Collagen peptides from Skipjack tuna (Katsuwonus pelamis) bones: Composition, characterization, and mechanism of high-fat diet-induced NAFLD amelioration.

This study aimed to analyze the amino acid composition and characterize the sequences of collagen peptides from Skipjack tuna bones (TBCPs) by liquid chromatography-tandem mass spectrometry (LC-MS/MS), and further investigate the function and mechanism of action of TBCPs in nonalcoholic fatty liver disease (NAFLD). The results showed that TBCPs contain 16 types of amino acids, among which glycine is the most abundant, and hydrophobic amino acids account for 40.75 %. Moreover, a total of twenty-eight active peptide sequences with an ALC ≥ 95 % were identified from TBCPs. In vivo test results re…

Spinal cord transection repair occurs when Nestin+ cells differentiate into neurons within a taxol-collagen-enhanced microenvironment in mice.

BACKGROUND: Previous studies have shown that taxol promotes axon regeneration in nerve repair, but fails to bridge the two ends of a completely transected spinal cord. Our prior in vitro research revealed that taxol, a microtubule-stabilizing agent, promotes neural stem cells (NSCs) differentiation into neurons while inhibiting astrocyte differentiation. In vivo studies further demonstrated that taxol-scaffold enhances functional recovery in animals with complete spinal cord injury (SCI). This study aims to directly validate the role of taxol-collagen in guiding NSCs to differentiate into neur…

NC410, a bivalent LAIR-2 construct, remodels collagen in the tumor microenvironment and abrogates neutrophil-driven T cell suppression.

We previously observed that circulating human neutrophils exposed to epithelial ovarian cancer (OC) ascites fluid supernatants (ASC) and malignant effusions from other tumors acquire T cell suppressor function. Collagen motifs ligate LAIR-1, an inhibitory SHP-1-dependent checkpoint broadly expressed on immune cells. We hypothesized that NC410, a bivalent LAIR-2 construct that inhibits LAIR-1-collagen binding, would rescue neutrophil-driven T cell non-responsiveness. NC410 remodeled ASC collagen resulting in neutrophil clustering and reduction in neutrophil-T cell contact, abrogated ASC-induced…

hUC-MSCs loaded collagen scaffold for refractory thin endometrium caused by Asherman syndrome: a double-blind randomized controlled trial.

In this single-center, double-blinded, randomized controlled trial, we investigated whether human umbilical cord-derived mesenchymal stromal cells loaded collagen scaffolds (hUC-MSC/CS) could improve the cumulative live-birth rate (cLBR) in infertile women with refractory thin endometrium (RTE). We randomly assigned 25 subfertile women with RTE, in a 1:1 ratio, to receive hysteroscopic adhesiolysis and plowing plus either hUC-MSC/CS or saline/CS (control) for intrauterine implantation. Uterine fluid was collected on the embryo transfer day for RNA-sequencing to explore the potential mechanisms…

Therapeutic potential of miR-10a overexpressing mesenchymal stem cell-derived extracellular vesicles in modulating inflammation in collagen-induced arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune condition that leads to joint damage. Mesenchymal stem cells (MSCs) are being recognized as a promising treatment option because of their capacity to modulate immune responses. Their therapeutic effects are mediated by released extracellular vesicles (EVs) which contain microRNAs known to influence inflammatory processes. This research focused on the impact of bone marrow MSC (BM-MSC)-derived EVs overexpressing miR-10a on cytokine production in a mouse model of collagen-induced arthritis (CIA). miR-10a was overexpressed in MSCs derived from bon…

Diabetic bone fragility through advanced glycation end product-collagen axis: Mechanisms and therapy of sodium glucose cotransporter 2 inhibitors.

Type 2 diabetes markedly elevates fracture risk despite normal or high bone mineral density, a paradox reflecting qualitative skeletal deficits rather than loss of mass. Chronic hyperglycemia fosters the accumulation of advanced glycation end products in bone; their nonenzymatic crosslinks stiffen type I collagen, impair mineralization, and erode mechanical strength. By engaging the receptor for advanced glycation end products, these adducts activate nuclear factorκB and mitogen-activated protein kinase cascades, amplifying oxidative stress, inflammation, osteoblast dysfunction, and osteoclast…

Comparative analysis of antioxidant activity and accelerated collagen expression of human skin fibroblasts with gelatin hydrolysates from different sources.

Gelatin hydrolysates have been extensively used as nutraceutical agents, exhibiting significant biological activity. This study aimed to compare the antioxidant activity and enhancement of collagen expression in human skin fibroblast (HDF-α) cells treated with gelatin hydrolysates from different sources. Results indicate that the gelatin hydrolysates exhibited remarkable DPPH·, ABTS·+ and OH· scavenging capabilities. Notably, the gelatin hydrolysates derived from bovine, donkey and tilapia skin showed significantly higher scavenging capacities than those from other sources (p < 0.05). Gelatin …

Engineered Drug-Amphiphile Conjugate Nanoparticles for Targeted Inhibition of AQP4-Mediated NLRP3 Inflammasome Signaling in Collagen-Induced Rheumatoid Arthritis.

