Molecular characterisation of catalase in Antarctic teleosts: insights into oxidative stress management in an extreme environment.

The permanently sub-zero temperatures and high oxygen solubility of the Southern Ocean promote the formation of reactive oxygen species (ROS) in the tissues of resident organisms, imposing persistent oxidative stress. Antarctic notothenioid fish have evolved efficient antioxidant systems to counter this pressure. This study reports the first molecular characterisation of the catalase (cat) transcript and deduced protein (CAT) sequence, together with tissue-specific CAT activity, in two Antarctic species with contrasting physiological traits: the red-blooded notothenioid Trematomus bernacchii a

Evaluating the Whole Blood Survival Advantage: How Citrate and Calcium Levels Drive Mortality in a Translational Swine Model of Polytrauma.

OBJECTIVES: Whole blood (WB) is increasingly used for trauma resuscitation, but clinical comparisons with component therapy (COMP) have reported mixed survival results, and physiologic contributors to any differences remain incompletely defined. We evaluated the association between excess citrate exposure, calcium (Ca2+) supplementation, hemodynamic stability, and survival during massive transfusion. METHODS: In a randomized swine (Sus scrofa) polytrauma model, 32 castrated male and nonpregnant females underwent tibial fracture, blunt liver injury, 30% controlled hemorrhage, and a 30-minute s

The efficacy of transdermal melatonin in dental anxiety in children: a randomised placebo-controlled clinical trial.

PURPOSE: Dental anxiety, a prevalent barrier to optimum dental care, frequently necessitates pharmacological intervention. Melatonin appears to be a safe anxiolytic in medical settings but remains underexplored in paediatric dental anxiety management. Our objective was to evaluate the efficacy of transdermal melatonin patches in managing dental anxiety in children undergoing invasive dental treatment. METHODS: 52 children aged 5-7 years undergoing dental treatment requiring infiltration local anaesthesia (LA) were randomly assigned to receive either 2 mg transdermal melatonin patches (n = 26)

Differential metabolomic effects of progestagen-based estrus synchronisation in South African Dohne Merino ewes.

This study represents one of the first to comprehensively profile the homeostatic cervicovaginal metabolome of South African Dohne Merino ewes using liquid chromatography high resolution mass spectrophotometry (LC-HRMS) and compare that metabolome immediately after estrus synchronisation using progestagen intravaginal pessaries. As the use of assisted reproductive technologies gains traction in various livestock production systems, characterising their effects using novel omics approaches is crucial to physiology, welfare and production. An ewe has well-developed metabolomic cascades that are

Targeting the protein tyrosine phosphatase 1B signaling axis in Alzheimer's disease: Emerging therapeutic potential of ivermectin.

This study attempts to repurpose ivermectin (IVM) for AD by inhibiting the PTP1B enzyme. IVM is a widely known, commercially approved antiparasitic drug. The PTP1B enzyme dephosphorylates multiple kinase substrates and is responsible for metabolic regulation by the PI3K/AKT downstream signaling cascade. A similar signaling pathway is present in the brain. We assessed binding affinity in BIOVIA Discovery Studio Visualizer version 2021 and inhibitory activity using a commercially available ELISA kit. The streptozotocin (3 mg/kg, intracerebroventricularly)- induced AD mouse model was used to stud

Blunted niacin skin flushing response with subtype-specific clinical associations in adolescent bipolar disorder.

BACKGROUND: Bipolar disorder (BD) typically emerges during adolescence and is associated with substantial functional impairment, but objective physiological markers are scarce. The niacin skin flushing response (NSFR) has been proposed as a potential biomarker in adults with BD. However, its characteristics in adolescents and its multivariate associations with behavioral phenotypes, particularly across BD subtypes, remain poorly characterized. METHODS: We recruited 402 adolescents (184 with BD [63 BD-I, 121 BD-II]; 218 healthy controls) and conducted quantitative niacin testing. Participants u

Intermittent exogenous ketosis does not reduce acute mountain sickness during early terrestrial high-altitude acclimatization.

PURPOSE: Intermittent exogenous ketosis via ketone monoester (KE) ingestion has been shown to enhance physiological responses and reduce acute mountain sickness (AMS) symptoms during acute normobaric hypoxia. Its effects during early acclimatization to terrestrial high-altitude, however, remain unclear. METHODS: Thirty-four participants were randomised to an intermittent exogenous ketosis (IEK, n=17) or placebo (PLA, n=17) group and underwent a near sea level trial (295 m) without supplementation, followed by a four-day sojourn at 3375 m during which they received regular KE (IEK) or placebo

Short-Term Psychological and Physiological Changes Following Coffee Aroma Exposure: A Multimodal Assessment.

Background/Objectives: Coffee aroma has been associated with psychological and physiological responses, although these responses have often been examined separately. This exploratory study used a multimodal approach to assess subjective mood, electroencephalographic (EEG) activity, heart rate (HR), and heart rate variability (HRV) following short-term coffee aroma exposure. Methods: Twenty healthy adults aged 20-29 years participated in a single-group, within-subject pre-post study and were exposed to coffee aroma for 5 min. Mood was assessed using the Profile of Mood States (POMS). Pre- and p

Haptoglobin Protects Against Increased Chlorine Inhalation Vulnerability in Sickle Cell Trait Mice.

