Predicting in vivo cannabinoid-drug interactions mediated via inhibition of UDP-glucuronosyltransferases using in vitro studies and physiologically based pharmacokinetic modeling and simulations.

We previously reported the inhibitory potential of the most extensively studied cannabinoids, cannabidiol (CBD) and Δ9-tetrahydrocannabinol (THC), along with their respective circulating metabolites, against a panel of cytochrome P450 (CYP) enzymes. In the current work, we characterized the inhibitory potential of these cannabinoids against a panel of recombinant human UDP-glucuronosyltransferases (UGTs). Upon screening CBD, THC, and their primary circulating metabolites (7-hydroxy CBD or 11-hydroxy THC) at 10 μM, cannabinoids likely to inhibit UGTs at pharmacologically relevant concentrations

Impact of Gut Microbiota and Probiotics on Rheumatoid Arthritis: A Potential Treatment Challenge.

Over the past few decades, there has been a surge in global study on the relationship between gut microbiota and human health. Numerous human illnesses have been linked to dysbiosis. Gram-positive firmicutes and Gram-negative bacteroidetes are the two leading bacterial phyla that make up 90% of the gut microbiome. Many symbionts in the gut environment establish intricate relationships with host defense to stop both local and non-native dangerous bacteria from colonizing and invading. Dysbiosis alters the paracellular route and damages the epithelium, enabling them to penetrate the epithelium a

Surgical revision in the presence of an S. aureus infection increases virulence factor expression and activates a multi-tissue inflammatory response.

Replacing implanted medical hardware due to infection often requires one or more revision surgeries. Each surgery triggers a tissue injury response and disrupts the established bacterial biofilm. However, the complex tissue response to reinjury and biofilm disturbance is not well understood. Our results show that with an existing infection, immunological niches such as the bone marrow, lymph nodes, and circulating blood further upregulate pro-inflammatory programs in response to revision. Rather than reducing bacterial burden, this heightened inflammation provokes virulence factor expression a

Innate Immune Response to Helminth Infections.

Helminths are the most common infectious parasites of humans, causing a global burden of disease, exceeding malaria and tuberculosis in developing countries. Helminth parasites have coevolved with humans and thus are quite well adapted to survive and flourish in them. The immune response induced by the parasite is predominantly pro-inflammatory, a Th1-mediated response involving antigen-presenting cells with elevated IgE and eosinophilia. At the later stage, the parasite is capable of polarizing the Th1 response to the Th2 response, which becomes favorable for their survival for a longer perio

New insights into the life cycle, host cell tropism, and infection amplification of Leishmania spp.

The genus Leishmania comprises several species of digenetic protozoan parasites that cause a spectrum of diseases known as leishmaniases, which are transmitted to humans and other mammals through the bite of hematophagous female sand flies. Leishmania spp. and their invertebrate vectors are widely distributed across the globe, putting more than a billion people at risk. Once inside mammalian hosts, these intracellular parasites reside within parasitophorous vacuoles of host cells. Although macrophages are the primary infected cells in lesions, Leishmania can also infect other cell types, whose

The Pain Is in the Brain With the Sugar-Coated Killer.

Cryptococcus neoformans and Cryptococcus gattii are encapsulated yeasts that have become increasingly common central nervous system (CNS) pathogens within an enlarging populationof immunocompromised host. CNS disease represents a major interaction between the host and the yeast. In this regard, we have examined the myriads of tools that this group of yeasts possess that allow them to become very efficient pathogens in the human CNS, causing fatal meningoencephalitis if untreated. In the age of molecular approaches to the investiagion of fungal pathogenesis, cryptococcosis has been studied exte

The Role of Myeloid-Derived Suppressor Cell (MDSC) in Fungal Infections and Its Potential as a Therapeutic Target.

BACKGROUND: The interaction between the immune system and fungi has long intrigued researchers, as fungi are among the most common organisms encountered by humans. Myeloid-derived suppressor cells (MDSCs) are specialized immune cells that play a key role in modulating immune responses during fungal infections by inhibiting T-cell activity. OBJECTIVES: This review aims to investigate the mechanisms by which MDSCs influence fungal infections and to explore their potential as therapeutic targets. METHODS: A comprehensive review of existing studies examining MDSC activity in various fungal infec

Transcriptomic profile of the zoonotic parasite Anisakis pegreffii upon in vitro exposure to human dendritic cells.

Anisakis pegreffii is a zoonotic marine nematode whose life-cycle involves marine organisms-small crustaceans, fish and squids as intermediate/paratenic hosts, and marine mammals, mainly cetaceans-as definitive ones. When its third-stage larvae (L3) are accidentally ingested by humans with the consumption of raw or undercooked parasitized fish and/or squids, the parasite fails to complete its life cycle, leading to human anisakiasis. Despite increasing interest in understanding the molecular basis of pathogenic effects in human anisakiasis, the transcriptomic response of A. pegreffii L3 to int

Toxoplasma effector TgROP1 establishes membrane contact sites with the endoplasmic reticulum during infection.

