Dual-type dynamic covalent chitosan/whey protein/Sr-doped bioactive glass carriers for hydrophobic compounds: injectable hydrogels and porous scaffolds.

Chitosan hydrogel-based systems are recognized as versatile platforms for drug delivery and regenerative medicine. However, incorporating hydrophobic bioactive substances remains a significant challenge, typically addressed through complex chemical modifications such as chitosan hydrophobization. This study introduces a straightforward strategy to incorporate lipophilic compounds - retinol and resveratrol - into chitosan hydrogel-based materials using whey protein isolate (WPI) as an amphiphilic carrier. They were developed as preformed injectable hydrogels and porous scaffolds. The hydrogel n

Dynamic PEGDA-GelMA-HAMA microcarriers for efficient ex vivo expansion of human hematopoietic stem/progenitor cells.

The clinical demand for hematopoietic stem cells (HSCs) far exceeds that of the available sources, driving the need for efficient ex vivo expansion technologies. A central challenge is the design of culture systems that promote HSCs proliferation while inhibiting differentiation, a balance that is poorly maintained in conventional static cultures. In this study, we engineered a novel biomimetic microcarrier (denoted as P7G3H2) by synthesizing a composite hydrogel from poly(ethylene glycol) diacrylate (PEGDA), gelatin methacryloyl (GelMA), and hyaluronic acid methacryloyl (HAMA). This material

The Structure of Zinc Chelidonate in the Crystalline Phase, Aqueous Solution and Assessment of the Interaction with Serum Albumin.

A zinc complex of chelidonic acid (4-oxo-4H-pyran-2,6-dicarboxylic acid) was obtained by reaction with zinc oxide under isothermal conditions. Its composition was confirmed by elemental and thermogravimetric analyses, and its molecular structure was characterized using NMR and IR spectroscopy. Single-crystal X-ray diffraction revealed that the complex crystallizes as a one-dimensional coordination polymer, [ZnChel(H2O)4]n, in the triclinic space group P-1, featuring a distorted octahedral Zn(II) center coordinated by two chelidonate ligands and four water molecules. This six-coordinate arrange

Bleeding Risk Associated with Novel Oral Anticoagulants in Critically Ill Patients: A Scoping Review.

Novel oral anticoagulants (NOACs) are widely used for the prevention and treatment of thromboembolic disease; nevertheless, critically ill patients were mostly excluded from the landmark randomized clinical trials. The purpose of this scoping review is to systematically map and synthesize the available evidence on bleeding risk associated with NOACs use in critically ill patients. Furthermore, we have also focused on ICU-specific risk factors, pathophysiological mechanisms and management strategies. Critical illness is characterized by dynamic pathophysiology, dysfunction of organs, and system

Mechanically robust polyurethane elastomers enabled by soft-segment-regulated hydrogen bonds and microphase separation for ultrasound imaging medical catheters.

Conventional strategies for enhancing the mechanical robustness of thermoplastic polyurethane elastomers (TPUs) rely on hard-segment engineering, such as introducing dynamic covalent/noncovalent bonds or optimizing chain extenders, yet overlook the critical role of soft segments in governing microphase separation. Here, we present a soft-segment-regulated design that leverages crystallizable polyols to synergize hierarchical hydrogen bonding, tunable microphase separation, and strain-induced crystallization (SIC), achieving excellent mechanical performance. Among them, PU-PTMEG exhibits except

Optimal control for anti-abeta treatment in Alzheimer's disease using a reaction-diffusion model.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that severely impairs survival and quality of life. While anti-amyloid beta (Aβ) therapies can slow disease progression, their efficacy depends on personalized dosing that maximizes benefits and minimizes risks, such as amyloid-related imaging abnormalities (ARIA). Mathematical modelling offers a powerful tool for understanding AD dynamics and optimizing treatment, yet most models focus solely on temporal behaviour, overlooking spatial heterogeneity within the brain. In this study, we propose a spatially explicit reaction-dif

Acceleration patterns of the Center of Mass are altered during walking in people with incomplete spinal cord injury.

Frequent falls in ambulatory people with spinal cord injury (SCI) makes balance an area of research priority. To assess dynamic stability, we investigated center of mass (CoM) acceleration during stance phase of people with SCI (n = 34) who did not use an assistive device (n = 18) and those who used a rolling walker (n = 16). Comparison data was collected from neurologically intact (NI) individuals (n = 15). CoM acceleration was analyzed as a discrete metric using the root mean square (RMS) and as a continuous metric using statistical non-parametric mapping (SnPM). Stance phase was assessed as

Polarisation of brain dynamics in mania and depression.

INTRODUCTION: This work investigated brain dynamics underlying manic and depressive states in bipolar disorder. METHODS: Resting-state fMRI data were obtained from 69 bipolar patients-34 manic, 35 depressed-and 73 healthy controls. Intrinsic brain activity was modelled as a temporal sequence of discrete quasi-stationary states (co-activation patterns), which were compared between groups and related to symptomatology. RESULTS: Mania was associated with increased occurrence of a co-activation pattern encompassing sensorimotor network areas, which positively correlated with manic symptomatology

Amphibian Strategies Against Attacks by Flies: Host-Specificity and Threats.

