Cognitive conflict following appetitive versus negative cues and smoking cessation failure.

RATIONALE: Attentional biases and executive control deficits may play a role in smoking cessation failure. OBJECTIVES: The object of this study was to determine whether smokers' pre-quit reaction times on a computerized modified Simon task (which assesses attentional biases and executive control deficits) predict abstinence following a quit attempt. METHODS: Participants (N = 365) in a larger smoking cessation clinical trial completed the modified Simon task twice (while 10-h nicotine-deprived vs. not deprived). In the task, two photographs (i.e., two digital slides) were displayed—one alway

Functional specialization and dynamic resource allocation in visual cortex.

We studied the spatiotemporal characteristics of cortical activity in early visual areas and the fusiform gyri (FG) by means of magnetoencephalography (MEG). Subjects performed a visual classification task, in which letters and visually similar pseudoletters were presented in different surrounds and under different task demands. The stimuli appeared in a cued half of the visual field (VF). We observed prestimulus effects on amplitudes in V1 and Cuneus relating to VF and task demands, suggesting a combination of active anticipation and specialized routing of activity in visual processing. Top-down and bottom-up attentional guidance: investigating the role of the dorsal and ventral parietal cortices.

Recent neuroimaging studies suggest that the superior parietal lobule (SPL) of the human cortex mediates goal-directed attentional orienting, while the temporo-parietal junction (TPJ) mediates stimulus-driven attentional orienting. Here, we investigated these brain-behavior correspondences by examining the performance of patients with an attentional deficit following a right hemisphere lesion. Patients completed two tasks, one sensitive to stimulus-driven attentional orienting and the other to goal-directed attentional orienting. Based on the behavioral profiles obtained on each task, patients

How do we empathize with someone who is not like us? A functional magnetic resonance imaging study.

Previous research on the neural underpinnings of empathy has been limited to affective situations experienced in a similar way by an observer and a target individual. In daily life we also interact with people whose responses to affective stimuli can be very different from our own. How do we understand the affective states of these individuals? We used functional magnetic resonance imaging to assess how participants empathize with the feelings of patients who reacted with no pain to surgical procedures but with pain to a soft touch. Empathy for pain of these patients activated the same areas (

An initial investigation of the orbitofrontal cortex hyperactivity in obsessive-compulsive disorder: exaggerated representations of anticipated aversive events?

Orbitofrontal cortical (OFC) dysfunction has been repeatedly involved in obsessive-compulsive disorder, but the precise significance of this abnormality is still unclear. Current neurocognitive models propose that specific areas of the OFC contribute to behavioral regulation by representing the anticipated affective value of future events. This leads to the hypothesis that these OFC areas are hyperactive in patients, reflecting ruminative preoccupation with future aversive events. In experimental situations, such hyperactivity should be triggered by negative affect in response to high likeliho

Individual differences in risk preference predict neural responses during financial decision-making.

We investigated the neural correlates of subjective valuations during a task involving risky choices about lotteries. Because expected value was held constant across all lotteries, decisions were influenced by subjective preferences, which manifest behaviorally as risk-seeking or risk-averse attitudes. To isolate structures encoding risk preference during choice, we probed for areas showing increased activation as a function of selected risk-level. Such response patterns were obtained in anterior (ACC) and posterior cingulate cortex (PCC), superior frontal gyrus, caudate nucleus, and substanti

Behavioural and neurophysiological correlates of human cataplexy: a video-polygraphic study.

OBJECTIVES: To investigate the behavioural and neurophysiological pattern of cataplexy. METHODS: Seven narcolepsy with cataplexy patients underwent daytime videopolygraphy using humorous movies or/and jokes to trigger cataplectic attacks. RESULTS: During segmental cataplectic attacks, EMG showed brief and irregular periods of silencing focally involving facial, neck, axial or limb muscles, sometimes coinciding with bursts of rapid eye movements. All patients enacted intentional movements in response to these segmental postural lapses. During global cataplectic attacks, EMG showed suppression

Frontal attentional responses to food size are abnormal in obese subjects: an electroencephalographic study.

