Graves' disease (GD) is an autoimmune disorder of the thyroid gland and both environmental and genetic factors contribute to disease aetiology. Cytokines, such as interleukin 4 (IL-4) and interleukin 10 (IL-10), are involved in the immune response and may be implicated in the autoimmune disease process. Associations have been reported between single nucleotide polymorphisms (SNPs) of IL-10 and the Ile50Val polymorphism of the IL-4 receptor gene (IL-4R) gene and atopy and autoimmune diseases such as systemic lupus erythematosus and rheumatoid arthritis. The autoimmune diseases cluster within fa

BACKGROUND: Chronic spontaneous urticaria (CSU) is a systemic immune-mediated disorder frequently comorbid with autoimmune thyroid disease, particularly Hashimoto's thyroiditis (HT). Emerging evidence implicates shared immunogenetic pathways, yet the role of thymic stromal lymphopoietin (TSLP), a key epithelial alarmin in this comorbidity remains unclear.
OBJECTIVE: To investigate the association between TSLP SNPs and the co-occurrence of CSU and HT.
METHODS: We enrolled 45 patients with CSU alone and 41 with CSU and concomitant HT from a reference center in the Colombian Caribbean. Diagnosi

BACKGROUND: Latent autoimmune diabetes in adults (LADA) is recognized as the most prevalent form of autoimmune diabetes in adulthood marked by a long-term preservation of endogenous insulin secretion capacity. Renal hypouricemia type 2 (RHUC2) is a rare genetic disorder characterized by defective uric acid reabsorption which results from variants in the SLC2A9 gene. Currently, there are no reported cases of RHUC2 combined with LADA. CASE PRESENTATION: A 39-year-old servicewoman visited our hospital with the complaint of elevated blood glucose for 2 years and polydipsia, polyuria, numb right th

BACKGROUND AND AIM: Autoimmune gastritis (AIG) was previously considered a rare disease in China, and its clinical characteristics were not fully understood. This study aimed to demonstrate the characteristics of AIG in China and evaluate gastric oxyntic mucosal atrophy using a modified AIG-atrophic staging.
METHODS: This was a single-center retrospective observational real-world study. The diagnosis of AIG was based on pathological results combined with parietal cell antibody (PCA) and intrinsic factor antibody (IFA) results, and endoscopic findings.
RESULTS: A total of 745 patients were en

RATIONALE: A 62-year-old male patient was admitted to our clinic in February 2016 with persistently elevated liver enzymes.
PATIENT CONCERNS: Clinical history involved a long time of poly-autoimmunity with a rheumatoid arthritis (in remission under tocilizumab therapy), an autoimmune thyroiditis, an eosinophilia as well as a hyper-immunoglobulin (IgG) 4-syndrome.
DIAGNOSES: Laboratory studies revealed a significant increase in liver enzymes with an alanine aminotransferase (ALT) level of 574 U/L and an aspartate aminotransferase (AST) level of 864 U/L (normal <50 U/L). Furthermore, the patie

Chronic spontaneous urticaria (CSU) is a common mast cell-driven disease characterized by the development of wheals (hives), angioedema (AE), or both for > 6 weeks. It is thought that autoimmunity is a common cause of CSU, which is often associated with autoimmune thyroiditis, whereas the link to other autoimmune disorders such as systemic lupus erythematosus (SLE) has not been carefully explored. Here, we systematically reviewed the existing literature for information on the prevalence of CSU in SLE (and vice versa) and we examined the possible clinical and pathogenetic relationship between C

In the present study, we describe the in vitro transcription-translation of human tyrosinase-related protein-1 (TRP-1) cDNA and subsequent use of the resulting 35S-labelled TRP-1 in a radioimmunoassay to analyse vitiligo sera for the presence of TRP-1 antibodies. Of 53 vitiligo sera examined in the assay, three (5.7%) were found to be positive for TRP-1 antibodies. In contrast, sera from 20 controls, 10 patients with Hashimoto's thyroiditis and 10 patients with Graves' disease were all negative for TRP-1 antibodies. Although glycosylation of the labelled protein was necessary for its immunopre

