Antiobesity Effect of Garlic Extract Fermented by Lactobacillus plantarum BL2 in Diet-Induced Obese Mice.

Obesity is viewed as a serious public health problem. This study aimed to investigate the antiobesity effects of fermented garlic extract by lactic acid bacteria (LAFGE) on obesity. Male C57BL/6J mice were fed with high-fat diet (HFD) to induce obesity. The HFD-induced obese mice were orally administrated with 250 or 500 mg/kg LAFGE for 8 weeks. Feeding HFD-fed mice with 250 or 500 mg/kg LAFGE reduced body weight by 14% and 18%, respectively, compared to HFD. HFD-fed mice with 500 mg/kg LAFGE administration had lower epididymal, retroperitoneal, and mesenteric adipose tissue mass by 36%, 44%, …

Adipokines and the female reproductive tract.

It is well known that adipose tissue can influence puberty, sexual maturation, and fertility in different species. Adipose tissue secretes molecules called adipokines which most likely have an endocrine effect on reproductive function. It has been revealed over the last few years that adipokines are functionally implicated at all levels of the reproductive axis including the gonad and hypothalamic-pituitary axis. Many studies have shown the presence and the role of the adipokines and their receptors in the female reproductive tract of different species. These adipokines regulate ovarian steroi…

Bone metabolism and adipokines: are there perspectives for bone diseases drug discovery?

INTRODUCTION: Over the past 20 years, the idea that white adipose tissue (WAT) is simply an energy depot organ has been radically changed. Indeed, present understanding suggests WAT to be an endocrine organ capable of producing and secreting a wide variety of proteins termed adipokines. These adipokines appear to be relevant factors involved in a number of different functions, including metabolism, immune response, inflammation and bone metabolism. AREAS COVERED: In this review, the authors focus on the effects of several adipose tissue-derived factors in bone pathophysiology. They also consi…

Excess nutrient supply in early life and its later metabolic consequences.

Suboptimal nutrition in early life, both in utero and during infancy, is linked to increased risk of adult obesity and its associated advers … during early life include resetting of hypothalamic energy sensing and appetite regulation, altered adipose tissue insulin sensitivity and impaired brown adipose tissue function. More detailed understanding of the mechanisms involved in metabolic programming could enable the development of therapeutic strategies for ameliorating its ill effects. Research in this …

Cord blood chemerin and obestatin levels in large for gestational age infants.

OBJECTIVE: To investigate possible alterations in cord blood levels of adipokines, chemerin and obestatin (secreted by adipose tissue and associated with later development of insulin resistance/metabolic syndrome), as well as insulin, in large for gestational age (LGA) and appropriate for gestational age (AGA) pregnancies, granted that these groups differ in body fat mass and metabolic/endocrine mechanisms. METHODS: Cord blood chemerin, obestatin, and insulin concentrations were prospectively measured in 40 LGA (9 born from diabetic and 31 from nondiabetic mothers) and 40 AGA singleton full-t…

Tissue-autonomous function of Drosophila seipin in preventing ectopic lipid droplet formation.

Obesity is characterized by accumulation of excess body fat, while lipodystrophy is characterized by loss or absence of body fat. Despite their opposite phenotypes, these two conditions both cause ectopic lipid storage in non-adipose tissues, leading to lipotoxicity, which has health-threatening consequences. The exact mechanisms underlying ectopic lipid storage remain elusive. Here we report the analysis of a Drosophila model of the most severe form of human lipodystrophy, Berardinelli-Seip Congenital Lipodystrophy 2, which is caused by mutations in the BSCL2/Seipin gene. In addition to reduc…

Olanzapine, but not aripiprazole, weight-independently elevates serum triglycerides and activates lipogenic gene expression in female rats.

Metabolic adverse effects such as weight gain and dyslipidaemia represent a major concern in treatment with several antipsychotic drugs, inc … a rat model, we showed that subchronic treatment with olanzapine induces weight gain and increases adipose tissue mass in rats with free access to food. This effect was also observed for aripiprazole, considered metabolically neutral in the clinical setting. In pair-fed rats with limited food access, neither olanzapine nor aripiprazole induced weight gain. Inte …

Management of obesity in the prevention of cardiovascular disease.

The prevalence of obesity is increasing in the United States and many other countries, and has now passed smoking as the most preventable cause of morbidity and mortality. Obesity is an excess of adipose tissue that results from a mixture of genetic predisposition, environmental influences (e.g., sedentary lifestyle), and behavioural components (e.g., food as a reward), and it is a chronic, relapsing condition. The rapid increase in obesity prevalence is, however, not due to genetic changes but rather is a societal mismatch between physiology and environment, where food is abundant and exercis…

Giant hypopharyngeal atypical lipomatous tumor.

