Carbon and hydrogen isotope fractionation of benzene and toluene during hydrophobic sorption in multistep batch experiments.

The application of compound-specific stable isotope analysis (CSIA) for evaluating degradation of organic pollutants in the field implies that other processes affecting pollutant concentration are minor with respect to isotope fractionation. Sorption is associated with minor isotope fractionation and pollutants may undergo successive sorption-desorption steps during their migration in aquifers. However, little is known about isotope fractionation of BTEX compounds after consecutive sorption steps. Here, we show that partitioning of benzene and toluene between water and organic sorbents (i.e. 1

Versatility in corrinoid salvaging and remodeling pathways supports corrinoid-dependent metabolism in Dehalococcoides mccartyi.

Corrinoids are cobalt-containing molecules that function as enzyme cofactors in a wide variety of organisms but are produced solely by a subset of prokaryotes. Specific corrinoids are identified by the structure of their axial ligands. The lower axial ligand of a corrinoid can be a benzimidazole, purine, or phenolic compound. Though it is known that many organisms obtain corrinoids from the environment, the variety of corrinoids that can serve as cofactors for any one organism is largely unstudied. Here, we examine the range of corrinoids that function as cofactors for corrinoid-dependent meta

Autoimmune thyroid diseases and nonorgan‑specific autoimmunity.

Autoimmune thyroid diseases (ATD), as the most common organ‑specific autoimmune disorder, is frequently accompanied by other organ- and nonorgan‑specific autoimmune diseases. Although the exact pathogenic mechanism of the coexistence of autoimmune disorders has not been clearly defined, genetic and environmental factors, immune defects, and hormonal changes, may play a key role in polyautoimmunity. The role of human leukocyte antigen (HLA) haplotypes, HLA-B8 and -DR3, in the overlapping of autoimmune disorders was well supported by higher frequency of these haplotypes in primary Sjögren's synd

Seroprevalence of Trichinella spiralis and Toxoplasma gondii in pigs from different housing systems in The Netherlands.

Prevalences of parasitic infections in pigs from different housing systems may vary, due to their contact with the environment, and this might have consequences for food safety. In this study, 40 organic, 9 free-range and 24 intensive farms were selected and a total of 845 serum samples were tested for antibodies specific for Toxoplasma and Trichinella using ELISA assays. The overall seroprevalence of Toxoplasma in the total number of 845 serum samples tested is 2.6%, ranging from 0.38% in intensively raised pigs to 5.62% in free-range pigs. Of the housing systems tested, 4% (intensive farms)

Potential for photoenhanced toxicity of spilled oil in Prince William Sound and Gulf of Alaska waters.

Photoenhanced toxicity is the increase in the toxicity of a chemical in the presence of ultraviolet light (UV) compared to a standard laboratory test conducted with fluorescent lighting (minimal UV). Oil products, weathered oil, and specific polycyclic aromatic compounds present in oil are 2 to greater than 1000 times more toxic in the presence of UV. The photoenhanced toxicity of oil to fish and aquatic invertebrates appears to occur through a process of photosensitization, rather than photomodification of the aqueous phase oil. In photosensitization, the bioaccumulated chemical transfers lig

Severe destructive autoimmune lesions with aging in murine Sjögren's syndrome through Fas-mediated apoptosis.

When we evaluated the age-associated changes in autoimmune exocrinopathy in a NFS/sld murine model for primary Sjögren's syndrome (SS), severe destructive autoimmune lesions developed in the salivary and lacrimal glands in the aged mice, compared with those observed in the younger model. We detected a decreased secretion of saliva and tear flow in the aged group. A significant increase of TUNEL(+)-apoptotic epithelial duct cells in the salivary glands was detected in the aged SS animal model. A higher proportion of mouse salivary gland cells bearing Fas was found in the aged group, whereas no

Autoimmune Diseases: Problems of Pathogenesis, Diagnostics and Treatment.

Autoimmune diseases (AID) are pathological states wherein a disturbance of the regulation of physiological autoimmune processes results in the appearance of antibodies, T lymphocytes, specific in respect to components of self tissues, causing structural and/or functional disturbances in target organs. Studies conducted in respect of the pathogenesis of AID deal with its main components: 1) mechanisms of oligo-polyclonal B-cellular activation; 2) participation of autoantibodies, immune complexes, DTH effectors in the development of autoimmune inflammation; 3) participation of cytotoxic lymphocy

[Autoimmune mechanisms in endocrinology--the polyglandular autoimmune endocrine syndrome].

Autoimmune endocrinopathies belong to so-called organ specific autoimmune diseases. These diseases combine very often and from the polyglandular autoimmune endocrine syndrome (PAES), where autoantibodies either destroy or stimulate individual endocrine glands or hormone receptors in target tissues. Thus a wide range of combinations of hypofunctional or hyperfunctional clinical syndromes develops. PAES is a good natural model of endocrine polyautoaggressiveness. In its development the following aetiopathogenetic factors participate: 1. Hereditary familial disposition, expressed e.g. by certain

Common Pathogens Associated with Diabetic Charcot Neuroarthropathy in the Indian Population: A Retrospective Observational Study.