A patient developed insulin-dependent diabetes mellitus at the age of 9 years, complicated by a sensory/autonomic polyneuropathy which presented with a respiratory arrest at the age of 41 years. The neuropathy increased in severity over the subsequent two decades. At the age of 52 years she had hypopituitarism, hypothyroidism and low normal adrenal function. Autopsy at the age of 59 years revealed loss of pituitary tissue with evidence of hypophysitis, a lymphocytic thyroiditis and severe adrenal atrophy with lymphocytic infiltration of the medulla. The pancreas showed reduced numbers and size

Thyroid diseases are the commonest endocrine disorders in the general population. In most of the cases, they are consistent with benign conditions which may be asymptomatic or affect people at a variable extent. Since they often represent chronic conditions their prevalence increases by age and reaches in elderly the highest rates. Thyroid nodules are a common clinical finding. Most subjects with thyroid nodules have few or no symptoms. Thyroid nodules are more commonly non-functioning. However, in elderly, toxic multinodular goiter is the most frequent cause of spontaneous hyperthyroidism and

INTRODUCTION: The interplay between genetic and environmental factors plays a critical role in the development of type 1 diabetes (T1D). While variants in non-HLA coding regions have been identified as potential therapeutic targets, their exact contribution to the pathophysiology of disease remains unclear. The aim of the study was to characterize six non-HLA variants in pediatric patients with T1D and explore their potential association with clinical parameters and other autoimmune diseases, such as thyroiditis and celiac disease (CD).
METHODS: We analyzed six variants (c.1858T>C, c.49A>G, c

INTRODUCTION: Immune checkpoint inhibitors (ICIs), such as nivolumab, have made significant advancements in the treatment of several malignancies. However, they have been associated with a new and diverse spectrum of immune-related adverse effects (irAEs). We report the case of a patient with metastatic renal cell carcinoma who developed encephalitis and other potential irAEs, including hypophysitis with isolated adrenocorticotropic hormone deficiency, transient autoimmune thyroiditis, and autoimmune hemolytic anemia, following combination therapy with nivolumab and cabozantinib. The patient u

A 47 year old Caucasian female with a background of type 2 diabetes mellitus, hypothyroidism, and autoimmune hepatitis presented with a painful, pruritic, papular eruption in a photosensitive distribution across the upper chest, neck, face, dorsal hands and forearms. On examination, lesions coalesced into annular plaques each with an active, raised margin and an atrophic, yellow centre. Histopathology demonstrated an absence of mucin and elastophagocytosis with giant cells engulfing dermal elastin fibres. These histopathological features favoured a diagnosis of annular elastolytic giant cell g

Type 1 diabetes mellitus (T1DM) and multiple sclerosis (MS) have been described as chronic organ-specific diseases, approached by different medical specialties. However, they share more etiologic and pathologic features than expected between two autoimmune diseases. The authors present the case of a 40-year-old Caucasian male, diagnosed with type 1 diabetes mellitus at age 18, with poor metabolic control in the early years after the diagnosis. Fourteen years after the diagnosis of diabetes, he started complaining of paresthesias in both feet and sexual dysfunction. Months later, he began to ha

Combination immunotherapy targeting the PD-1 and CTLA-4 checkpoint inhibitor pathways provides substantial clinical benefit in patients with advanced-stage cancer but at the risk of dose-limiting inflammatory and autoimmune toxicity. The delicate balance that exists between unleashing tumor killing and promoting systemic autoimmune toxicity represents a major clinical challenge. We hypothesized that targeting anti-CTLA-4 so that it perfuses tumor-draining lymph nodes would provide a significant therapeutic advantage and developed an injectable hydrogel with controlled antibody release characte
