The Na(+)-dependent or E1 stages of the Na,K-ATPase reaction require a few micromolar ATP, but submillimolar concentrations are needed to accelerate the K(+)-dependent or E2 half of the cycle. Here we use Co(NH(3))(4)ATP as a tool to study ATP sites in Na,K-ATPase. The analogue inactivates the K(+) phosphatase activity (an E2 partial reaction) and the Na,K-ATPase activity in parallel, whereas ATP-[(3)H]ADP exchange (an E1 reaction) is affected less or not at all. A calcium-dependent protein kinase (PK-III), not requiring calmodulin for activity, was purified from extracts of ripening mango fruit tissue. PURPOSE: Chloride efflux plays an important role in aqueous humor production. Chloride currents have been described in bovine non-pigmented ciliary epithelium (NPE), in transformed cultured human NPE, and in bovine volume-activated chloride channels described in the latter. It is the basolateral membranes of the NPE of ocular ciliary processes that comprise the exit pathway for the process of aqueous secretion performed by the double syncytial layer of ciliary epithelium, however. In this report we describe that the functional capacity of the mitochondrial oxidative phosphorylation system can be studied in cultured skin fibroblasts permeabilized with a limited amount of digitonin. By using a variety of different oxidizable substrates, information can be obtained on the functional activity of complex I, for instance, which is important since different diseases in man have recently been identified in which complex I is deficient. The method described may contribute to the biochemical characterization of patients suffering from one of a variety of encephalomyopathies.