Membrane contact sites (MCS) are essential for organelle communication in eukaryotic cells. Pathogens also establish MCS with host organelles, but the mechanisms underlying these interactions and their role in infection remain poorly understood. Here, using a fluorescence sensor and CRISPR-based loss-of-function screening, together with imaging and proteomics, we identify the parasite effector mediating MCS between host endoplasmic reticulum (ER) and the vacuole containing the intracellular parasite Toxoplasma gondii. TgROP1 acts as a tether and mimics a canonical FFAT motif to bind the host E

Lipid transport proteins in Toxocara canis: Host lipid acquisition and immune modulation.

Toxocara canis is unable to synthesize sufficient lipids de novo to meet its biological requirements and therefore depends on host-derived lipids for survival. The parasite expresses a diverse set of lipid transport proteins spanning all major classes, such as pseudocoelomic fluid lipoproteins (vitellogenins), nematode polyprotein antigens/allergens, intracellular carriers (fatty-acid binding proteins, phosphatidylinositol-transfer proteins), secreted lipid-binding proteins (fatty acid-and retinol-binding proteins, venom allergen-like proteins), membrane-associated transporters (Niemann-Pick C

The endothelium at the interface between tissues and Staphylococcus aureus in the bloodstream.

SUMMARYStaphylococcus aureus is a major human pathogen. It can cause many types of infections, in particular bacteremia, which frequently leads to infective endocarditis, osteomyelitis, sepsis, and other debilitating diseases. The development of secondary infections is based on the bacterium's ability to associate with endothelial cells lining blood vessels. The success of endothelial colonization and infection by S. aureus relies on its ability to express a wide array of cell wall-anchored and secreted virulence factors. Establishment of endothelial infection by the pathogen is a multistep pr

The final gate: bacterial subversion of autophagosome-lysosome fusion as a conserved immune evasion strategy.

Autophagy is a vital component of the host cell intracellular defense arsenal, culminating in the fusion of autophagosomes with lysosomes to degrade invading pathogens. While autophagosome formation has been extensively studied, recent insights reveal that the final fusion step constitutes a critical immunological bottleneck that is highly vulnerable to microbial sabotage. In this review, we synthesize evidence from diverse pathogens, including Mycobacterium tuberculosis, Salmonella enterica, Treponema pallidum, Helicobacter pylori, Coxiella burnetii, Yersinia pestis, and Porphyromonas gingiva

From past to present: the crosstalk between type VI secretion system of Salmonella and host.

Salmonella encounters dual selective pressures during infection - from the host immune system and competing gut microbiota. The Type VI secretion system (T6SS) is a versatile transmembrane nanomachine that delivers effector proteins into target cells or the extracellular environment, enabling diverse functions including bacterial competition, virulence, and niche adaptation. Emerging evidence demonstrates that some T6SS effectors exhibit dual targeting capability, affecting both eukaryotic and prokaryotic cells. This dual-targeting strategy confers competitive advantages by simultaneously elim

Host cell-specific metabolism of linoleic acid controls Toxoplasma gondii growth in cell culture.

The obligate intracellular parasite Toxoplasma gondii can infect and replicate in any warm-blooded cell tested to date, but much of our knowledge about T. gondii cell biology comes from just one host cell type: human foreskin fibroblasts (HFFs). To expand our knowledge of host-parasite lipid interactions, we studied T. gondii in intestinal epithelial cells, the first site of host-parasite contact following oral infection and the exclusive site of parasite sexual development in feline hosts. We found that highly metabolic Caco-2 cells are permissive to T. gondii growth even when treated with hi

Eosinophils as modulators of host defense during parasitic, fungal, bacterial, and viral infections.

Eosinophils, traditionally associated as central innate effector cells with type 2 immunity during allergic and helminth parasitic diseases, have recently been revealed to have important roles in tissue homeostasis as well as host defense in a broader variety of infectious diseases. In a dedicated session at the 2023 biennial conference of the International Eosinophil Society titled "Eosinophils in Host Defense," the multifaceted roles eosinophils play against diverse pathogens, ranging from parasites to fungi, bacteria, and viruses, were presented. In this review, the session speakers offer a

Hofmeister effect enhanced SiO2/gelatin-based hydrophobically associated hydrogels and their lubricating properties.

In recent years, hydrogel materials with suitable energy dissipation mechanisms and excellent mechanical properties have attracted much attention in tissue engineering due to their ability to mimic the natural cartilage structure. However, in cartilage tissue's regeneration and repair process, hydrogel materials should also possess satisfactory lubrication properties and biocompatibility. Therefore, preparing biocompatible low friction, high toughness hydrogels remain a challenge. In this paper, a new strategy is proposed to use gelatin, acrylamide (AM), lauryl methacrylate (LMA) and SiO2 to c