BACKGROUND: Diabetic Charcot neuroarthropathy (CN) is a limb-threatening complication of long-standing diabetes mellitus and is frequently complicated by secondary infection. India-specific data on the microbiological spectrum and antimicrobial susceptibility patterns of infections complicating diabetic CN are limited. MATERIALS AND METHODS: This retrospective observational study included 82 adult patients with diabetic CN and associated foot ulcers. Deep tissue or ulcer-base samples were collected under aseptic precautions and processed for bacterial culture, organism identification, and ant

Aberrant gene expression in organs of bovine clones that die within two days after birth.

Cloning by somatic nuclear transfer is an inefficient process in which some of the cloned animals die shortly after birth and display organ abnormalities. In an effort to determine the possible genetic causes of neonatal death and organ abnormalities, we used real-time quantitative reverse transcription-polymerase chain reaction to examine expression patterns of eight developmentally important genes (PCAF, Xist, FGFR2, PDGFRa, FGF10, BMP4, Hsp70.1, and VEGF) in six organs (heart, liver, spleen, lung, kidney, and brain) of both cloned bovines that died soon after birth (n=9) and normal control

Parallel systems for social and spatial cognition reaching the cortical apex.

How are systems supporting high-level cognition organized in the human brain? We hypothesize that cognitive processes involved in understanding people and places are implemented by distinct neural systems with parallel anatomical organization. We test this hypothesis using precision neuroimaging of individual human brains on diverse tasks involving perception and cognition in the domains of familiar people, places, and objects. We find that thinking about people and places elicits responses in distinct areas of high-level association cortex within the default mode network, spanning the frontal

Microbiome and Communication Disorders: A Tutorial for Clinicians.

PURPOSE: Emerging research in the field of microbiology has indicated that host-microbiota interactions play a significant role in regulating health and disease. Whereas the gut microbiome has received the most attention, distinct microbiota in other organs (mouth, larynx, and trachea) may undergo microbial shifts that impact disease states. A comprehensive understanding of microbial mechanisms and their role in communication and swallowing deficits may have downstream diagnostic and therapeutic implications. METHOD: A literature review was completed to provide a broad overview of the microbi

The landscape of immune dysregulation in Crohn's disease revealed through single-cell transcriptomic profiling in the ileum and colon.

Crohn's disease (CD) is a chronic gastrointestinal disease that is increasing in prevalence worldwide. CD is multifactorial, involving the complex interplay of genetic, immune, and environmental factors, necessitating a system-level understanding of its etiology. To characterize cell-type-specific transcriptional heterogeneity in active CD, we profiled 720,633 cells from the terminal ileum and colon of 71 donors with varying inflammation status. Our integrated datasets revealed organ- and compartment-specific responses to acute and chronic inflammation; most immune changes were in cell composi

Spatial tuning of face part representations within face-selective areas revealed by high-field fMRI.

Regions sensitive to specific object categories as well as organized spatial patterns sensitive to different features have been found across the whole ventral temporal cortex (VTC). However, it is unclear that within each object category region, how specific feature representations are organized to support object identification. Would object features, such as object parts, be represented in fine-scale spatial tuning within object category-specific regions? Here, we used high-field 7T fMRI to examine the spatial tuning to different face parts within each face-selective region. Our results show

Age-related and disease locus-specific mechanisms contribute to early remodelling of chromatin structure in Huntington's disease mice.

Temporal dynamics and mechanisms underlying epigenetic changes in Huntington's disease (HD), a neurodegenerative disease primarily affecting the striatum, remain unclear. Using a slowly progressing knockin mouse model, we profile the HD striatal chromatin landscape at two early disease stages. Data integration with cell type-specific striatal enhancer and transcriptomic databases demonstrates acceleration of age-related epigenetic remodelling and transcriptional changes at neuronal- and glial-specific genes from prodromal stage, before the onset of motor deficits. We also find that 3D chromati

In Vivo Targets of Pasteurella Multocida Toxin.

Many Pasteurella multocida strains are carried as commensals, while some cause disease in animals and humans. Some type D strains cause atrophic rhinitis in pigs, where the causative agent is known to be the Pasteurella multocida toxin (PMT). PMT activates three families of G-proteins-Gq/11, G12/13, and Gi/o-leading to cellular mitogenesis and other sequelae. The effects of PMT on whole animals in vivo have been investigated previously, but only at the level of organ-specific pathogenesis. We report here the first study to screen all the organs targeted by the toxin by using the QE antibody th

Modular organization of the brainstem noradrenaline system coordinates opposing learning states.

Noradrenaline modulates global brain states and diverse behaviors through what is traditionally believed to be a homogeneous cell population in the brainstem locus coeruleus (LC). However, it is unclear how LC coordinates disparate behavioral functions. We report a modular LC organization in rats, endowed with distinct neural projection patterns and coding properties for flexible specification of opposing behavioral learning states. LC projection mapping revealed functionally distinct cell modules with specific anatomical connectivity. An amygdala-projecting ensemble promoted aversive learning

In-situ hybridization for the detection and identification of Histomonas meleagridis in tissues.

For use in an in-situ hybridization method, three probes (HM, TR and BL) were designed to hybridize, respectively, with (1) Histomonas meleagridis, (2) Tetratrichomonas gallinarum, and (3) a broad range of micro-organisms, including Blastocystis spp. Mono-eukaryotic cultures were used to test the specificity of the three oligonucleotides and to optimize the hybridization procedure before applying the probes to archived samples of various tissues and to a culture of Trichomonas gallinae. Specific detection of H. meleagridis was possible with the HM probe, but the other two probes were less spec
