Screening Based on Structural and Biological Verification of Stachyose as a PPARγ-Modulating Ligand for the Treatment of Non-Alcoholic Fatty Liver Disease.

Peroxisome proliferator-activated receptor gamma (PPARγ) is a critical therapeutic target for metabolic disorders like non-alcoholic fatty liver disease (NAFLD). However, PPARγ full agonists such as rosiglitazone (ROSI) exhibit limited efficacy and off-target effects. Intriguingly, transcriptomic analyses revealed dynamic PPARγ expression during NAFLD progression-compensatory upregulation in early stages and downregulation in advanced disease-highlighting the need for novel modulators. This study investigates the therapeutic potential of stachyose (STA), a natural bioactive compound, in NAFLD

Distinct epigenetic and transcriptional profiles of Epstein-Barr virus-positive and negative primary CNS lymphomas.

BACKGROUND: Epstein-Barr virus (EBV)+ and EBV- primary CNS lymphomas (PCNSL) carry distinct mutational landscapes, but their transcriptional and epigenetic profiles have not been integrated and compared. This precludes further insights into pathobiology and molecular differences, relevant for classification and targeted therapy. METHODS: Twenty-three EBV- and 15 EBV+ PCNSL, histologically classified as diffuse large B-cell lymphomas, were subjected to RNA-sequencing and EPIC methylation arrays. Unsupervised clustering analyses were performed. Differentially expressed and differentially methyl

Programmed Cell Death via Type IV Photodynamic Therapy Using Internalized Two-Photon Activated Molecular Nanomachines.

Direct photodynamic therapy (PDT) is a growing research area currently being explored as an alternative treatment for various cancers. Compared to traditional, indirect PDT, which exploits the reaction of oxygen with the photosensitizer (PS) to damage specially targeted cells, direct PDT utilizes the PS itself to disrupt the target cell, meaning no reactive oxygen species (ROS) are generated. The activation of Type IV technologies specifically induces a structural change within the photosensitizer, resulting in the activation of its therapeutic effect. In contrast to traditional invasive surge

Moving Through Genetic Alterations and PD-L1 Expression in High-Grade Fetal Adenocarcinoma of the Lung: A Case Report and Literature Review.

BACKGROUND: Lung cancer, the most common malignancy in humans, can be classified into a wide range of subtypes and molecular variants. Fetal adenocarcinoma of the lung (FLAC) is an infrequent subtype of lung adenocarcinoma with distinct histological characteristics. However, its incidence is extremely low (0.1-0.5% of primary lung cancers), and its biological features remain poorly understood. In the era of immunotherapy and targeted therapy, characterizing the genetic alterations of FLAC is essential for developing personalized treatment strategies and improving patient outcomes. Here, we rep

Andrographolide Promotes Ferroptosis in Pancreatic Cancer via Targeting and Activating HSP90/GPX4 Ubiquitination.

Pancreatic adenocarcinoma accounts for 90% of pancreatic cancer cases, the deadliest kind. PC patients' poor immunotherapy, chemotherapy, and other responses lead to a generally failed treatment strategy. Thus, understanding molecular processes is essential for creating novel PC therapies. The natural chemical andrographolide (ADG) from Andrographis paniculata shows anticancer properties against various cancer types. The method by which ADG fights pancreatic cancer is unknown. In PC cell lines, ADG inhibited cell proliferation and migration, caused G0/G1 phase arrest, and caused cell death due

Establishing a pediatric solid tumor PDX biobank for precision oncology research.

Developing advanced preclinical models and targeted therapies is essential for reducing cancer-related deaths in children with solid tumors. Patient-derived xenografts (PDX) have the potential to replicate key elements of the original tumor, including morphology, genetic alterations, and microenvironment, making them valuable tools for studying tumor biology and drug response. We implanted 124 pediatric solid tumor samples, collected for 1 y, into NOD/SCID/IL2Rg (NSG) mice. Tumor fragments were placed subcutaneously, and the animals were monitored for up to 1 y. Histopathology, Short Tandem Re

Tumor lysis syndrome associated with bortezomib: A post-hoc analysis after signal detection using the US Food and Drug Administration Adverse Event Reporting System.

