Persicaria odorata (Lour.) contains considerable amounts of phytochemicals, including phenolics and essential oils, which have been reported to inhibit α-glucosidase activity. However, which bioactive compounds play a key role in inhibiting the α-glucosidase activity is unclear. To address this issue, the methanolic extract of P. odorata was assayed via in vitro studies, and forty-four compounds in P. odorata extracts were elucidated using in silico studies. The extract strongly inhibited yeast α-glucosidase with an IC50 value of 0.31 µg/mL, which was 450-fold more potent than acarbose (IC50 =

Phytochemical investigation of the flowers of Viburnum macrocephalum f. macrocephalum (VMM), a sterile horticultural form native to China, led to the isolation of two new cadinane-type sesquiterpenes, viburetones A (1) and B (2), together with fifteen known compounds (3-17). Compounds 1 and 2 represent the first examples of this skeletal type from the genus Viburnum. The structures of the new compounds were elucidated by comprehensive analysis of their 1D and 2D NMR, HRESIMS, and ECD data. All isolates were evaluated for their inhibitory activities against α-amylase and α-glucosidase, key enzy

Keratin is an abundant structural fibrous protein and extremely recalcitrant biopolymer. β-Keratin is the major constituent of feathers, which, due to the widespread poultry industry, has become a major waste product. Biotechnological upcycling of feather waste has gained interest as various bacteria and fungi capable of degrading keratin have been isolated. These microorganisms produce proteases, termed keratinases, responsible for the enzymatic hydrolysis of keratin. The structural properties that confer keratinolytic activity to proteases are, however, not well understood. Here, we investig

Accelerating molecular probe discovery and lead optimization requires accurate and efficient binding affinity prediction. Here we present PBCNet2.0, a Cartesian tensor-based Siamese neural network for protein-ligand relative binding affinity prediction. Trained on 8.6 million protein-ligand complex pairs, PBCNet2.0 achieves zero-shot accuracy similar to computationally intensive physics-based simulations while remaining highly efficient. Retrospective prioritization experiments show that PBCNet2.0 improves optimization efficiency by 7.18-fold and reduces resource use by 41%. Mechanistic analys

Discovery of active ingredients from complex natural products remains a critical challenging task in drug development, primarily due to the lack of rapid and precise analytical tools for bioactive compound screening. To address this challenge, we developed a biosensing-assisted computer-aided screening strategy. A covalent organic framework (COF) derived photocatalytic artificial oxidase was synthesized with robust catalytic capability toward TMB under natural light excitation. Leveraging arbutin as the chromogenic substrate, a sensitive biosensing assay was developed for α-glucosidase (α-Glu)

Developing efficient, low-cost, and portable screening methods for α-glucosidase inhibitors (AGIs) is of great significance for discovering natural antidiabetic drugs. Accordingly, this study constructed a colorimetric dual-platform system utilizing Fe-doped MoOx (Fe-MoOv) nanozymes with superior peroxidase-like activity, featuring both high-throughput capability and portability. This system functions by linking α-glucosidase (α-Glu) activity to a colorimetric readout: active α-Glu leads to a colorless output, whereas its inhibition by an AGI preserves an intense blue color, allowing for strai

In this study, a phytochemical investigation of Bauhinia brachycarpa led to the isolation of 23 compounds, including three new ones: bausesquitate C (1), 5-(benzofuran-2-yl)-2,3-dimethoxyphenol (2), and its β-D-glucoside (3). In addition, 14 known compounds were reported from this species for the first time. Compound 1 exhibited moderate α-glucosidase inhibitory activity (IC50 = 5.39 ± 0.30 μM) and acted as a mixed-type inhibitor (Ki = 0.29, Kis = 21.3), with stronger affinity for the free enzyme. Its binding mode and conformational stability were further elucidated by molecular docking and mo

Pain and inflammation resulting from orthopedic trauma have a significant negative impact on early recovery and rehabilitation. A proteolytic enzyme called serrapeptase (serratiopeptidase), which has anti-inflammatory properties, has been used with increasing frequency in surgery and trauma-related surgery. Nevertheless, clinical evidence of its efficacy in orthopedic trauma remains scant. The objective of the study was to determine the effectiveness, safety, and tolerability of high-dose serrapeptase (serratiopeptidase; DANZ-N 30 mg) used as an adjunctive therapy in reducing inflammation and

