Aseptic arthritis in a man with toxic shock syndrome.

Synovitis in toxic shock syndrome (TSS) is an unusual finding. A 31-year-old man presented with pain and swelling in both knees and was found to have TSS, secondary to a septic bursitis caused by Staphylococcus aureus. Immune complexes were not detected in serum or synovial fluid, and the S aureus was not recovered from the inflamed joints. Antibodies against the TSS toxin-1 were detected in serum and synovial fluid, but in lower levels than would be seen in a normal control serum. Complement studies implicated alternative pathway activation by a marked diminution in C3 levels compared with C4

Evidence against HLA and immunological dependence of disease outbreak in SLE. Immunological characterisation of identical twins clinically discordant for SLE.

Identical female twins clinically discordant for 20 years for SLE were studied. Their HLA-haplotype was A1,28; B8w6,w35; Cw3,w7; Dr3,4. Both twins had a raised erythrocyte sedimentation rate, autoantibodies, and circulating immune complexes. The diseases sibling had a reversed OKT4/OKT8 ratio (0.43), decreased helper T cell number, defective pokeweed mitogen (PWM) induced plasma cell differentiation, and overactive hydrocortisone sensitive suppressor cells. Immunological abnormalities may be only partly HLA related (B8; Dr3), but are most probably secondary to the disease process in the siblin

Anergy in tuberculosis.

Anergy in tuberculosis is of considerable clinical and immunological interest. Although negative skin reactions may be secondary to improper tuberculin testing or to certain diseases, drugs, vaccinations and constitutional factors known to affect cell-mediated immunity, often no underlying explanation is apparent. Unexplained negative reactions occur with a highly variable frequency in a wide variety of tuberculous infections. Anergy in tuberculosis is usually generalized, is often accompanied by other immunologic abnormalities, and frequently disappears as the disease is treated. Most anergic

Hymenolepis citelli (Cestoda) and Nematospiroides dubius (Nematoda): interspecific interaction in mice.

In mice concurrently infected with Hymenolepis citelli and Nematospiroides dubius, survival of the tapeworm was prolonged, and there was an impairment of the efferent arm of the response to the cestode. The immunological rejection of a six cysticercoid primary H. citelli infection was delayed by the N. dubius infection. The growth of the cestode was poorer in concurrently infected mice, and this effect was rapid, being evident within 4 days of the N. dubius infection. Maximum biomass in the controls was reached on Day 20, whereas in the concurrently infected mice it was reached on Day 25. The

[Circulating immune complexes in uveitis].

The serum of 63 patients suffering from various clinical types of endogenous uveitis with unknown etiology was investigated for the presence of circulating immune complexes (CIC). None of the patients was suffering from a systemic disease usually associated with CIC. Significantly elevated CIC levels were detected in 2 women with chronic cyclitis. One of them was also suffering from paresis of the facial nerve. The investigation reported here indicates that C1q-binding CIC are of no importance in the pathogenesis of endogenous uveitis. On the one hand one should consider the possibility of loc

Histological and immunological study in patients with rheumatoid arthritis showing isolated abnormalities of salivary scintigraphy.

We report the histological and immunological findings in 20 rheumatoid arthritis (RA) patients with impaired salivary scintigraphy but without sicca complex. This condition was stable over 2-5 years and associated with mild infiltrates on lip biopsies. Increased levels of IgG, immune complexes, and antinuclear antibodies were found in seven patients (35 vs 69% in secondary Sjögren's syndrome). Two of the seven patients later developed xerostomia. The above-mentioned immunological abnormalities may increase the likelihood of developing a clinical impairment of salivary function in patients with

[Immunological and morphological comparisons in toxic goiter].

Histostructural peculiarities of the toxic goiter tissue were compared with humoral parameters of the autoimmune aggression and with the lymphoblasttransformation intensity in 29 patients operated on for thyrotoxicosis. The intensity of intradermal hypersensitivity reaction of delayed type in response to homologous and autologous tissue of the toxic goiter was also investigated. The majority of the patients displayed marked signs of autoimmune aggression, regardless of the structure of the goiter parenchyma and the intensity of ilt lymphoid infiltration. Immunological disturbances increase wit

Action of mannitol in various immunological experimental models.