Tumor lysis syndrome (TLS) is a cancer chemotherapy-associated oncologic emergency. Although there have recently been substantial developments in cancer chemotherapy, these may increase the risk of TLS. In this study, we aimed to identify anticancer agents that increase TLS risk, as classified by a TLS panel consensus, using the US Food and Drug Administration Adverse Event Reporting System (FAERS) database. TLS reports were retrieved from the FAERS database, and reporting odds ratios (RORs) were used to estimate associations between TLS and old and new anticancer agents or their combinations.

Leukemia stem cells: the root of chronic myeloid leukemia.

Chronic myeloid leukemia (CML) is a clonal myeloproliferative disorder characterized by a chromosome translocation that generates the Bcr-Abl oncogene encoding a constitutive kinase activity. Despite remarkable success in controlling CML at chronic phase by Bcr-Abl tyrosine kinase inhibitors (TKIs), a significant proportion of CML patients treated with TKIs develop drug resistance due to the inability of TKIs to kill leukemia stem cells (LSCs) that are responsible for initiation, drug resistance, and relapse of CML. Therefore, there is an urgent need for more potent and safer therapies against

Natural Agents Modulating Ferroptosis in Cancer: Molecular Pathways and Therapeutic Perspectives.

Ferroptosis, a controlled cell death influenced by iron-dependent lipid peroxidation, presents potential therapeutic targets for cancer treatment due to its unique molecular pathways and potential drug resistance. Natural compounds, such as polyphenols, flavonoids, terpenoids and alkaloids, can influence ferroptosis via important signalling pathways, such as Nrf2/Keap1, p53, and GPX4. These are promising for combinational therapy due to their ability to cause ferroptotic death in cancer cells, exhibit tumour-specific selectivity and reduce systemic toxicity. Furthermore, these compounds, when

Essential procedures of single-cell RNA sequencing in multiple myeloma and its translational value.

Multiple myeloma (MM) is a malignant neoplasm characterized by clonal proliferation of abnormal plasma cells. In many countries, it ranks as the second most prevalent malignant neoplasm of the hematopoietic system. Although treatment methods for MM have been continuously improved and the survival of patients has been dramatically prolonged, MM remains an incurable disease with a high probability of recurrence. As such, there are still many challenges to be addressed. One promising approach is single-cell RNA sequencing (scRNA-seq), which can elucidate the transcriptome heterogeneity of individ

Pre-silencing of TNF-α by targeted siRNA delivery mitigates glucocorticoid resistance of dexamethasone in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic, progressive inflammatory autoimmune disease marked by relentless synovial inflammation and joint destruction, for which long-term remission remains challenging. Although dexamethasone (DEX) is commonly employed to rapidly control disease activity, its therapeutic effectiveness is often undermined by the development of glucocorticoid resistance (GCR) and cumulative systemic toxicities. Recent insights suggest that TNF-α-driven inflammation not only perpetuates joint pathology but also sustains a molecular landscape that favors GCR, underscoring an urgent

Carboplatin desensitization in the era of target therapies: still worthwhile?

PURPOSE: Unresectable pediatric low-grade gliomas (LGG) usually need adjuvant therapy, and carboplatin hypersensitivity reaction (HR) commonly leads to premature treatment cessation of a standard chemotherapy regimen. In the molecular era, advances in understanding tumor genetic characteristics allowed the development of targeted therapies for this group of tumors; however, cost-effectiveness assessment of treatments, especially in low-income countries, is crucial. The aim is to describe the results of carboplatin desensitization protocol in a single center in a middle-income country. METHOD:

Molecular basis of muscle wasting and emerging therapeutic strategies targeting degenerative muscle disorders.

Muscle degenerative conditions, including sarcopenia, muscular dystrophies, and trauma-induced muscle loss, severely compromise mobility, metabolism, and overall health. These disorders result from multifactorial causes such as imbalances in protein homeostasis, satellite cell dysfunction, mitochondrial stress, and chronic inflammation. Central molecular regulators such as FOXO, AMPK, mTOR, and the myostatin/SMAD axis, play pivotal roles in driving muscle atrophy. Current regenerative strategies seek to restore muscle structure and function through stem cell-based therapies (MSCs, iPSC-derived