The so-called "giant fibrovascular polyps" of the esophagus and hypopharynx typically present as sausage-like pedunculated structures that protrude into the lumen and cause obstructive symptoms. Most are cured by local resection but they may recur. Microscopically, they display an admixture of fibrovascular and adipose tissue that is coated by unremarkable squamous mucosa. Here, we report a case that had scattered hyperchromatic cells and lipoblasts within the adipose tissue component. In other anatomic sites similar appearing lesions have been interpreted as pedunculated liposarcomas/atypical…

Reduced adiposity and high-fat diet-induced adipose inflammation in mice deficient for phosphodiesterase 4B.

The concept that obesity is an inflammatory state has changed our understanding of this condition and suggested that pharmacological interve … iets (HFDs). PDE4B deficiency suppressed TNF-alpha mRNA levels and macrophage infiltration in white adipose tissue in mice on HFD, but insulin sensitivity was unaltered. PDE4B-null mice on HFDs had increased locomotor activity. These results suggest a previously unappreciated role for PDE4B in the regulation of energy balance and that PDE4B inhibitors could have …

A mouse model of conditional lipodystrophy.

Lipodystrophies are syndromes of adipose tissue degeneration associated with severe defects in lipid and glucose homeostasis. We report here the generation and analysis of Pparg(ldi), a targeted allele that confers conditional dominant lipodystrophy in mice. The Pparg(ldi) allele was generated by insertion of the Tet activator (tTA) and a tTA-regulated Flag-Pparg1 transgene into the Pparg gene. Unexpectedly, tTA elicits mild lipodystrophy, insulin resistance, and dyslipidemia, and the Flag-PPARgamma1 transgene surprisingly exacerbates these traits. Doxycycline can both completely prevent and r…

True thymic hyperplasia associated with a unilocular thymic cyst: an unusual combination not previously reported.

A mediastinal tumor mass in a 53-year-old woman with the sole complaint of persistent cough was studied and found to be a true thymic hyperplasia associated with a unilocular thymic cyst, felt to be of the congenital type. The bilobed thymus obtained was macroscopically and microscopically normal except for the size (12 x 12 x 3.5 cm) and weight (97 g). It showed normal thymic tissue segregated in lobules separated by adipose septa and with good corticomedullary separation. There were numerous Hassall corpuscles in the medullary portion. The thymus-to-adipose ratio was approximately 80:20. In

Localization of the gene for familial partial lipodystrophy (Dunnigan variety) to chromosome 1q21-22.

Obesity is strongly implicated in the pathophysiology of insulin resistance, diabetes mellitus and dyslipidemia. The mechanisms, however, by which obesity causes these complications are not known. The study of single-gene disorders affecting adipose tissue may elucidate some of the mechanisms involved in these processes. Familial partial lipodystrophy, Dunnigan variety, (FPLD, OMIM 308980) is an autosomal-dominant condition characterized by marked loss of subcutaneous adipose tissue affecting the trunk and extremities but with excess fat deposition in the head and neck areas. Affected individu…

Adolescent sex differences in adiponectin are conditional on pubertal development and adiposity.

OBJECTIVE: Adiponectin is an adipose tissue protein with important insulin-sensitizing, anti-inflammatory, and cardioprotective properties but is paradoxically lower in obese individuals. Sex differences in adiponectin have been reported in adults and adolescents but not in prepubertal children. In this study, we hypothesized that sex differences in adiponectin would develop during puberty and would be influenced by level of adiposity. RESEARCH METHODS AND PROCEDURES: Adiponectin levels were measured in 1196 white and African-American adolescents. Insulin resistance was estimated using the ho…

Adipokines: therapeutic targets for metabolic syndrome.

For a long time it has been known that obesity (adiposity) is linked to insulin resistance. Recently, many investigators have reported that adipocytes secrete a variety of bioactive molecules, termed adipokines (adipocytokines), including TNFalpha, IL-6, leptin, adiponectin, resistin and so on. These adipokines play pivotal roles in energy homeostasis by affecting insulin sensitivity, glucose and lipid metabolisms, food intake, the coagulation system and inflammation. This review provides a summary of these adipose tissue-secreting biomolecules and discusses their feasibilities as drug targets…

From body mass index to body composition in female infertility.

Obesity - a global epidemic that the World Obesity Federation expects will affect one in five women by 2030 - is an important factor influencing reproductive health. Its impact extends beyond subfertility, encompassing menstrual irregularities, oligo-anovulation, early pregnancy loss, and complications during pregnancy and delivery. The interplay between excess adiposity, metabolic health and reproductive function is complex, as adipose tissue secretes several adipokines, such as adiponectin and leptin, targeting reproductive and endocrine tissues. However, investigating the role of obesity on…

Adiponectin: novelties in metabolism and hormonal regulation.

Adiponectin is a protein secreted exclusively by white adipose tissues and is abundantly present in human plasma. Adiponectin was found decreased in obese and diabetes mellitus type 2 patients and increased with weight reduction. A negative correlation between circulating adiponectin levels and body mass index and insulin resistance has been demonstrated. Plasma adiponectin concentrations were found lower in diabetes mellitus type 2 patients with coronary artery disease. Moreover, studies in aortic endothelial cells revealed that the protein exerts a dose-dependent decrease of the surface expr…