OBJECTIVE: Are obese subjects characterized by a reduction of attentional cortical responses to the enlargement of food or body images? METHODS: Electroencephalographic data were recorded in 19 obese and 15 normal-weight adults during an "oddball" paradigm. The subjects were given frequent (70%) and rare (30%) stimuli depicting faces (FACE), food (FOOD), and landscapes (CONTROL), and clicked the mouse after the rare stimuli. These stimuli depicted the same frequent stimuli graphically dilated by 25% along the horizontal axis. Bioelectrical impedance indexed subjects' body fat percentage. Cort

Activity in preserved left hemisphere regions predicts anomia severity in aphasia.

Understanding the neural mechanism that supports preserved language processing in aphasia has implications for both basic and applied science. This study examined brain activation associated with correct picture naming in 15 patients with aphasia. We contrasted each patient's activation to the activation observed in a neurologically healthy control group, allowing us to identify regions with unusual activity patterns. The results revealed that increased activation in preserved left hemisphere areas is associated with better naming performance in aphasia. This relationship was linear in nature;

Severity of coronary artery disease but not degree of coronary stenosis is correlated to cerebrovascular reactivity.

BACKGROUND: A decade difference in peak incidences of coronary and cerebral ischemia assumes a protection of the brain vasculature. Therefore, we hypothesize that early indicators of cerebrovascular disease such as parameters of the neurovascular coupling show a higher correlation to coronary artery disease than morphologic parameters. METHODS: Stenotic degree (%) of coronary arteries (RCA, LCA, LAD, RCX) was determined together with the Gensini score by angiography in 96 patients (80 men; 62 +/- 10 years), which were grouped according to disease severity (sclerosis, 1-vessel disease (VD), 2-

Objective perimetry using functional magnetic resonance imaging in patients with visual field loss.

In ophthalmic clinics, subjective perimetry is a standard examination method. However, for certain patients, objective perimetry is useful since it avoids the need for subjective judgments. The purpose of the present study is to demonstrate the feasibility of objective perimetry using functional magnetic resonance imaging (fMRI). fMRI was performed in 8 patients with visual field defects caused by cerebral lesions. The composite stimulus was either the combination of an expanding ring and a clockwise rotating wedge, or a contracting ring and a counter-clockwise rotating wedge. The largest radi

"Resting" CBF in the epileptic baboon: correlation with ketamine dose and interictal epileptic discharges.

BACKGROUND: Photosensitive epileptic (SZ) baboons demonstrate different cerebral blood flow (CBF) activation patterns from asymptomatic controls (CTL) during intermittent light stimulation (ILS). This study compares "resting" CBF between PS and CTL animals, and CBF correlations with ketamine dose and interictal epileptic discharges (IEDs) between PS and CTL animals. METHODS: Continuous intravenous ketamine was administered to eight PS and eight CTL baboons (matched for gender and weight), and maintained at subanesthetic doses (4.8-14.6 mg/kg/hr). Preservation of mouth region processing in two cases of prosopagnosia.

Although most adults are considered experts in face recognition, brain trauma can produce a selective loss in this ability, a condition referred to as prosopagnosia. This study examined the processing strategies of prosopagnosic patients LR and HH using the Face Dimensions Test. In this test, featural and configural information in the upper and lower halves of the face was parametrically varied and sensitivity to these changes measured. We found that relative to age-matched control participants, LR and HH exhibited an impaired ability to discriminate differences in the eye region, but a preser

Autoimmune optic neuritis in the common marmoset monkey: comparison of visual evoked potentials with MRI and histopathology.