The authors describe the inhibiting action of mannitol after repeated administration of low subcutaneous doses in a number of experimental immunological models. For example, in the rat it produces a reduction of the secondary arthritis of Freund's adjuvant polyarthritis and also of the pleurisy due to Bordetella pertussis hypersensitivity. In the mouse it reduces the reaction of delayed hypersensitivity to sheep red cells. Its action is also marked against ovalbumin-induced active skin anaphylaxis in the albino guinea-pig and on IgE synthesis in the rat. Moreover, after several injections it p

The nasal bony dorsum and cartilaginous vault. Pitfalls in management.

Our literature is replete with technical descriptions of rhinoplasty techniques designed to avoid certain complications. Analysis of 153 secondary rhinoplasties indicates that no procedure is immune to undesirable results and no specific technique can be employed to manage all potential problems. Critical scrutiny of these cases both preoperatively and intraoperatively indicated a decided recurrence of certain complications in the nasal bony dorsum and cartilaginous vault. Fifty-eight defects of the bony dorsum were encountered. These defects included open vault, wide dorsum, asymmetry, excess

Characteristics of Secondary Malignancy Among Children With Primary Immunodeficiency Disorders in Saudi Arabia.

Background Children with primary immunodeficiency disorders (PIDDs) are at an increased risk of developing malignancies due to impaired immune surveillance and genomic instability. In Saudi Arabia, limited data exist regarding the frequency, characteristics, and outcomes of secondary malignancies in this vulnerable population. Therefore, this study aimed to evaluate the clinical features, types of PIDD, associated secondary malignancies, treatment modalities, and outcomes among pediatric patients with PIDD treated at a tertiary care center in Saudi Arabia. Methods This retrospective observatio

Chemokine CXCL10 and Incident Stroke Risk in the Northern Manhattan Study.

Inflammation contributes to cerebrovascular disease, but prospective studies of broad immune marker panels and incident stroke are limited. We aimed to identify plasma immune markers independently associated with incident stroke. Stroke-free Northern Manhattan Study participants aged ≥ 50 years had 60 plasma immune-related proteins measured by Luminex. Least absolute shrinkage and selection operator (LASSO) Cox regression selected markers associated with incident all-cause stroke. Selected markers were tested in Cox models of stroke risk sequentially adjusted for vascular risk, cardiac comorbi

Can a mosquito-borne virus become a cancer therapy? rethinking Getah virus through the lens of oncolytic virotherapy.

Getah virus (GetV) is an arthropod-borne alphavirus historically recognized as an emerging zoonotic pathogen of veterinary significance, particularly in livestock and equine populations across Asia and parts of the Western Pacific. Over the past several decades, its expanding ecological range, broad mosquito vector competence, and increasing frequency of animal outbreaks have positioned GetV as a growing concern for animal health surveillance, diagnostics, and vaccine development. However, beyond its established role in veterinary virology, a critical and underexplored dimension of GetV biolog

Early Immune Phenotype-Guided Immunomodulatory Strategies and Intensive Care Outcomes in Septic Shock: A Single-Center Retrospective Cohort Study.

BACKGROUND: Septic shock is characterized by profound immune dysregulation, with marked heterogeneity in host inflammatory and immunosuppressive responses. This biological variability may contribute to the inconsistent effects observed in prior immunomodulatory interventions. Whether early immune phenotype-guided immunomodulation is associated with differential patterns of shock resolution and organ recovery remains incompletely understood. METHODS: We conducted a single-center retrospective observational cohort study of adult patients with septic shock. Immune phenotyping using routinely ava

A Paradigm Shift: Arrhythmogenic Cardiomyopathy Is an Inflammatory Disease.

