Chimeric antigen receptor macrophages (CAR-M) sensitize HER2+ solid tumors to PD1 blockade in pre-clinical models.

We previously developed human CAR macrophages (CAR-M) and demonstrated redirection of macrophage anti-tumor function leading to tumor control in immunodeficient xenograft models. Here, we develop clinically relevant fully immunocompetent syngeneic models to evaluate the potential for CAR-M to remodel the tumor microenvironment (TME), induce T cell anti-tumor immunity, and sensitize solid tumors to PD1/PDL1 checkpoint inhibition. In vivo, anti-HER2 CAR-M significantly reduce tumor burden, prolong survival, remodel the TME, increase intratumoral T cell and natural killer (NK) cell infiltration, …

Gasotransmitter-Nanodonor for Spatial Regulation of Anxiety-Like Behavior and Bone Metastasis.

Anxiety is highly prevalent among cancer patients, significantly impacting their prognosis. Current cancer therapies typically lack anxiolytic properties and may even exacerbate anxiety. Here, a gasotransmitter-nanodonors (GND) system is presented that exerts dual anxiolytic and anti-tumor effects via a "tumor-brain axis" strategy. The GND, synthesized by co-embedding Fe2⁺ and S2⁻ ions along with glucose oxidase (GOx) within bovine serum albumin (BSA) nanoparticles (FSG@AB), enables the controlled release of the gasotransmitter hydrogen sulfide (H₂S) in the acidic tumor microenvironment. H₂S a…

In vivo CRISPR screens identify Mga as an immunotherapy target in triple-negative breast cancer.

Immune evasion is not only critical for tumor initiation and progression, but also determines the efficacy of immunotherapies. Through itera … le-cell RNA sequencing analyses revealed that Mga influences various immune-related pathways in the tumor microenvironment. Our findings suggest that Mga may play a role in modulating the tumor immune landscape, though the precise mechanisms require further investigation. Interestingly, we observed that low MGA expression in breast cancer patients correlates with a favo …

Targeting pancreatic cancer immune evasion by inhibiting histone deacetylases.

The immune system plays a vital role in maintaining the delicate balance between immune recognition and tumor development. Regardless, it is not uncommon that cancerous cells can intelligently acquire abilities to bypass the antitumor immune responses, thus allowing continuous tumor growth and development. Immune evasion has emerged as a significant factor contributing to the progression and immune resistance of pancreatic cancer. Compared with other cancers, pancreatic cancer has a tumor microenvironment that can resist most treatment modalities, including emerging immunotherapy. Sadly, the u…

Liposomal oxaliplatin prodrugs loaded with metformin potentiate immunotherapy for colorectal cancer.

Tumor hypoxia is confirmed to be associated with the formation of tumor immunosuppression, a general feature of solid tumors, and thus attenuates the effectiveness of various cancer therapies in clinic. We herein develop a tumor microenvironment (TME) modulating liposomal nanomedicine by encapsulating metformin with amphiphilic oxaliplatin prodrug constructed liposomes to potentiate cancer immunotherapy. While metformin could regulate metabolisms of tumor cells to reduce their oxygen consumption and relieve tumor hypoxia, oxaliplatin is a chemotherapy drug that induces immunogenic cell death (…

Host autophagy mediates organ wasting and nutrient mobilization for tumor growth.

During tumor growth-when nutrient and anabolic demands are high-autophagy supports tumor metabolism and growth through lysosomal organelle turnover and nutrient recycling. Ras-driven tumors additionally invoke non-autonomous autophagy in the microenvironment to support tumor growth, in part through transfer of amino acids. Here we uncover a third critical role of autophagy in mediating systemic organ wasting and nutrient mobilization for tumor growth using a well-characterized malignant tumor model in Drosophila melanogaster. Micro-computed X-ray tomography and metabolic profiling reveal that

Hypoxia Inducible Factors' Signaling in Pediatric High-Grade Gliomas: Role, Modelization and Innovative Targeted Approaches.

The brain tumor microenvironment has recently become a major challenge in all pediatric cancers, but especially in brain tumors like high-grade gliomas. Hypoxia is one of the extrinsic tumor features that interacts with tumor cells, but also with the blood-brain barrier and all normal brain cells. It is the result of a dramatic proliferation and expansion of tumor cells that deprive the tissues of oxygen inflow. However, cancer cells, especially tumor stem cells, can endure extreme hypoxic conditions by rescheduling various genes' expression involved in cell proliferation, metabolism and angio…

Metformin - its potential anti-cancer and anti-aging effects.

Organoids are a class of multicellular structures with the capability of self-organizing and the characteristic of original tissues, they are generated from stem cells in 3D culture in vitro. Organoids can mimic the occurrence and progression of original tissues and widely used in disease models in recent years. The ability of tumor organoids to retain characteristic of original tumors make them unique for tumorigenesis and cancer therapy. However, the history of organoid development and the application of organoid technology in cancer therapy are not well understood. In this paper, we reviewe

[Development of incurable cancer cell targeted agents using viral technology].

