Evolving 10-year epidemiological landscape of amyotrophic lateral sclerosis (ALS) in Ceará, Brazil.

Objective: Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder that affects upper and lower motor neurons with variable cognitive impairment. Despite its rarity, ALS has a major social and economic impact and increasing incidence worldwide. There are few population-based studies of ALS in Brazil. Methods: We present a descriptive and quantitative study, with a retrospective and prospective design, of patients with ALS followed at the Walter Cantídio University Hospital (HUWC) from January 2013 to December 2023, with the aim of describing the clinical, epidemiological, and genet

Evaluation of the ivermectin effects on porcine granulosa cell function.

Ivermectin (IVM) is used as antiparasitic drug in veterinary medicine and it is also the only avermectin approved in humans in several parasitic infestations. We examined this substance because potential critical issues associated with the use were highlighted. Moreover, criticisms related to the ecological-environmental sphere, as well as food contamination arise. Nevertheless, current in vitro studies are lacking. This study aimed to investigate the direct impact of IVM on reproductive function using swine granulosa cells as reproductive cell model. The results show that progesterone, ATP an

Treating SOD1-ALS with tofersen results in nonprogressive chronic ALS-a case series from Iceland.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder. We describe four patients with hereditary ALS caused by the p.Gly94Ser SOD1 mutation who were treated monthly with the intrathecal antisense oligonucleotide tofersen in a clinical setting at Landspitali University Hospital of Iceland. After initiating treatment 15-26 months ago, no significant clinical deterioration was observed, and three patients showed signs of clinical improvement, with some recovery of motor function. All four patients currently present with chronic nonprogressive ALS, a phenotype not previou

Nose-to-brain delivery of a SOD1-stabilizing small molecule ameliorates pathology in an ALS mouse model.

Exposure of a pathogenic β6/β7 loop neo-epitope has been proposed to contribute to the pathogenesis of misfolded Cu/Zn superoxide dismutase (SOD1) in amyotrophic lateral sclerosis (ALS) by mediating early events in its noxious structural transformation and prion-like activity. Antibody-mediated blockade of this epitope was shown to ameliorate disease phenotype in an ALS animal model. Here, as an alternative strategy, we sought to block this epitope using a small molecule designed to occupy the inter-subunit cavity framed by the two β6/β7 loops. Using a structure-based virtual screen targeting

Coumarin application modulates antioxidant defense to enhance resistance against Fusarium in mung bean.

Plant-derived bioactive compounds contribute a major role in plant defense against fungal pathogens. Among these, coumarin proved to be a major controller in root-pathogen interactions, exhibiting diverse protective mechanisms. This study investigates the effects of coumarin (50 mg.L- 1), applied through seed priming and foliar spray, on the biochemical responses of Mung bean and its impact on root colonization by Fusarium solani. Mung bean seeds pre-treated with coumarin were sown under both infected and non-infected conditions. Once the seedling established up to two-leaf stage, weekly folia

Exogenous melatonin modulates growth, antioxidant defense, and secondary metabolism of Matthiola incana under drought stress.

Drought stress is a major environmental constraint that adversely affects plant growth, physiological performance, and the accumulation of bioactive compounds. The use of biostimulants such as melatonin (MT) has emerged as a promising strategy to enhance plant tolerance to abiotic stress. In this study, the effects of exogenous MT on growth, physiological traits, antioxidant defense, and secondary metabolism of Matthiola incana under different irrigation regimes (100, 70, 40, and 25% field capacity, FC) were investigated. The experiment was conducted as a factorial design with four MT concentr

Antioxidant Activity of Ethanolic Litchi chinensis Seed Extract in Oxidative Stress Model Mice and Identification of Blood-Entering Prototype Components.

Litchi chinensis seeds are rich in flavonoids and exhibit potent antioxidant activity. This study constructed a D-galactose-induced oxidative stress model in mice and applied ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS), network pharmacology, and molecular docking to clarify the antioxidant activity and material basis of ethanolic litchi seed extract. Litchi seed extract was orally given by gavage at 100 and 200 mg/kg in antioxidant tests, whereas a dosage of 500 mg/kg was adopted for the detection of absorbed constituents in plasma. The results showed that the tot

Paternal lead exposure induces transgenerational oxidative stress and down-regulation of antioxidant genes in Drosophila melanogaster.

