Changes in cartilage structure and composition are commonly observed during the progression of osteoarthritis (OA). Importantly, quantitative magnetic resonance imaging (MRI) methods, such as T1rho relaxation imaging, can noninvasively provide in vivo metrics that reflect changes in cartilage composition and therefore have the potential for use in early OA detection. Changes in cartilage mechanical properties are also hallmarks of OA cartilage; thus, measurement of cartilage mechanical properties may also be beneficial for earlier OA detection. However, the relative predictive ability of compo…

OBJECTIVES: To identify molecular differences between chondrocytes from osteophytic and articular cartilage tissue from OA patients.
METHODS: We investigated genes and pathways by combining genome-wide DNA methylation, RNA sequencing and quantitative proteomics in isolated primary chondrocytes from the cartilaginous layer of osteophytes and matched areas of low- and high-grade articular cartilage across nine patients with OA undergoing hip replacement surgery.
RESULTS: Chondrocytes from osteophytic cartilage showed widespread differences to low-grade articular cartilage chondrocytes. These d…

BACKGROUND: Posttraumatic osteoarthritis (PTOA) is directly associated with early acute articular cartilage injury. Inhibition of cartilage destruction immediately following joint damage can effectively slow or prevent PTOA progression. Therefore, we sought to determine intervention targets and therapeutic strategies in the acute stage of cartilage injury. The benefits of chronic intermittent hypobaric hypoxia (CIHH) extend to various body tissues, but its impact on acute cartilage injury remains unclear. We selected PTOA initiation as the therapeutic window and administered CIHH treatment imm…

BACKGROUND: Arthropathy is a prevalent and invalidating complication of acromegaly with a characteristic radiographic phenotype. We aimed to further characterize cartilage and bone abnormalities associated with acromegalic arthropathy using magnetic resonance imaging (MRI).
METHODS: Twenty-six patients (23% women, mean age 56.8 ± 13.4 years), with active (n = 10) and controlled acromegaly (n = 16) underwent a 3.0 T MRI of the right knee. Osteophytes, cartilage defects, bone marrow lesions and subchondral cysts were assessed by the Knee Osteoarthritis Scoring System (KOSS) method. Cartilage th…

OBJECTIVE: To investigate the parathyroid hormone-related peptide (PTHrP) nuclear localisation segment (PTHrP-NLS) and parathyroid hormone 1 receptor (PTH1R) signal-dependent rehabilitative effects of CD90+ chondrocytes (CD90-CCs) on degenerative temporomandibular joint (TMJ) cartilage.
DESIGN: Changes in PTHrP and PTH1R expression in CD90-CCs in TMJ cartilage in response to bilateral anterior elevation (BAE), which promotes TMJ cartilage thickening, were measured in female mice. A genetic modulation strategy was adopted. CD90-CCs were isolated from PTHrP-NLS conditional mutation (CD90-CreER;…

Meckel's and condylar cartilages are key to mandible development, with Meckel's cartilage acting as a template and condylar cartilage as a growth center. In achondroplasia, the most common form of genetic dwarfism, abnormalities of these cartilages lead to micrognathia, with significant functional repercussions for affected individuals. How FGFR3 overactivation in achondroplasia disrupts Meckel's and condylar cartilages is largely unknown. Our aim was to identify the pathways driving these disruptions by analyzing the genes expressed in these cartilages in a mouse model mimicking achondroplasi…

PURPOSE: Congenital tracheal stenosis is a disease in which complete tracheal cartilage rings (CTCR) cause airway narrowing. Although tracheal cartilage malformation has been suggested as a cause of CTCR, no histological studies have been performed. Here, we report a comparison of the tissues from CTCR and normal tracheal cartilage.
METHODS: Thirty-one infants who underwent slide tracheoplasty at our institution from May 2016 to August 2019 were included. Tissues from ten autopsy cases without tracheal lesions were used as controls. The survey items were tracheal cartilage cell density, carti…