RATIONALE: Sickle cell trait (SCT), carried by millions worldwide, is generally considered benign. However, SCT is increasingly linked to life‑threatening complications under environmental stress. OBJECTIVES: We tested whether sickle cell trait confers heightened vulnerability to oxidant inhalation injury and whether post-exposure haptoglobin mitigates injury. METHODS: Using humanized SCT (AS) and control hemoglobin (AA) mice carrying human hemoglobin knock‑in genes, we examined physiologic, biochemical, histologic, mitochondrial, and proteomic responses to chlorine (Cl₂) inhalation at conce

Antibodies elicited by a chimeric peritrophin antigen reduce Aedes aegypti survival after blood feeding.

Vector-borne diseases transmitted by Aedes aegypti, including dengue, Zika, chikungunya, and yellow fever, represent a persistent and expanding global health threat. Limitations of conventional vector control strategies, such as insecticide use and environmental source reduction, particularly in urban settings, underscore the need for novel and complementary tools. Anti-vector vaccination offers one such strategy by inducing host-derived antibodies that are ingested during vector feeding and interfere with vector physiology or survival. Although this concept has been successfully explored in s

Non-uniform growth probability of Mycobacterium tuberculosis across inoculum densities partially restored by culture filtrate: Implications for supplemented growth assays.

Differentially culturable (DC) Mycobacterium tuberculosis (Mtb) phenotypes reduce the sensitivity of sputum culture and may be associated with adverse treatment outcomes in tuberculosis patients. Detecting and quantifying DC Mtb in clinical samples remains an important research objective; current approaches rely on culture filtrate (CF) supplemented growth assays that assume CF selectively resuscitates DC Mtb rather than broadly stimulating growth from low density inocula - an untested assumption for Mtb. We performed a half-logarithmic dilution series of Mtb from approximately 10,000-0 total

The sRNA-encoded peptide TimP rewires the Escherichia coli envelope via LamB to sensitize bacteria to polymyxins.

The rise of multidrug-resistant (MDR) Escherichia coli has marginalized last-line polymyxins (PMs) owing to evolving resistance and narrow therapeutic windows. Revitalizing these agents requires the identification of novel regulatory circuits governing the integrity of the gram-negative envelope. In this study, we identified a stress-responsive peptide TimP and demonstrated that its intracellular overexpression (TimP+), a synthetic variant (FT-Pep), and biogenic TimP+-derived extracellular vesicles (TimP EVs) exhibited equivalent functionality in sensitizing E. coli to PMs, consistently induci

Comparative Responses of Invasive and Native Plant Species to Combined Cd and Microplastic Pollution.

The co-occurrence of heavy metal contamination and biodegradable microplastic (polylactic acid, PLA) pollution poses increasing risks to terrestrial plant communities and soil functioning, yet species-specific responses to combined stress remain poorly understood. Cd and microplastics frequently co-occur in agricultural soils, where microplastics can alter cadmium mobility, bioavailability, and transport pathways, potentially modifying metal toxicity and plant stress responses compared with single-pollutant exposure. We investigated the responses of the invasive Bidens pilosa and the native So

Overexpression of StBIN2 Improves Salt Stress Tolerance in Potato.

Salt stress has become one of the major abiotic stress factors limiting sustainable crop production worldwide, and potato, as the fourth largest food crop globally, is particularly severely affected by saline and other environmental stresses in terms of its growth, development, yield, and quality. StBIN2 belongs to the GSK3 family of proteins, and numerous studies have confirmed that GSK3 family members widely regulate diverse abiotic stress responses and developmental processes in plants. However, the specific function and underlying mechanism of StBIN2 in the salt stress response of potato r

The kinase LRRK2 is required for the physiological function and expression of the glial glutamate transporter EAAT2 (SLC1A2).

Neurotransmitter transporters (NTTs) control synaptic responses by modulating the concentration of neurotransmitters at the synaptic cleft. Glutamate is the most abundant excitatory neurotransmitter in the brain and needs to be finely tuned in time and space to maintain a healthy brain and precise neurotransmission. The glutamate transporter EAAT2 (SLC1A2) is primarily responsible for glutamate clearance. EAAT2 impairment has been associated with Alzheimer's disease (AD), Huntington's disease (HD), amyotrophic lateral sclerosis (ALS), and Parkinson's disease (PD). Mutations in leucine-rich rep

Self-disinfecting adhesive chitosan nanospray therapeutics for on-demand antibiotic prophylaxis against healthcare-associated infections.

Healthcare-associated infections (HAIs) present a critical clinical challenge that significantly increases patient morbidity and healthcare costs. While pH-responsive antibacterial nanoparticles (NPs) have emerged as promising therapeutic platforms, current strategies are hampered by poor tissue adhesion and premature drug release under physiological conditions. Here, we propose sprayable adhesive antibacterial NPs based on catechol-conjugated chitosan with dynamic Cu2+ coordination for infection-triggered vancomycin release in HAI prevention. This multifunctional therapeutic system uniquely i