Lead (Pb) is one of the most prevalent environmental toxicants and is of great concern due to its adverse effects. Despite lead's oxidative properties, studies on the transgenerational effects of paternal Pb exposure remain inadequate. This study aimed to evaluate these effects on oxidative stress and the expression of specific genes using the D. melanogaster model. Male fruit flies were fed a normal diet supplemented with lead acetate as the Pb source for 20 consecutive days. The Pb-exposed males were mated with unexposed females, producing F1 offspring. F1 flies were fed a normal diet, and t

Protective Effect of L-Arginine and Filgrastim Against Valproic Acid-Induced Histopathological Changes on the Gastric and Lingual Mucosa.

Valproic acid (VPA) is a commonly used anticonvulsant prescribed for various neurological disorders. Adverse effects may arise from chronic use and high dosages. This study aims to evaluate the protective effects of L-arginine and Filgrastim against the harmful impact of VPA on gastric and lingual mucosa. Forty male albino rats were divided into four groups (10 for each). The control group received saline, while the VPA group administered VPA (500 mg/kg) orally for 14 days. The VPA + L-arginine and VPA + Filgrastim groups received L-arginine (300 mg/kg) and Filgrastim (40 μg/kg), respectively,

Lung Vascular Remodeling and Oxidative Damage Induced by Chronic Intermittent Hypoxia.

High-altitude workers in the Los Andes Mountains, known as "the Chilean miner model," are exposed to chronic intermittent hypobaric hypoxia (CIHH). This intermittent condition differs from other models of chronic hypoxia, mainly due to the hypoxic pattern and the cardiovascular and pulmonary effects. There are reports of cardiopulmonary dysfunction and remodeling in human and animal models. However, research on some mechanisms of vascular function and the consequences of lung remodeling induced by CIHH is still lacking. Therefore, this study aims to characterize the effects of CIHH exposure on

COMMD1 Induces Copper Deficiency of SOD1 by Inhibiting the Palmitoylation of CCS in ALS.

Mutations in superoxide dismutase 1 (SOD1) compromise its metal-binding capacity, resulting in protein misfolding and aggregation, which ultimately induces cellular apoptosis in amyotrophic lateral sclerosis (ALS). Copper metabolism domain containing 1 (COMMD1), a gene implicated in copper homeostasis, has not been thoroughly characterized in the context of ALS pathogenesis. In this study, we identified elevated COMMD1 expression in ALS, potentially contributing to diminished copper incorporation into SOD1. Knockdown of COMMD1 enhanced palmitoylation of the copper chaperone for SOD1 (CCS), fac

Nitroxoline exerts potent anti-Aspergillus fumigatus activity by disrupting copper homeostasis and inducing oxidative stress.

Invasive pulmonary aspergillosis (IPA) caused by Aspergillus fumigatus remains a clinical challenge due to limited therapies and rising antifungal resistance. Here, we evaluated nitroxoline (NTX), an FDA-approved 8-hydroxyquinoline derivative, for its antifungal potential and mechanism of action. NTX exhibited potent in vitro activity against 64 clinical isolates and the AF293 reference strain, delaying conidial maturation and significantly improving survival, reducing pulmonary fungal burden, and alleviating lung inflammation and hyphal invasion in an IPA mouse model. Transcriptomic analysis

Network analysis of the cerebrospinal fluid proteome reveals shared and unique differences between sporadic and familial forms of amyotrophic lateral sclerosis.

BACKGROUND: Amyotrophic Lateral Sclerosis (ALS), a neurodegenerative disease involving loss of motor neurons, typically results in death within 3-5 years of disease onset. Although roughly 10% of cases can be linked to a specific inherited mutation (e.g., C9orf72 hexanucleotide repeat expansion or SOD1 mutation), the cause(s) of most cases are unknown. Consequently, there is a critical need for biomarkers that reflect disease onset and progression across ALS subgroups. METHODS: We employed tandem mass tag mass spectrometry (TMT-MS) based proteomics on cerebrospinal fluid (CSF) to identify and