Autosomal recessive hypophosphatemic rickets type 2 (ARHR2) caused by biallelic ENPP1 mutations is a rare disorder with a broad phenotypic spectrum. We describe 3 affected siblings from a consanguineous family who presented with markedly heterogeneous clinical features. The proband exhibited classical signs of rickets with progressive lower-limb deformities, short stature, and elevated alkaline phosphatase. Her older sister demonstrated limited elbow extension, conductive hearing loss, and vascular stenoses, while the youngest sibling developed early biochemical abnormalities before overt skel

Mycoplasma gallisepticum is a major poultry pathogen responsible for chronic respiratory disease and substantial global economic losses. Its ability to establish chronic infections reflects its effective immune evasion strategies, but the mechanisms underlying this remain poorly understood. Some pathogenic mammalian mycoplasmas use the Mycoplasma Immunoglobulin Binding-Protease (MIB-MIP) system to capture and cleave host immunoglobulins (Ig), but the functionality and host-specificity of this system in M. gallisepticum have not been examined. We aimed to functionally characterise all the MIB-M

INTRODUCTION: Mutations in myogenesis regulating glycosidase (MYORG), result in autosomal recessive (AR) form of Primary Familial Brain Calcification (PFBC) which is a rare neurodegenerative disease. PFBC is characterized by symmetric brain calcifications, particularly in the thalami, cerebellum, basal ganglia, and subcortical white matter. To date, eight genes have been linked with PFBC, however, currently about half of people with PFBC remain without a genetic diagnosis. Among these genes, MYORG, JAM2, CMPK2, and NAA60 are associated with an AR-PFBC. Within AR-PFBCs, the frequency of mutatio

Glycerophosphodiester phosphodiesterases (GDPDs) enzymes are known to be involved in phospholipids degradation pathways, where glycerophosphodiesters are hydrolyzed to glycerol-3-phosphate (G3P) and corresponding alcohol. In plants, GDPDs are involved in phosphate deficiency adaptive responses and have been shown to impact root length, but the precise mechanism remains unclear. This study focuses on the rice GDPD5 gene and its role in regulating primary root growth. Our research demonstrates that OsGDPD5 encodes a functional GDPD enzyme and could hydrolyze glycerophosphocholine and glycerophos

Melanoma is an aggressive skin cancer with a high metastatic potential, influenced by both genetic and environmental factors. Proteases play a key role in shaping the tumor microenvironment and enabling transformed cells to actively colonize distant sites (metastasis). We performed proteomic mapping of protease cleavage sites in formalin-fixed paraffin-embedded tissue samples and profiled potentially active proteases in samples from melanoma patients with distinct prognostic outcomes. Although protein abundance alone did not indicate potential markers of disease progression, the observed cleav

Background: INPP5E-related retinopathy (INPP5E-RR) is a rare genetic disorder caused by biallelic pathogenic variants in the INPP5E gene, which encodes an enzyme critical for phosphoinositide signaling. While early-onset rod-cone dystrophy is a hallmark feature, detailed longitudinal data on the phenotype are scarce. This study aims to report a 6-year longitudinal assessment of retinal structure and function in a case of non-syndromic INPP5E-RR. Methods: A 42-year-old female proband with compound heterozygous pathogenic missense variants in INPP5E (p.Arg486Cys and p.Arg378Cys) was monitored fr

This study aimed to determine whether dietary bee pollen protects common carp (Cyprinus carpio) from deltamethrin (DM)-induced oxidative stress by modulating serum paraoxonase (PON) and arylesterase (ARE) activities. DM, a widely used pyrethroid insecticide, can impair antioxidant defenses in aquatic organisms. PON and ARE, two HDL-associated enzymes with antioxidant properties, were used as biomarkers to evaluate oxidative stress. Fish were exposed to two sublethal concentrations of DM (0.018 and 0.036 µg/L) for 14 days, with or without dietary supplementation of 2% chestnut-origin bee pollen
