suggesting that a muscle activation reflex involving lower body muscles plays an important role. To our knowledge, there is no literature exploring how sympathetic activation affects IOH. We hypothesized involuntary muscle contractions before standing would significantly reduce the drop in BP seen in IOH while increasing sympathetic activity would not.
METHODS: Study participants performed 4 sit-to-

Sympathetic vasoconstriction is mediated by α-adrenergic receptors under resting conditions. During exercise, increased sympathetic nerve activity (SNA) is directed to inactive and active skeletal muscle; however, it is unclear what mechanism(s) are responsible for vasoconstriction during large muscle mass exercise in humans. The aim of this study was to determine the contribution of α-adrenergic receptors to sympathetic restraint of inactive skeletal muscle and active skeletal muscle during cycle exercise in healthy humans. In ten male participants (18-35 yr), mean arterial pressure (intra-ar

Previous studies have been described changes in brain regions contributing to the sympathetic vasomotor overactivity in Goldblatt hypertension (2K1C). Furthermore, changes in the spinal cord are also involved in the cardiovascular and autonomic dysfunction in renovascular hypertension, as intrathecal (i.t.) administration of Losartan (Los) causes a robust hypotensive/sympathoinhibitory response in 2K1C but not in control rats. The present study evaluated the role of spinal γ-aminobutyric acid (GABA)-ergic inputs in the control of sympathetic vasomotor activity in the 2K1C rats. Hypertension wa
