Causal relationship between gut microbiota and rheumatoid arthritis: A bidirectional 2-sample Mendelian randomization study.

To evaluate the bidirectional causal relationship between gut microbiota and Rheumatoid arthritis using Mendelian randomization (MR). Using gut microbiota data from the IEU OpenGWAS database (n = 5959, Finnish cohort) and rheumatoid arthritis genetic data from the Finnish database (6236 cases and 1,47,221 controls), forward and reverse MR analyses were conducted with gut microbiota and rheumatoid arthritis as exposure factors and outcomes, respectively. The significance threshold for forward MR was set at 1 × 10-5, and the significance threshold for reverse MR analysis was set at 5 × 10-8. The…

Microbiota-targeted therapeutic strategies for elderly-onset rheumatoid arthritis: based on the gut-joint axis.

Elderly-onset rheumatoid arthritis (EORA) presents distinct clinical challenges, including more refractory disease activity, higher comorbidity burden, and increased disability and mortality compared to younger-onset RA. Emerging evidence implicates the gut-joint axis-specifically the synergistic interplay between immunosenescence, inflammaging, and gut microbiota dysbiosis-in the pathogenesis of EORA. This review aims to synthesize current evidence on the role of the gut microbiota in EORA, elucidate the mechanistic links between age-related immune changes and microbial dysbiosis, and evaluat…

Targeting gut-microbiota-dependent choline metabolite trimethylamine N-oxide ameliorates bone health in ovariectomized mice.

AIMS: Postmenopausal osteoporosis (PMO) arises from estrogen deficiency and imposes a substantial global health burden due to its high fracture risk and related complications. Although choline has been suggested as a nutritional bone-protective factor, its gut microbiota-derived metabolite trimethylamine N-oxide (TMAO) has been reported to exert adverse vascular and metabolic effects, leaving its overall impact on bone health unclear. This study aims to elucidate how choline-derived TMAO regulates bone metabolism under estrogen-deficient conditions and to explore its potential therapeutic valu…

Gut Microbiota-Derived Indole-3-Propionic Acid Alleviates Diabetic Osteoporosis Through Nrf2-Mediated Ferroptosis Suppression.

Diabetic osteoporosis combines impaired bone formation with disproportionate skeletal fragility, yet the microbial metabolites linking diabetes-associated dysbiosis to bone dysfunction remain unclear. This study implicates indole-3-propionic acid (IPA), a gut microbiota-derived tryptophan metabolite, in the maintenance of skeletal homeostasis under diabetic conditions. Circulating IPA was lower in diabetic mice and in a small exploratory cohort of patients with diabetic osteoporosis. IPA concentrations were positively associated with bone mass. Metagenomic profiling linked lower IPA to impaire…

Inflammatory protein mediators linking gut microbiota to degenerative lumbar spine disorders: cross-disease genetic evidence.

BACKGROUND: Degenerative lumbar spine disorders (DLSD), including intervertebral disc disorders (IDD), degenerative spondylolisthesis, and lumbar spinal stenosis (LSS), are major contributors to low back pain and disability. Associations among gut microbiota (GM), inflammatory proteins, and DLSD have been demonstrated in prior studies. Yet, two key questions persist: whether specific circulating inflammatory proteins (IPs) mediate this association, and whether such mediation is shared across different diseases. METHODS: We performed two-sample Mendelian randomization (MR) to evaluate causal a…

Early antioxidant capacity, intestinal barrier integrity and gut microbiota drive DHAV-3 resistance in ducks.

BACKGROUND: Selective breeding for disease resistance is an effective strategy to control duck hepatitis A virus type 3 (DHAV-3) in waterfowl. However, the mechanism underlying resistance remains poorly understood, particularly those associated with antioxidant defense, intestinal development and host-microbiota interactions. METHOD: A total of 100 1-day-old Pekin ducklings were used in this study with 50 DHAV-3 susceptible and resistant ducks, respectively. Samples were collected at 7 days post-hatching (D7), D21 and D42, 10 birds per group. We compared DHAV-3 resistant and susceptible ducks…

Ultra long-lived plasma cells in the human small intestine produce microbiota-reactive IgA antibodies.

A large fraction of the intestinal microbiota is highly coated with secretory IgA, and bacteria-specific IgA is believed to shape the composition of the microbiota. A hallmark of the adaptive immune system is immunological memory to specific antigens. However, whether there is strong and persistent memory of secretory antibodies to bacterial antigens has not been determined. Here we show that ultra long-lived CD19-CD45- (age > 20 years) plasma cells (PCs) residing in the human small intestine produce IgA that binds to most taxa of a diverse anaerobic microbiota culture. Long-lived CD19-CD45+ (…

Comparative immunomodulatory and gut microbiota-regulating effects of macroalgal polysaccharides in cyclophosphamide-induced immunosuppressed mice.

