Sea Cucumber Fucoidan Inhibits Helicobacter pylori Gastritis via MAPK/NF-κB Signaling and Gut Microbiota Modulation.

Antibiotic-based eradication therapy forHelicobacter pylori faces challenges such as the increasing number of antibiotic-resistant strains and gut microbiota dysbiosis. The study evaluated the effects of fucoidan from sea cucumber cooking liquid (Fuc-SC) on Helicobacter pylori Sydney strain SS1 (Hp SS1)-associated gastritis. The backbone structure of Fuc-SC was composed of alternating → 3)-α-L-Fucp-(1 → 4)-α-L-Fucp-(1 → linked fucose residues. The mouse experiments demonstrated that Fuc-SC reduced the colonization of Hp SS1 in the gastric mucosa through decreasing the level of urease, Hp-IgG, …

Helicobacter pylori Eradication Therapy-Associated Drug-Induced Liver Injury in a Patient With Primary Biliary Cholangitis.

Drug-induced liver injury (DILI) is a rare but serious complication of Helicobacter pylori eradication therapy, especially in patients with pre-existing liver disease. We report a 75-year-old man with well-controlled primary biliary cholangitis (PBC) who developed jaundice and pruritus after treatment with vonoprazan, amoxicillin, and clarithromycin. Liver biopsy showed an enlarged portal area, enlarged hepatocytes around the central vein, eosinophil infiltration, and focal necrosis, but PBC deterioration was ruled out. A positive drug lymphocyte stimulation test confirmed DILI. This case high…

Helicobacter pylori VacA-induced gastric mucosal atrophy: a comparative analysis with other forms of atrophic gastritis.

This study aimed to evaluate the atrophic changes induced by Helicobacter pylori VacA and compare them with other forms of atrophic gastritis.A comprehensive histomorphological analysis and immunohistochemical evaluation were performed on 984 endoscopic gastric mucosal biopsy samples from patients with endoscopically confirmed atrophic gastritis. H. pylori primarily adheres to surface mucus cells, where it proliferates and produces the vacuolating cytotoxin VacA. The underlying mechanism involves VacA inducing the upward migration and compensatory proliferation of cells located in the deeper r…

Innovative approaches in HELICOBACTER pylori therapy: Clinical trials and advancements in Gastro-Retentive drug delivery systems.

HELICOBACTER PYLORI (H. PYLORI) identified in 1982, infects more than half of the world's population and is a major cause of gastrointestinal issues like gastritis, peptic ulcers, and an elevated risk of stomach cancer. The infection triggers chronic gastritis through the immune response and bacterial toxins such as VacA and CagA, which harm the stomach lining. This persistent inflammation can lead to peptic ulcers by compromising the protective mucous barrier, allowing stomach acid to create lesions. Prolonged infection with more aggressive strains increases the likelihood of gastric cancer b…

Helicobacter pylori CagA protein induces gastric cancer stem cell-like properties through the Akt/FOXO3a axis.

The presence of Helicobacter pylori (H. pylori) infection poses a substantial risk for the development of gastric adenocarcinoma. The primary mechanism through which H. pylori exerts its bacterial virulence is the cytotoxin CagA. This cytotoxin has the potential to induce inter-epithelial mesenchymal transition, proliferation, metastasis, and the acquisition of stem cell-like properties in gastric cancer (GC) cells infected with CagA-positive H. pylori. Cancer stem cells (CSCs) represent a distinct population of cells capable of self-renewal and generating heterogeneous tumor cells. Despite ev…

Impacts of Matrix Metalloproteinase-8 Genotypes, Smoking, Alcohol Drinking, and Helicobacter Pylori Infection on Gastric Cancer.

BACKGROUND/AIM: In gastric cancer (GCa) tissues, mRNA expression of matrix metalloproteinase-8 (MMP-8) is notably reduced compared to healthy tissues. Furthermore, abnormally low or elevated serum levels of MMP-8 have been linked to a significantly poor prognosis. The involvement of MMP-8 genotypes in susceptibility to GCa remains underexplored. We aimed to assess the influence of MMP-8 genotypes on GCa susceptibility and their potential interactions with smoking, alcohol consumption, and Helicobacter pylori (H. pylori) infection. PATIENTS AND METHODS: The study utilized polymerase chain reac…

Seroprevalence of Helicobacter pylori and incident myocardial infarction - A population-based Swedish nested case-control study.

