Occupational Infection Prevention Among Nurses and Laboratory Technicians Amidst Multiple Health Emergencies in Outbreak-Prone Country, D.R. Congo.

Millions of healthcare workers experience percutaneous exposure to bloodborne communicable infectious disease pathogens annually, with the risk of contracting occupationally acquired infections. In this study, we aimed to assess the status of occupational safety and outbreak preparedness in Congolese nurses and laboratory technicians in Kongo central and the Katanga area, amidst multiple ongoing public health emergencies in the Democratic Republic of the Congo (DRC). This was a multicenter analytical cross-sectional study conducted in five referral hospitals located in Kongo central province a

Pediatric simple triage score: A simplified approach for triaging pediatric patients with fever in the emergency department.

INTRODUCTION: The initial 24-h period following admission to a hospital holds profound significance for pediatric patients, representing a critical window where proactive interventions can substantially influence outcomes. We devised a simple triage system, pediatric simple triage score (PSTS), to see whether rapid triage of sick pediatric patients with fever can be done using the new triage system in the emergency department (ED) to predict hospital admission. METHODS: This was a prospective observational study, conducted at the department of emergency medicine of a tertiary care teaching ho

Ticks without borders: Microbial communities of immature Neotropical tick species parasitizing migratory landbirds along northern Gulf of Mexico.

The long-distance, seasonal migrations of birds make them an effective ecological bridge for the movement of ticks. The introduction of exotic tick species to new geographical regions can lead to the emergence of novel tick-borne pathogens or the re-emergence of previously eradicated ones. This study assessed the prevalence of exotic tick species parasitizing resident, short-distance, and long-distance songbirds during spring and autumn at stopover sites in the northern Gulf of Mexico using the mitochondrial 12S rDNA gene. Birds were captured for tick collection from six different sites from l

Louis Pasteur-The life of a controversial scientist with a prepared mind, driven by curiosity, motivation, and competition.

Louis Pasteur, born December 27, 1822 in Dole, France, showed in his childhood and youth great abilities as an artistic painter; however by an age of 19, his interest changed toward science, and he moved to Paris to study chemistry and physics at École Normale Supérieure. During graduation, he initiated research on chiral crystallography and stereochemistry and got his doctorates in 1847 in both chemistry and physics. In 1848, he started as high school teacher in Dijon, but shortly after he became a deputy professor at the University of Strasbourg in chemistry and got married to the rector's d

Antigenic Characterization of New Lineage II Insect-Specific Flaviviruses in Australian Mosquitoes and Identification of Host Restriction Factors.

We describe two new insect-specific flaviviruses (ISFs) isolated from mosquitoes in Australia, Binjari virus (BinJV) and Hidden Valley virus (HVV), that grow efficiently in mosquito cells but fail to replicate in a range of vertebrate cell lines. Phylogenetic analysis revealed that BinJV and HVV were closely related (90% amino acid sequence identity) and clustered with lineage II (dual-host affiliated) ISFs, including the Lammi and Nounané viruses. Using a panel of monoclonal antibodies prepared to BinJV viral proteins, we confirmed a close relationship between HVV and BinJV and revealed that

Role of Zika Virus Envelope Protein Domain III as a Target of Human Neutralizing Antibodies.

Zika virus (ZIKV) is a flavivirus that is structurally highly similar to the related viruses, dengue virus (DENV), West Nile virus, and yellow fever virus. ZIKV causes an acute infection that often results in mild symptoms but that can cause severe disease in rare instances. Following infection, individuals mount an adaptive immune response, composed of antibodies (Abs) that target the envelope (E) glycoprotein of ZIKV, which covers the surface of the virus. Groups have studied monoclonal antibodies and polyclonal immune sera isolated from individuals who recovered from natural ZIKV infections

Aedes Anphevirus: an Insect-Specific Virus Distributed Worldwide in Aedes aegypti Mosquitoes That Has Complex Interplays with Wolbachia and Dengue Virus Infection in Cells.

Insect-specific viruses (ISVs) of the yellow fever mosquito Aedes aegypti have been demonstrated to modulate transmission of arboviruses such as dengue virus (DENV) and West Nile virus by the mosquito. The diversity and composition of the virome of A. aegypti, however, remains poorly understood. In this study, we characterized Aedes anphevirus (AeAV), a negative-sense RNA virus from the order Mononegavirales AeAV identified from Aedes cell lines was infectious to both A. aegypti and Aedes albopictus cells but not to three mammalian cell lines. To understand the incidence and genetic diversity

Vaccination with Gag, Vif, and Nef gene fragments affords partial control of viral replication after mucosal challenge with SIVmac239.

UNLABELLED: Broadly targeted cellular immune responses are thought to be important for controlling replication of human and simian immunodeficiency viruses (HIV and SIV). However, eliciting such responses by vaccination is complicated by immunodominance, the preferential targeting of only a few of the many possible epitopes of a given antigen. This phenomenon may be due to the coexpression of dominant and subdominant epitopes by the same antigen-presenting cell and may be overcome by distributing these sequences among several different vaccine constructs. Accordingly, we tested whether vaccina

Patent literature on mosquito repellent inventions which contain plant essential oils--a review.