Arrhythmogenic cardiomyopathy (ACM) is a genetic myocardial disorder marked by progressive cardiomyocyte loss, fibro-fatty replacement, ventricular arrhythmias, and risk of sudden cardiac death. Traditionally considered a structural and electrical disease driven by desmosomal dysfunction, emerging evidence redefines ACM as an inflammatory cardiomyopathy in which immune activation plays a central role. This review integrates genetic, molecular, experimental, and clinical data to highlight inflammation as a unifying feature of ACM. Desmosomal gene variants impair cell adhesion and also activate

Circulating microbial metabolites and the gut-prostate axis in prostate cancer: Implications for laboratory biomarkers and therapeutic response.

Prostate cancer progression and treatment response are influenced not only by tumor genomics and androgen receptor signaling but also by systemic host-microbiome interactions along the gut-prostate axis. Increasing evidence indicates that gut microbial metabolism produces bioactive compounds that circulate in human body fluids and can influence immune regulation, hormone metabolism, and therapeutic outcomes. This review synthesizes current evidence on microbiome-derived metabolites that may serve as measurable biomarkers relevant to prostate cancer biology and clinical laboratory diagnostics.

Triptolide-Induced Male Germ Cell Damage Leads to Non-Infectious Epididymitis in Mice.

BACKGROUND: Triptolide is a traditional Chinese medicine used to treat autoimmune and inflammatory diseases. However, one adverse side effect of triptolide is male infertility. While triptolide's testicular toxicity is well-documented, its impact on epididymal function, particularly its role in triggering immune dysregulation (e.g., epididymitis secondary to germ cell damage), remains unclear. OBJECTIVES: The objective is to explore the damage to male germ cells in mice after triptolide administration and analyze how the male germ cells' damage leads to epididymitis. MATERIALS AND METHODS: T

USP4 controls neutrophil spreading through stabilising IQGAP1 during lung inflammation.

BACKGROUND: Protein availability critically influences innate immune reactivity and the extent of immune-mediated tissue injury. A key unresolved question is how specific protein homeostasis networks within immune cells such as neutrophils can be modulated to mitigate the detrimental outcomes of acute inflammation. METHODS: We established an acute lung injury (ALI) model in immune-specific ubiquitin-specific peptidase 4 (USP4) knockout chimeric (BM-Usp4-/-) and control (BM-WT) mice. Immune (dys)function in obsessive-compulsive disorder.

BACKGROUND: It has been suggested that peripheral inflammation and immune-mediated neuronal dysfunction play a decisive role in the pathophysiology of obsessive-compulsive disorder (OCD). However, work addressing this hypothesis is mostly based on small samples and heterogeneous methodologies, and has led to inconsistent results. Here we conducted the largest study to date comparing an extensive panel of immune markers in the peripheral blood between adult patients with OCD and healthy controls. METHODS: One-hundred and thirty-nine patients with OCD and 131 age and sex-matched controls were a

Epigenetic profiling reveals M1 macrophages as the primary drivers of trained immunity in HSV-1 latently infected mice.

Macrophages play multifaceted and critical roles in controlling diverse pathogenic infections. Building on our recently published observations of macrophages exhibiting memory responses to HSV-1 infection, our current report investigates the macrophage subtype responsible for generating trained immunity against virus-induced immunopathogenesis. Using ATAC-seq (Assay for Transposase-Accessible Chromatin using sequencing), an epigenetic profiling technique, we identified chromatin accessibility changes in both M1 and M2 macrophages associated with the acquisition of IRGM1, a marker of the traine

Immunoinformatics-guided design of a universal chimeric multi-epitope subunit vaccine against Marburg virus disease and Ravn virus co-infection.

Marburg Virus Disease (MVD) is a life-threatening hemorrhagic fever, caused by two viruses-Marburg virus (MARV) and Ravn virus (RAVV)-that belong to the family Filoviridae. Due to its high mortality rate and the lack of licensed vaccines, MVD remains a major global health concern. In this study, a comprehensive immunoinformatics workflow was used to design a universal chimeric multi-epitope subunit vaccine targeting conserved antigenic regions of both viruses to reduce MVD-related mortality. Antigenicity analysis identified glycoproteins (GP) of MARV and RAVV as the most immunogenic viral prot