AIMS: Helicobacter pylori (H. pylori) and its cytotoxin-associated gene A (CagA) have been associated with myocardial infarction (MI), but existing data are conflicting possibly due to limitations in study designs and lack of data on important confounders. The aim of this study was to determine whether H. pylori or CagA seropositivity is associated with incident MI, including MI phenotypes, and to describe temporal trends. METHODS: We used the Northern Sweden Health and Disease study, a prospective biobank with data from residents enrolled in a population-based cohort from health examinations…

AUF1-mediated inhibition of autophagic lysosomal degradation contributes to CagA stability and Helicobacter pylori-induced inflammation.

CagA, a virulence factor of Helicobacter pylori (H. pylori), is known to drive inflammation in gastric epithelial cells and is typically degraded through autophagy. However, the molecular mechanism by which CagA evades autophagy-mediated degradation remains elusive. This study found that H. pylori inhibits autophagic flux by upregulating the expression of AU-rich element RNA-binding factor 1 (AUF1). We confirmed that AUF1 does not affect autophagy initiation but instead hampers lysosomal clearance, as evidenced by treatments with 3-MA, CQ and BafA1. Upregulated AUF1 stabilizes CagA protein lev…

Helicobacter pylori promotes gastric cancer progression by activating the TGF-β/Smad2/EMT pathway through HKDC1.

Helicobacter pylori (H. pylori) infection is widely acknowledged as the primary risk factor for gastric cancer, facilitating its progression via the Correa cascade. Concurrently, Hexokinase Domain Containing 1 (HKDC1) has been implicated in the mediation of aerobic glycolysis, contributing to tumorigenesis across various cancers. However, the precise role of HKDC1 in the inflammatory transformation associated with H. pylori-induced gastric cancer remains elusive. In this study, transcriptome sequencing revealed a significant correlation between HKDC1 and H. pylori-induced gastric cancer. Subse…

Helicobacter pylori positive oral squamous cell carcinoma demonstrate higher pathological tumor staging and poorer overall survival.

BACKGROUND: Helicobacter pylori (H pylori), a bacterium characterized by its spiral shape and gram-negative nature, impacts approximately half of the global population, showing a greater prevalence in developing nations. There are various factors that contribute to the pathogenicity of H pylori in the gastric mucosa, leading to gastric ulcer, gastritis and gastric cancers. The relationship between H pylori and gastric cancers has been well documented. The association between Oral Squamous Cell Carcinoma (OSCC) and H pylori still remains a grey field. The study aimed to evaluate the presence of…

Oral Live-Carrier Vaccine of Recombinant Lactococcus lactis Inducing Prophylactic Protective Immunity Against Helicobacter pylori Infection.

Helicobacter pylori infects the gastric mucosa and induces chronic gastritis, peptic ulcers, and gastric cancer. Research has demonstrated that vaccination can induce a protective immune response and prevent H. pylori infection. Oral administration of the Lactococcus lactis live-carrier vaccine is safe and easily complied with by the public. In this study, two recombinant L. lactis strains were constructed that expressed antigens of H. pylori urease subunit alpha (UreA) and UreA fused with Escherichia coli heat-labile toxin B subunit (LTB-UreA), named LL-UreA and LL-LTB-UreA, respectively. The…

Bioactive Properties of Campomanesia lineatifolia: Correlation Between Anti-Helicobacter pylori Activity, Antioxidant Potential and Chemical Composition.

Helicobacter pylori is found in the stomach of patients with chronic gastritis and peptic ulcers, infecting approximately half of the world's population. Current treatment for H. pylori infection involves a multi-drug therapeutic regime with various adverse effects, which leads to treatment abandonment and contributes to the emergence of resistant strains of H. pylori. Previously, we demonstrated that the essential oil of Campomanesia lineatifolia leaves exhibited an anti-H. pylori activity. In this study, we aimed to evaluate the phenolic content of the phenolic-rich ethanol extract (PEE) fro…

A new multi-epitope DNA vaccine against Helicobacter Pylori infection in a BALB/c mouse model.

BACKGROUND: Helicobacter Pylori (H. Pylori) is a pathogen that may invade the human stomach. This bacterial strain is now causing widespread concern and considerable health issues worldwide. In contrast to antibiotic treatment, which may lead to drug resistance, vaccination therapy is emerging as a possible immunotherapy option for H. Pylori. DNA vaccines are a potential option to traditional vaccines among vaccine research methods. Furthermore, the multiepitope DNA vaccination may induce a broader immune response to suppress H. Pylori infection. METHODS: Four target antigenic proteins (outer…