Aquaporins (AQPs) are transmembrane proteins that transport water, small solutes, and molecules across cell membranes. Studies have reported the role of AQPs in the activation, migration, and proliferation of immune cells, thus modulating the pathogenesis of autoimmune disease. In joints, the enhanced AQP4 expression exaggerates pathological changes like hydrarthrosis, acidosis, and hyperosmotic stress-inducing dysfunction of the articular chondrocytes, leading to articular cartilage destruction in collagen-induced arthritis (CIA). Acetazolamide (AZM), a sulfonamide carbonic anhydrase inhibito…

Bioengineered Chitosan-Collagen-Honey Sponges: Physicochemical, Antibacterial, and In Vitro Healing Properties for Enhanced Wound Healing and Infection Control.

Bacterial-mediated infections represent a major risk factor for chronic wounds. Numerous polymeric dressings have been proposed to reduce this incidence and promote wound healing. In the present investigation, chitosan/collagen/honey-based sponges were prepared by freeze-drying. The effect of honey incorporation at different concentrations on the physicochemical and antibacterial properties of the sponges was evaluated. The SEM images showed that the surface and cross-sections of all samples had a porous structure. The pore size gradually increased in the range of 78.14 to 126.9 μm due to the …

Collagen-Coated 3D-Printed Magnesium Alloy Scaffold with Controlled Mg2+ Release for Enhanced Repair of Critical-Sized Bone Defects.

Critical-sized bone defects pose a significant clinical challenge due to the limitations of existing repair strategies. An ideal bone scaffold is expected to provide mechanical support while promoting osteogenesis and angiogenesis. However, biodegradable magnesium (Mg) alloys face challenges in controlling rapid degradation, leading to detrimental local Mg2+ concentrations and premature mechanical failure. To address this, a collagen-coated 3D-printed magnesium alloy scaffold (COL/Mg) was developed via electrostatic adsorption. The 3D-printed Mg scaffold provides initial structural support mat…

Characterization of Recombinant Human Type II Collagen from CHO Cells, Functional Assessment of Chondrocytes and Alleviation of Cartilage Degeneration.

Type II collagen (Col2), a crucial structural protein in hyaline cartilage, is essential for cartilage integrity and facilitating injury repair. However, research on recombinant type II collagen still faces many challenges, such as structure and yield, which limit the application of recombinant Col2 in biomedical fields. In this study, we achieved high-yield expression of full-length human Col2 (rhCol2) in CHO cells. The physical and chemical properties of rhCol2 were very close to native Col2, including molecular weight, triple helix structure, thermal stability and self-assembly capacity. Fu…

Development of silk fibroin/collagen film containing GI-20 peptide-loaded PLGA nanoparticles against corneal herpes simplex virus-1.

Herpes simplex virus-1 (HSV-1) is the primary cause of infectious blindness. Despite impressive therapeutic outcomes of conventional treatments, HSV-1 drug resistance can be easily developed. Thus, more constructive strategies should be implemented. Led by this inspiration, this work describes the potential utility of a biodegradable silk fibroin/collagen (SF/Col) film combined with GI-20-loaded poly lactic-co-glycolic acid (PLGA) nanoparticle to provide efficient and sustained delivery platform for synthetic GI-20 peptide against HSV-1. A non-irritant film containing 90 % SF and 10 % Col inco…

Human embryonic stem cell-derived immunity-and-matrix-regulatory cells on collagen scaffold effectively treat rat corneal alkali burn.

Corneal alkali burns (CAB) are a severe form of ocular injury that often leads to significant vision loss, with limited effective treatment options available beyond corneal transplantation. Immunity and matrix-regulatory cells (IMRCs) have emerged as a promising alternative due to their ability to modulate immune responses and support tissue repair. This study investigates the efficacy of IMRCs on collagen scaffolds (IMRCs-col) for treating CAB in a rat model. We developed a novel treatment combining IMRCs with a collagen scaffold to align with the ocular surface structure. In vitro analyses s…

Collagen I-induced VCAN/ERK signaling and PARP1/ZEB1-mediated metastasis facilitate OSBPL2 defect to promote colorectal cancer progression.

The global burden of colorectal cancer (CRC) has rapidly increased in recent years. Dysregulated cholesterol homeostasis facilitated by extracellular matrix (ECM) remodeling transforms the tumor microenvironment. Collagen I, a major with ECM component is highly expressed in colorectal tumors with infiltrative growth. Although oxysterol binding protein (OSBP)-related proteins accommodate tumorigenesis, OSBPL2, which is usually involved in deafness, is not associated with CRC progression. Therefore, we aimed to investigate the pathological function of OSBPL2 and identify the molecular link betwe…