Polysaccharides derived from macroalgae exhibit potent immunomodulatory and gut-regulatory activities; however, comprehensive comparisons of … stronger immunostimulatory effects, particularly in B-cell restoration and cytokine secretion. Gut microbiota profiling revealed that both polysaccharides ameliorated CTX-induced dysbiosis by enriching Lactobacillus, reducing Desulfovibrio, and increasing short-chain fatty acid levels. Collectively, these findings demonstrate that SP and UP enhance immune f …

Small intestine microbiota development prevents early-life adiposity via IL-22-mediated intestinal PPARα suppression.

Perturbation to the early-life microbiota has long-term detrimental effects on health and development, leading to increased risk for metabolic dysfunction and childhood obesity. Despite the central role of the small intestine (SI) in energy balance, the impact of SI microbiota establishment on the regulation of host metabolism and early-life adiposity remains unclear. Here, we report that disruption of a critical SI microbiota-intestinal epithelial cell circuit, specifically during a critical early-life period, drives long-lasting obesity. We demonstrate that the SI microbiota expands in abund…

The interplay between gut microbiota and Alzheimer's disease: mechanistic insights from dysbiosis to disease modulation.

Alzheimer's disease (AD) is a chronic, progressive, neurodegenerative condition marked by memory loss and cognitive decline. It is character … of amyloid deposition, affecting gastrointestinal motility and inflammatory susceptibility. The gut microbiota also produces key bioactive compounds, including neurotransmitters such as serotonin, dopamine, acetylcholine, histamine, and gamma-aminobutyric acid (GABA), which influence the central nervous system (CNS) through neural, immune, and endocrine path …

Curdlan alleviates non-alcoholic fatty liver disease by enriching butyrate-producing gut microbiota-liver axis.

The gut microbiota-liver axis is a critical target for non-alcoholic fatty liver disease (NAFLD) intervention, yet effective strategies to restore gut microbiota balance and mitigate hepatic metabolic inflammation remain limited. Curdlan, a Food and Drug Administration-approved microbial exopolysaccharide, is widely used as a food additive, but its role in modulating the gut-liver axis to combat NAFLD has never been explored. Here, we demonstrate that curdlan alleviates high-fat and fiber-deficient (HFFD) diet-induced NAFLD by orchestrating gut microbiota remodeling and enhancing gut-liver cro…

Targeting the microbiota-gut-brain axis in post-stroke insomnia: a phase-dependent therapeutic framework.

Post-stroke insomnia (PSI) is a critical biological barrier to neurorehabilitation afflicting over half of all stroke survivors. Traditional sedatives often force clinicians into a therapeutic dilemma between sleep efficacy and cognitive suppression. The microbiota-gut-brain (MGB) axis has recently emerged as a transformative target to resolve this impasse. Acute stroke triggers profound autonomic dysfunction, causing immediate intestinal barrier collapse. This "leaky gut" facilitates the systemic translocation of lipopolysaccharides (LPS) and activates the NLRP3 inflammasome. The resulting in…

Exploring the chicken respiratory microbiome: a comprehensive meta-analysis of factors shaping chicken respiratory microbiota.

Poultry is the main source of meat production worldwide. The respiratory microbiota of chickens plays a critical role in avian health and disease prevention. To better understand its composition and dynamics, we conducted a comprehensive meta-analysis focusing on factors such as age, breed, SPF status, and respiratory organs. We collected 16S rRNA data from 13 different studies available on NCBI’s PubMed and Scopus, including data on the nasal cavity, trachea, and lungs from healthy chickens. All the data were processed using QIIME2 and then analysed together. Metadata from each study was comp…

A polysaccharide from Pueraria lobata ameliorates hepatic fibrosis via gut microbiota-dependent suppression of ferroptosis.

BACKGROUND: Hepatic fibrosis, driven by oxidative stress and subsequent hepatocellular injury, represents a major worldwide health challenge … LP2's protection. Notably, the hepatoprotective effects of PLP2 were predominantly dependent on gut microbiota integrity, as direct PLP2 treatment failed to protect hepatocytes in vitro. This role was further confirmed by the abolition of protection with antibiotic treatment and the transfer of benefits via fecal microbiota transplantation. CONCLUSION: Thes …
