Spleen metabolomics coupled with gut microbiome analysis to elucidate the immunomodulatory mechanisms of longan polysaccharides against cyclophosphamide-induced immunosuppression in mice.

Longan polysaccharide (LP) has exhibited excellent immunomodulatory activities by modifying gut microbiota but the specific regulatory mechanism remains unclear. Therefore, spleen metabolomics and metagenomic sequencing of gut microbiota were combined to investigate the immunomodulatory mechanism of LP in cyclophosphamide (CPA)-induced immunosuppressed mice with an intact and antibiotic-depleted microbiota. The results indicated that LP significantly restored thymic and splenic indices, increased lymphocyte proliferation, and mitigated damage to immune organs. LP up-regulated the ratio of CD4+

Impact of FCGR2A and FCGR3A Gene Variants on the Response to Rituximab in Patients With Glomerular Diseases.

Rituximab is an anti-CD20 monoclonal antibody used in autoimmune diseases, including glomerular diseases. The FCGR2A (rs1801274) and FCGR3A (rs396991) variants have been suggested to affect rituximab efficacy; this study evaluates their impact on rituximab efficacy in glomerular diseases. A clinical trial was performed including adults with glomerular diseases who required rituximab (NEFRTX; EudraCT: 2020-000484-23). Patients received 1.0 g or 0.5 g of rituximab on day 1 (± day 14); biochemical parameters and rituximab concentration were measured, and FCGR2A and FCGR3A variants were characteri

The influence of immune factors on bone metabolism in autoimmune thyroid diseases.

Autoimmune thyroid diseases (AITD) primarily includes Graves' disease (GD) and Hashimoto's thyroiditis (HT), which share common features such as the presence of thyroid autoantibodies and lymphocyte infiltration. Studies have shown that immune cells and their secreted cytokines in AITD not only participate in thyroid autoimmune responses but also regulate osteoblast and osteoclast activity in bone metabolism, affecting bone resorption and formation. Additionally, thyroid autoantibodies may directly affect bone metabolism through mechanisms like oxidative stress. This review emphasizes the dual

Evaluation of the trypanocidal and immunomodulatory effects of LDT409, a cardanol derivative from cashew nut shell liquid.

INTRODUCTION: Chagas disease (CD), caused by Trypanosoma cruzi, is a neglected tropical illness with significant public health impact, particularly due to its cardiac manifestations. Current etiological treatments, benznidazole and nifurtimox, are limited by prolonged administration, adverse effects, and low cure rates, especially in chronic cases. Natural products, such as cashew nut shell liquid (CNSL), are a promising source of bioactive compounds with trypanocidal and immunomodulatory properties, and the added benefit of low production costs, a valuable advantage in addressing neglected tr

Case report: Parvovirus B19 encephalitis following rituximab therapy in a patient with immune-mediated thrombocytopenia.

Parvovirus B19 infection is mostly asymptomatic in immunocompetent individuals but may cause severe manifestations when humoral immunity is impaired. Anti-CD20 therapies such as rituximab induce prolonged B-cell depletion and hypogammaglobulinemia, affecting the immune coordination and lowering the threshold for viral neuroinvasion and persistence within the central nervous system (CNS). We present a case of a 61-year-old patient who developed new-onset seizures 9 months after completing rituximab as fourth-line therapy for immune-mediated thrombocytopenia. Brain MRI showed a focal hyperintens

Utility of mouse precision cut lung slices as an in vitro model for interrogating the lung immune response against bacterial pathogens in the context of immunomodulatory therapeutics.

High rates of respiratory infections have been observed in patients following treatment with immunomodulatory therapeutics, yet preclinical assessment and mechanistic understanding of drug-associated infection risk remains a challenge. Here, an ex vivo infection model of mouse precision-cut lung slices (PCLS) is described to address this gap. Naïve mouse PCLS were pre-treated with immunomodulatory drugs previously reported to exacerbate clinical infection risk (Idelalisib, Anakinra and Tofacitinib), followed by incubation with the lung pathogen Streptococcus pneumoniae. Bacterial uptake by the

Immune reconstitution and tolerance-inducing therapies as promising therapeutic approaches in multiple sclerosis.

