Risk factors for Overweight/Obesity among people living with HIV on antiretroviral therapy: An ambidirectional cohort study at a tertiary health facility in Zambia.

BACKGROUND: Overweight and obesity are major concerns among people living with HIV (PLWH), particularly those on integrase inhibitors, as they elevate the risk of cardiovascular diseases. However, longitudinal data on the burden and risk factors for overweight/obesity in sub-Saharan Africa (SSA) remain limited. This study aimed to estimate the incidence and identify factors associated with overweight and obesity among PLWH who switched to a dolutegravir (DTG)-based ART regimen at Livingstone University Teaching Hospital. METHODS: We enrolled 249 adults aged ≥18 years living with HIV on ART [n

KT-253, a Novel MDM2 Degrader and p53 Stabilizer, Has Superior Potency and Efficacy than MDM2 Small-Molecule Inhibitors.

Murine double minute 2 (MDM2) is an E3 ligase that inhibits the tumor suppressor protein p53. Clinical trials employing small-molecule MDM2/p53 interaction inhibitors have demonstrated limited activity, underscoring an unmet need for a better approach to target MDM2. KT-253 is a highly potent and selective heterobifunctional degrader that overcomes the MDM2 feedback loop seen with small-molecule MDM2/p53 interaction inhibitors and induces apoptosis in a range of hematologic and solid tumor lines. A single intravenous dose of KT-253 triggered rapid apoptosis and sustained tumor regression in p5

Mcl-1 is an important target protein for kaempferol from persimmon leaves in sensitizing ABT-199 to induce apoptosis in hepatoma cancer cells.

Overexpression of Mcl-1 causes hepatocellular carcinoma resistance to Bcl-2 inhibitors, but there are currently no direct Mcl-1 inhibitors available for clinical application. Our previous research demonstrated that kaempferol from persimmon leaves (KPL) can sensitize ABT-199 to inhibit liver cancer cell proliferation. This study further explored the effect of KPL sensitizing ABT-199 on liver cancer cell apoptosis and its potential mechanisms. The inhibitory effects of KPL and ABT-199, both individually and in combination, on the proliferation of HepG2, Huh7, and HCCLM3 cells were evaluated. Ce

A Population Pharmacokinetics Study of Venetoclax Concomitant with Voriconazole in Patients with Hematologic Malignancies.

BACKGROUND: Venetoclax is a selective small-molecule BCL-2 inhibitor that has been approved for treating hematologic malignancies. Co-administration with CYP3A inhibitors, such as voriconazole, poses a high risk of drug-drug interactions (DDIs) that can increase venetoclax exposure. This study aimed to develop a population pharmacokinetics (PopPK) model to characterize the PK properties of venetoclax when co-administered with voriconazole. METHODS: Patients (≥18 years of age) treated with venetoclax for hematologic malignancies and concomitant with voriconazole were enrolled. A PopPK model of

Rethinking the feasibility and safety of venetoclaxobinutuzumab in chronic lymphocytic leukemia: nontraditional factors may play a role in clinical practice.

The concept of fitness to receive treatment with novel agents in chronic lymphocytic leukemia (CLL) remains debated. Comorbidities and treatment-related logistics are increasingly recognized as key factors in treatment feasibility. Venetoclax- obinutuzumab (VO) has demonstrated efficacy in both fit and unfit patients in clinical trials, yet real-world data remain limited. This retrospective, multicenter study analyzed disease- and patient-related factors affecting VO management and outcomes in 271 patients. Fitness was assessed using comorbidity indices (CLL-CI, CIRS, CCI), Eastern Cooperative

Venetoclax promotes acute myeloid leukemia cells apoptosis by decreasing ac4C modification and mRNA expression of IMP3 through downregulating NAT10.

Venetoclax, a common treatment for acute myeloid leukemia (AML), exhibits unclear mechanisms. This study investigates whether venetoclax regulates AML apoptosis via NAT10, which is upregulated in AML and linked to poor prognosis. HL60 and KG-1 cell lines were treated with varying venetoclax concentrations, and cell viability, apoptosis, and protein levels of apoptosis-related markers were assessed using CCK-8, flow cytometry, and Western blotting. Molecular docking confirmed the interaction between venetoclax and NAT10. Venetoclax inhibited cell viability and promoted apoptosis in a dose-depen

Epidemiologic Characteristics Determining the Choice of Direct-Acting Antiviral Therapy in HCV Patients: An Italian Real-World Evidence Study.

Pangenotypic direct-acting antivirals (pDAAs) have transformed hepatitis C virus (HCV) treatment. In Italy, sofosbuvir/velpatasvir (SOF/VEL) and glecaprevir/pibrentasvir (GLE/PIB) are available. While both show similar efficacy, differences in patient profiles and potential drug-drug interactions (DDIs) may influence treatment choice. This study examined factors affecting pDAA selection and potential prescribing gaps. Using administrative databases (2018-2023) covering 3.7 million citizens, HCV patients were divided into SOF/VEL and GLE/PIB cohorts and compared by demographic, clinical, and th

HIV Dolutegravir resistance and multiclass failure in Mozambique: findings from a real-world cohort.

BACKGROUND: Dolutegravir (DTG) is the anchor drug of the first-line agent for HIV treatment globally, including low- and middle-income countries. Although clinical trials report low rates of integrase inhibitor resistance, real-world data from sub-Saharan Africa suggest a different scenario. We aimed to assess the prevalence and patterns of DTG resistance in Mozambique, building on prior preliminary findings with an expanded cohort and extended observation period. METHODS: We conducted a retrospective observational study in five DREAM centers in Mozambique. HIV-positive individuals on DTG-bas