Species interactions are fundamental to ecological and evolutionary processes, shaping ecosystem dynamics and driving biodiversity. Among those, interactions between flies and amphibians are common in tropical areas, yet most aspects of their ecology and evolution are understudied. Using the PRISMA method, we systematically review the literature to examine the direct and indirect threats imposed by Diptera flies attacking amphibians and the behavioral, physiological, and acoustic defenses they elicit. We delve, for instance, into the eavesdropping behavior of some dipteran species, which use a

Computational Insights into the Dissociation Pathway of the Charybdotoxin-Shaker Complex: Voltage Dependency and Potassium Modulation.

The blockade of potassium channels by protein toxins is influenced by both voltage and permeant ions, yet the structural sequence of events leading to their unbinding is not resolved. Charybdotoxin (CTX) occludes the outer exit of the pore with Lys27 positioned in the S1 site, but the intermediate states that guide its dissociation remain unclear. Using long-timescale all-atom molecular dynamics, meta-eABF free-energy calculations, and residue-level contact analysis, we mapped the dissociation pathway of CTX from the Shaker channel. External electric fields triggered an early destabilization c

Temporal dynamics of tuberculosis incidence in Piauí, northeast Brazil, 2010-2024: a time-series and decomposition analysis.

BACKGROUND: Tuberculosis (TB) remains a major public health challenge in Brazil, characterised by marked regional heterogeneity. This study aimed to analyse the temporal dynamics of TB in a state of northeast Brazil. METHODS: This was an ecological time-series study using official TB surveillance data on newly reported TB cases from 2010 to 2024. Age-standardised incidence rates (ASIRs) were analysed through seasonal-trend decomposition by Loess (STL) and Joinpoint regression models. Furthermore, the rates were decomposed to quantify the contributions of population ageing, population growth a

Development of Transferable Coarse-Grained Lipid Models with Optimized Structural and Elastic Membrane Properties.

Lipid membranes play crucial roles in cellular functions and offer diverse engineering applications. Studying their properties is critical but computationally demanding through atomistic simulations. In this work, we develop coarse-grained (CG) models for phosphocholine lipids, aimed at balancing computational efficiency and predictive accuracy with chemical and temperature transferability. We introduce chargeless CG beads with 2:1 or 3:1 mapping and optimize the force fields through a combination of systematic and accelerated approaches that integrate particle swarm optimization algorithm wit

Macroevolutionary role reversals in the earliest radiation of bony fishes.

The evolution of jaws is hypothesized to have fueled radiations among vertebrates, contributing to their overwhelming success in the present day. Past work shows rapid early expansion of diversity in jaw structure in many lineages; however, the evolutionary dynamics underlying this pattern are unclear and hindered by the lack of a robust comparative framework. Here, using a macroevolutionary approach, we explore the diversification of lower jaws in early bony fishes, a major contributor to this initial radiation. Using newly generated three-dimensional mandibular shape data from 86 species, we

Alzheimer's disease and memantine effects on NMDA-receptor blockade: non-invasive in vivo insights from magnetoencephalography.

To accelerate new treatments for Alzheimer's disease, there is the need for human pathophysiological biomarkers that are sensitive to treatment and disease mechanisms. In this proof-of-concept study, we assess new biophysical models of non-invasive human MEG imaging to test the pharmacological and disease modulation of NMDA-receptor inhibition. Magnetoencephalography was recorded during an auditory mismatch negativity paradigm from (1) neurologically-healthy people on memantine or placebo (n = 19, placebo-controlled crossover design); (2) people with Alzheimer's disease at baseline and 16-mont

Evaluating the abnormal neural variability in schizophrenia: A TMS-EEG study.

BACKGROUND: The combination of transcranial magnetic stimulation (TMS) and electroencephalography (EEG) (TMS-EEG) has become a pivotal tool for investigating neurophysiological mechanisms in schizophrenia (SZ), a disorder marked by pronounced clinical and neurobiological heterogeneity. However, conventional analyses of TMS-EEG data frequently neglect trial-to-trial neural variability, a critical dimension of dynamic brain responses that may reflect core pathophysiological features of SZ. METHODS: The TMS-EEG data and MCCB were collected from 45 SZ patients and 45 healthy controls (HC). Trial-

Single-Cell Transcriptomics on PRPF31-Mutated Retinal Organoids Reveal Early Müller Glial Activation and Progressive Photoreceptor Degeneration.

Background: Retinitis pigmentosa (RP) encompasses a group of inherited retinal disorders characterized by progressive degeneration of rod and cone photoreceptors, leading to vision loss. Among RP subtypes, RP11 is linked to mutations in PRPF31, a key spliceosome component, resulting in retinal cell dysfunction. Although PRPF31 is ubiquitously expressed, its mutations predominantly impact retinal cells, leading to the progressive loss of photoreceptors. Despite significant progress, studies have focused on photoreceptor and retinal pigment epithelium dysfunction in late disease stages, leaving