Relapsing polychondritis, a comparatively rare condition systemically affecting organs containing collagen, such as cartilage tissue, is diagnosed based on clinical manifestations and pathological findings. Our case report involves a respiratory symptom alone. A 65-year-old man seen for fever and dyspnea was found in computed tomography and endoscopy to have subglottic and tracheal stenosis, necessitating tracheostomy and tracheal cartilage biopsy. Pathologically bony cartilage change was seen but no definite auricular inflammation findings. Additional auricular cartilage biopsy, showed no inf…

This study describes the morphology of the laryngeal apparatus in bowhead whales (Balaena mysticetus) with respect to respiration, deglutition, and vocalization. We also examined the intrinsic cricoarytenothyroid muscle (Musculus (M.) diverticuli laryngei) which forms the laryngeal diverticulum, to ascertain its interactions with the laryngeal cartilages during respiration and sound production. Five fetal larynges and four from adult whales were studied using noninvasive imaging, as well as macroscopic and microscopic techniques. The larynx extends from the skull base into the thoracic inlet. …

Until now it is not known whether osteophytes of the femoral head develop because of pathological joint alterations or arise from normal remodeling processes secondary to osteoarthrosis. Firstly, we analysed the topographical localization of osteophytes. We then compared the extracellular matrix components of macroscopically normal cartilage from the margin of osteophytes with osteophytic cartilage from weight bearing and non-weight bearing zones by histochemical staining of low and heavily sulfated glycosaminoglycans. For examination 65 femoral heads were taken during endoprosthetic hip surge…

Bony and cartilage defects of the nose should be reconstructed for both esthetic and functional purposes. In the past, autogenous bones, such as rib bones, cranial bones, and other bone grafts, have been used to reconstruct bony defects of the nose. Rib and ear cartilage have been used to reconstruct cartilage defects of the nose. A congenital subtotal bony-cartilaginous defect of the nose was reconstructed using cranial bone for the nasal bony defect, ear cartilage for the tip cartilage deficiency, and rib cartilage for the septal defect, in the present patient.

Posttraumatic osteoarthritis (PTOA) results in joint pain, loss of joint function, and impaired quality of daily life in patients with limited treatment options. We previously demonstrated that epidermal growth factor receptor (EGFR) signaling is essential for maintaining chondroprogenitors during articular cartilage development and homeostasis. Here, we used a nonsurgical, loading-induced PTOA mouse model to investigate the protective action of EGFR signaling. A single bout of cyclic tibial loading at a peak force of 6 N injured cartilage at the posterior aspect of lateral femoral condyle. Si…

INTRODUCTION: Repairing damaged cartilage poses significant challenges, particularly in cases of congenital cartilage defects such as microtia or congenital tracheal stenosis, or as a consequence of traumatic injury, as the regenerative potential of cartilage is inherently limited. Stem cell therapy and tissue engineering offer promising approaches to overcome these limitations in cartilage healing. However, the challenge lies in the size of cartilage-containing organs, which necessitates a large quantity of cells to fill the damaged areas. Therefore, pluripotent stem cells that can proliferat…

We investigated the proximal tibiae in adult, middle-aged and old (6,12, and 30 months) guinea pigs with spontaneous arthrosis by quantitative morphometry. Since the cartilage destruction develops predominantly in the medial tibial plateau, the lateral side may serve as an internal control, In established lesions, cartilage destructions were focal, surrounded by a brim of thickened cartilage that was grossly intact, but showed cell clustering, hypertrophy and increased metachromasia. Peripherally, there was a distinct transition to normal-appearing cartilage. Subjacent to the cartilage ulcerat…

Osteochondrosis (OC) of the articular epiphyseal cartilage complex (AECC) is a developmental disease that is present in the first weeks of life. It is characterized by focal chondronecrosis and retention of growth cartilage due to failure of endochondral ossification. Fissures may extend from the lesion through the overlying articular cartilage to create a cartilage flap and an osteochondral fragment. This articular form is known as osteochondritis dissecans (OCD). There have been many hypotheses about the etiopathogenesis of OC of the AECC including, amongst others, ischemia of growth cartila…