Bites Bites of mosquitoes belonging to the genera Anopheles Meigen, Aedes Meigen, Culex L. and Haemagogus L. are a general nuisance and are responsible for the transmission of important tropical diseases such as malaria, hemorrhagic dengue and yellow fevers and filariasis (elephantiasis). Plants are traditional sources of mosquito repelling essential oils (EOs), glyceridic oils and repellent and synergistic chemicals. A Chemical Abstracts search on mosquito repellent inventions containing plant-derived EOs revealed 144 active patents mostly from Asia. Chinese, Japanese and Korean language pate

Vaccinations administered during off-clinic hours at a national community pharmacy: implications for increasing patient access and convenience.

PURPOSE: Approximately 50,000 adults die annually from vaccine-preventable diseases in the United States. Most traditional vaccine providers (eg, physician offices) administer vaccinations during standard clinic hours, but community pharmacies offer expanded hours that allow patients to be vaccinated at convenient times. We analyzed the types of vaccines administered and patient populations vaccinated during off-clinic hours in a national community pharmacy, and their implications for vaccination access and convenience. METHODS: We retrospectively reviewed data for all vaccinations given at t

Temporal dissection of hepatitis C virus and orthoflavivirus infections by continuous luminescence monitoring.

Virus infection is a dynamic process consisting of rapidly evolving viral and host events, yet conventional endpoint assays provide only fragmented snapshots of these processes. Here, we established a unified temporal framework for dissecting virus-host interactions by integrating continuous bioluminescence monitoring with molecular and functional analyses. Real-time monitoring of hepatitis C virus (HCV) single-round infectious particles (SRIPs) revealed a biphasic replication profile consisting of an initial transient phase, followed by replication-dependent amplification. This transition was

Physical and chemical characteristics of Aedes larval habitats in Metema District, Northwest, Ethiopia.

BACKGROUND: Aedes-transmitted viral diseases such as dengue, chikungunya, yellow fever, and Zika are increasing public health and economic challenge globally. In the absence of treatments and vaccines for arboviral diseases, surveillance and control of Aedes larvae remains a top priority. However, Aedes larvae control strategies relay to a large extent on the knowledge of the distribution and characteristics of their habitats. The study assessed habitats, indices, habitat physico-chemical characteristics and composition of Aedes larvae and pupae in three towns. METHODS: Aedes larvae and pupae

Schistosome and malaria exposure and urban-rural differences in vaccine responses in Uganda: a causal mediation analysis using data from three linked randomised controlled trials.

BACKGROUND: Vaccine immunogenicity and effectiveness vary geographically. Chronic immunomodulating parasitic infections including schistosomes and malaria have been hypothesised to be mediators of geographical variations. METHODS: We compared vaccine-specific immune responses between three Ugandan settings (schistosome-endemic rural, malaria-endemic rural, and urban) and did causal mediation analysis to assess the role of Schistosoma mansoni and malaria exposure in observed differences. We used data from the control groups of three linked randomised trials investigating the effects of intensi

Lives saved with vaccination for 10 pathogens across 112 countries in a pre-COVID-19 world.

BACKGROUND: Vaccination is one of the most effective public health interventions. We investigate the impact of vaccination activities for Haemophilus influenzae type b, hepatitis B, human papillomavirus, Japanese encephalitis, measles, Neisseria meningitidis serogroup A, rotavirus, rubella, Streptococcus pneumoniae, and yellow fever over the years 2000-2030 across 112 countries. METHODS: Twenty-one mathematical models estimated disease burden using standardised demographic and immunisation data. Impact was attributed to the year of vaccination through vaccine-activity-stratified impact ratios

Alignment-free k-mer-guided design of a pan-Orthoflavivirus RT-qPCR assay.

The co-circulation and rapid expansion of the genus Orthoflavivirus, including dengue virus (DENV), Zika virus (ZIKV), West Nile virus (WNV), Yellow fever virus (YFV), and Japanese encephalitis virus (JEV), pose significant global health challenges. Developing inclusive pan-genus molecular diagnostics is hindered by high nucleotide divergence (>25-30%) and the computational limitations of traditional multiple sequence alignment in detecting conserved motifs across large data sets. To overcome these limitations, we developed a systematic alignment-free design pipeline that uses rigorous k-mer a

Constructing a predictive model for early-onset sepsis in neonatal intensive care unit newborns based on SHapley Additive exPlanations explainable machine learning.

BACKGROUND: The clinical characteristics of neonatal sepsis (NS) are subtle and non-specific, posing a serious threat to the lives of newborn infants. Early-onset sepsis (EOS) is sepsis that occurs within 72 hours after birth, with a high mortality rate. Identifying key factors of NS and conducting early diagnosis are of great practical significance. Thus, we developed a robust machine learning (ML) model for the early prediction of EOS in neonates admitted to the neonatal intensive care unit (NICU), investigated the pivotal risk factors associated with EOS development, and provided interpreta

Flavivirus NS1-triggered endothelial dysfunction promotes virus dissemination.

UNLABELLED: The Flaviviridae are a family of viruses that include the important arthropod-borne human pathogens dengue virus (DENV), West Nile virus, Zika virus, Japanese encephalitis virus, and yellow fever virus. Flavivirus nonstructural protein 1 (NS1) is essential for virus replication but is also secreted from virus-infected cells. Extracellular NS1 acts as a virulence factor during flavivirus infection in multiple ways, including triggering endothelial dysfunction and vascular leak via interaction with endothelial cells. While the role of NS1 in inducing vascular leak and exacerbating pa