PURPOSE: To assess the use of visual evoked potentials (VEPs) for the in vivo detection of impaired visual function in a marmoset model of multiple sclerosis. The sensitivity of the VEP recordings was determined by comparison with magnetic resonance imaging (MRI) and histopathology. METHODS: Baseline VEPs were recorded in six healthy marmoset monkeys in response to light-flash stimulation. Experimental autoimmune encephalomyelitis (EAE) was induced in four of the six monkeys. Clinical scores were assessed daily, and VEPs were recorded every second week. In vivo MRI and subsequent histopatholo

Electroretinographic analyses of Rpe65-mutant rd12 mice: developing an in vivo bioassay for human gene therapy trials of Leber congenital amaurosis.

PURPOSE: Dramatic restoration of retinal function has followed subretinal viral-mediated gene therapy in RPE65-deficient animal models of human Leber congenital amaurosis (LCA) caused by RPE65 mutations. Progress in early-phase clinical trials of RPE65-LCA prompted us to begin development of an in vivo bioassay of clinical grade vector stability for later-phase trials. METHODS: Naturally-occurring Rpe65-mutant rd12 mice (2-4 mo of age) were studied with full-field electroretinograms (ERGs). Flash stimuli (range, -4.1 to 3.6 log scot-cd x s x m(-2)) were used to evoke ERGs in anesthetized, dar

Corticolimbic blood flow during nontraumatic emotional processing in posttraumatic stress disorder.

CONTEXT: Recent brain imaging studies implicate dysfunction of limbic and paralimbic circuitry, including the amygdala and medial prefrontal cortex (MPFC), in the pathogenesis of posttraumatic stress disorder (PTSD) during traumatic recollection and imagery. However, the relationship between activity in these regions and general emotional processing unrelated to traumatic experience has not been fully examined. OBJECTIVE: To investigate activity in the limbic and paralimbic brain regions in PTSD in response to a challenge with emotionally salient generic visual images. DESIGN: Cross-sectiona

Altered resting state networks in mild cognitive impairment and mild Alzheimer's disease: an fMRI study.

Activity and reactivity of the default mode network in the brain was studied using functional magnetic resonance imaging (fMRI) in 28 nondemented individuals with mild cognitive impairment (MCI), 18 patients with mild Alzheimer's disease (AD), and 41 healthy elderly controls (HC). The default mode network was interrogated by means of decreases in brain activity, termed deactivations, during a visual encoding task and during a nonspatial working memory task. Deactivation was found in the default mode network involving the anterior frontal, precuneus, and posterior cingulate cortex. MCI patients

Hormonal cycle modulates arousal circuitry in women using functional magnetic resonance imaging.

Sex-specific behaviors are in part based on hormonal regulation of brain physiology. This functional magnetic resonance imaging (fMRI) study demonstrated significant differences in activation of hypothalamic-pituitary-adrenal (HPA) circuitry in adult women with attenuation during ovulation and increased activation during early follicular phase. Twelve normal premenopausal women were scanned twice during the early follicular menstrual cycle phase compared with late follicular/midcycle, using negative valence/high arousal versus neutral visual stimuli, validated by concomitant electrodermal acti

A novel method for the topographic analysis of neural activity reveals formation and dissolution of 'Dynamic Cell Assemblies'.

The study of synchronous oscillations in neural systems is a very active area of research. However, cognitive function may depend more crucially upon a dynamic alternation between synchronous and desynchronous activity rather than synchronous behaviour per se. The principle aim of this study is to develop and validate a novel method of quantifying this complex process. The method permits a direct mapping of phase synchronous dynamics and desynchronizing bursts in the spatial and temporal domains. Two data sets are analyzed: Numeric data from a model of a sparsely coupled neural cell assembly a

Visual evoked potentials in disproportionately growth-retarded human neonates.

To study brain function in the neonatal period, disproportionately growth-retarded (n = 33) and appropriately grown (n = 21) infants were examined using Doppler flow velocities prenatally and visual evoked potentials postnatally. Visual evoked potentials recordings were made at gestation of 40 and 46 weeks. The group of growth-retarded infants had significantly prolonged latencies to both of the two major peaks (designated P and N), most pronounced for the P peak. This result was observed at both ages investigated and corresponds to a developmental delay of 3 weeks. For individuals, the increa