OBJECTIVES: Multiple sclerosis (MS) is a complex, progressive neurodegenerative autoimmune disease and a major cause of neurological disability worldwide. MS affects approximately 2.8-3 million people, predominantly presenting as relapsing-remitting MS (RRMS) that frequently converts to secondary progressive disease, while effective options for progressive phenotypes remain limited. METHODS: This review reframes MS immunotherapy through a tolerance-centric paradigm, distinguishing continuous maintenance disease-modifying therapies (DMTs) from immune reconstitution therapies (IRTs) and emergin

Preparation and characterization of immunopeptides isolated from pig spleen and evaluation of their immunomodulatory properties in vitro and in vivo.

The importance of small bioactive peptides derived from pig spleen have been used to enhance immune responses and support intestinal health. However, there is a lack of information regarding the conformational relationship and their effects on immune function of pig spleen proteins (PSPs). The objective of this study was to prepare and assess the immunomodulatory characteristics of immunopeptides from PSP. Firstly, enzymatic hydrolysates from PSP were prepared using alkaline protease and aminopeptidase, and small hydrolysate fractions with a <3 kDa were separated by SDS-PAGE and GPC. The bioac

Immunomodulatory activity of single chitooligosaccharides in LPS-induced RAW264.7 cells and endotoxemia mice.

Chitooligosaccharides is a green and non-toxic amino sugar, and its immunomodulatory activity has attracted much attention in recent years. However, most of the test samples in the existing chitooligosaccharides activity studies are mixtures, and the immunomodulatory structure-activity relationship is difficult to understand. In the present study, we successfully isolated five single chitooligosaccharides from dimer to hexamer by ion-exchange chromatography, and further investigated the size effects of chitooligosaccharides on its immunomodulatory activity. In vitro experiments showed that chi

Concomitant anti-CGRP and immunomodulatory treatments in patients with migraine: towards integrated management strategies.

BACKGROUND: Preclinical evidence supports the immunoregulatory role of calcitonin gene-related peptide (CGRP) in migraine pathophysiology. The increasing use of anti-CGRP therapies in patients with migraine and other comorbidities raises the question whether the potential use of anti-CGRP monoclonal antibodies (CGRP-mAbs) therapies in combination with other immunological therapies is effective and safe. METHODS: This multicenter study included patients with migraine receiving CGRP-mAbs combined with immunosuppressive and immunomodulatory treatments. Clinical and demographic data, treatment hi

Monitoring Brain Activity with EEG Source Localization in Rituximab-Treated Anti-NMDAR Encephalitis: A Case Study.

Anti-N-methyl-D-aspartate receptor (anti-NMDAR) encephalitis is a severe autoimmune encephalitis that often demonstrates a favorable response to immunotherapy, including rituximab. While disease outcomes have been widely documented, longitudinal characterization of brain activity changes following treatment remains limited. Electroencephalography (EEG) source localization provides a non-invasive approach for assessing regional brain dynamics. We report a case of a 17-year-old male patient with anti-NMDAR encephalitis who underwent serial EEG recordings before and after rituximab administration

Efficacy of early rituximab treatment in anti-mGluR1 encephalitis: a case report.

BACKGROUND: Anti-mGluR1 encephalitis is a form of autoimmune encephalitis, with limited reports globally and only two cases reported from Japan. There are uncertainties regarding the optimal immunosuppressive agents and individual drug responses. Herein, we report a case of anti-mGluR1 encephalitis from Japan, notable for a favorable outcome after an early rituximab initiation and an observed increase in cerebellar blood flow during the early disease stages. CASE PRESENTATION: The patient was a 65-year-old woman who experienced dizziness for 3 months, followed by worsening gait instability. S

Immunomodulatory effects of Cryptococcus neoformans capsular polysaccharides on macrophage infected with Trypanosoma cruzi.

Cryptococcus neoformans is an opportunistic fungal pathogen with a global distribution and is the causative agent of cryptococcosis, a disease that primarily affects immunocompromised individuals. The infection typically begins in the lungs and, in severe cases, can progress to meningoencephalitis and even death. Biochemical studies have shown that the fungal capsule is predominantly composed of glucuronoxylomannan (GXM), which accounts for approximately 88% of the total composition, followed by glucuronoxylomannogalactan (GXMGal, ∼10%) and mannoproteins (∼2%). Purified capsular components hav

BACH2 and HIV: partners in crime?

HIV-1 persists lifelong despite effective antiretroviral therapy, yet the mechanisms underlying this persistence remain incompletely understood. Recent work by Wei et al. and Gao et al. reveals that the transcription factor BACH2 orchestrates CD4+ T cell memory programs fostering long-term memory formation while limiting effector differentiation, thereby promoting HIV-1 persistence.