The safety and anti-tumor effect of oncolytic virus have been reported in a clinical study conducted in Japan. We have engineered a novel multimutated tumor-specific oncolytic herpes virus, harboring a smooth muscle-specific calponin promoter. Since tumor cells present in a hypoxic environment are known to be resistant to chemotherapy and radiotherapy, we also engineered a novel oncolytic herpes virus targeting a specific tumor microenvironment, which harbors a gene encoding a fusion protein of oxygen-dependent degradation (ODD) domain of HIF1α and ICP4, a master viral transcription factor req…

Congenital adrenal hyperplasia.

The tumor stroma, which is essential to support growth and metastasis of malignant cells, provides targets for active immunotherapy of cance … ession of the co-inhibitor PD-1. In two mouse melanoma models, depleting FAP+ stroma cells from the tumor microenvironment (TME) upon vaccination with an adenoviral-vector reduces frequencies and functions of ISCs. This is associated with changes in the cytokine/chemokine milieu in the TME and decreased activity of STAT6 signaling within ISCs. Decreases in ISCs upon FAP …

Combining CAI with 1-MT or DMF disrupted PD-1 expression and promoted IFN-γ production in CD8+ T cells, and it also increased T lymphocyte infiltration in the tumor microenvironment, inhibited tumor growth …

A Cascade-Immunomodulating Poly(amino acid)-Metal Coordination Nanoplatform for Enhanced Cancer Immunotherapy.

The limited efficacy of cancer immunotherapy arises not only from insufficient immune activation, but more fundamentally from the absence of a systems-level strategy to coordinate the anti-cancer immunity cycle. Here, a poly(amino acid)-metal coordination nanoplatform comprising methoxy poly(ethylene glycol)-poly(glutamic acid), manganese ion (Mn2+), and quercetin (Quer) is engineered to orchestrate the cancer-immunity cycle dynamically. Mn2+-mediated Fenton-like reaction induces immunogenic cell death and fosters immune-activating tumor microenvironments (TMEs), as supported by a 1.7-fold inc…

Why only 3 months? Immunological constraints on CAR-T therapy in gastrointestinal cancers.

In June 2026, satri-cel/CT041, a chimeric antigen receptor (CAR) T-cell product targeting Claudin18.2, received approval from China's National Medical Products Administration for the treatment of Claudin18.2-positive, HER2-negative advanced gastric/gastroesophageal junction adenocarcinoma after failure of at least two prior lines of therapy. This marks the first CAR-T product approved worldwide for solid tumours. The approval ends the long-standing debate over whether CAR-T cells can be effective against solid malignancies, yet it simultaneously raises a more pointed issue: in its pivotal rand

Niches in cancer: from primary tumors to metastatic disease.

Cancer progression does not occur in isolation: malignant-cell states and fates are shaped by specialized microenvironments, or cancer niches. Within primary tumors, vascular, stromal, immune and neural interactions create local hypoxic, metabolic and mechanical conditions that together influence malignant-cell behavior. These local environments sustain stem-like states, regulate immune surveillance and vascular remodeling, and influence dissemination and treatment response. Tumor-derived extracellular vesicles and other systemic signals connect primary tumors to distant organs, conditioning t

Evasive encounters: Immunomodulatory CAFs interact with Tregs in the lung cancer microenvironment.

Ringham et al.1 identify CHL1+CXCL9+ cancer-associated fibroblasts at lung tumor margins that recruit suppressive CXCR3+ regulatory T cells, revealing how stromal chemokine gradients can promote immune evasion. Viewed alongside immune-stimulatory CCL19+ fibroblastic reticular cells, these findings suggest that immune-interacting fibroblasts are context-dependent organizers of tumor immunity.

Exploring tumor-derived exosomes as promising antigenic platforms: novel vaccine and immunotherapy strategies for "cold" colorectal cancer.

Colorectal cancer (CRC) remains a formidable clinical challenge, largely due to the prevalence of microsatellite-stable tumors characterized by an immunologically "cold" tumor microenvironment (TME) that is resistant to immune checkpoint inhibitors (ICIs). Tumor-derived exosomes (TEXs) play a paradoxical dual role in this landscape. Native TEXs actively orchestrate immune evasion by delivering immunosuppressive molecules, such as programmed cell death 1 ligand 1 (PD-L1) and transforming growth factor-beta (TGF-β), which impair cytotoxic T-cell function and promote the expansion of regulatory T…

Spatiotemporal transcriptomics for tumour microenvironment remodelling in cancer progression and therapy.

BACKGROUND: Spatiotemporal transcriptomics (STT) integrates spatially resolved gene expression with temporal information derived from staged samples, paired treatment specimens, or computational inference, providing a framework for reconstructing tissue-level dynamics. In the tumour microenvironment (TME), STT extends static molecular and cellular maps by revealing how cellular neighbourhoods and functional niches are reorganised during tumour evolution. MAIN TEXT: This review examines how STT characterises TME remodelling during malignant transformation, invasion, metastasis, ageing, and tre

Neuron-tumor interplay in colorectal cancer: from mechanisms of onset and progression to targeted therapies.

Colorectal cancer (CRC) is a major cause of cancer-related mortality worldwide, traditionally attributed to genetic mutations and epigenetic alterations. However, with the emergence of cancer neuroscience, a paradigm shift has occurred, revealing the central role of the nervous system in the initiation, progression, and metastasis of CRC. The gastrointestinal tract, being the only organ in the body with an independent peripheral nervous system - the enteric nervous system (ENS) - provides a unique anatomical and functional basis for neuro-tumor interactions. This review systematically explores
