[Determination by ultrasound of chorionicity in twin pregnancy].

Twins, specially those of monochorionic and monoamniotic pregnancies, are exposed to many perinatal risks and complications. The objective of this study was to evaluate the usefulness of ultrasonographic determination of the chorionicity of human placental by counting the number of layers in amniotic membranes. Thirty-eight patients with twin pregnancies were studied prospectively. The ultrasonographic evaluation of membranes layers was made only once, between the weeks and 16 and 30 of pregnancy. When two layers were identified, the placentation was determined as monochorionic, and when four …

From venous congestion to placental hypoxia: the underappreciated role of chronic venous disease in impaired placenta development and pregnancy health.

Chronic Venous Disease (CVD) is a common vascular disorder, primarily affecting the lower extremities, with a significantly higher incidence in women. Pregnant women represent a particularly high-risk population for CVD. Early screening and assessment of CVD severity and progression during pregnancy are imperative for preventing Venous Thromboembolism (VTE). Despite its high prevalence, CVD in pregnancy often remains underestimated, frequently being managed by clinicians as a localized and benign condition. However, emerging evidence suggests that CVD may exert broader systemic effects, potent

Exploratory evaluation of equine placental extract supplementation on ageing indicators in geriatric dogs: a single-arm pre-post study.

BACKGROUND: Recent advances in preventive veterinary healthcare, nutritional management, and owner engagement have extended the longevity of domestic dogs. However, this increased lifespan has led to a higher prevalence of age-associated physiological and functional decline. Equine placental extract contains a variety of bioactive constituents, with reported antioxidative and anti-inflammatory properties. Although the use of equine placental extract in veterinary practice is expanding, empirical evidence supporting its effects on ageing-related clinical outcomes in dogs remains limited. In thi

Human Placental Mesenchymal Stem Cell-Derived Exosomes in Wound Healing and Scar Therapy: A Systematic Review and Meta-analysis.

Exosome therapy represents an emerging regenerative medicine approach for optimizing wound healing and reducing scarring. Extensive pre-clinical research on human placental mesenchymal stem cell-derived exosomes (hpMSC-exosomes) demonstrates significant therapeutic potential in aesthetic and reconstructive surgery applications. Despite compelling preclinical evidence supporting the efficacy of exosomes in wound healing, human clinical studies remain scarce. The author's current study includes 738 patients treated with hpMSC-exosomes over a 7-year period across multiple aesthetic and reconstruc

Trajectories of Maternal Placental Growth Factor Levels from Early to Late Pregnancy: Prenatal Factors and Adverse Perinatal Outcomes.

BACKGROUND: Knowledge on longitudinal trajectories of maternal placental growth factor (PlGF) levels, their prenatal factors, and their associations with adverse perinatal outcomes is lacking. We sought to identify PlGF trajectories, examine associations of prenatal factors with these trajectories, and determine whether these trajectories are associated with adverse perinatal outcomes. METHODS: This retrospective cohort study included 438 pregnant women with multiple PlGF measurements. Group-based trajectory modeling identified distinct PlGF trajectories. Logistic and linear regression examin

Placental iron utilisation in fetal growth restriction: alterations in mitochondrial haem synthesis and iron-sulphur cluster assembly pathways.

Fetal growth restriction (FGR) affects ∼10% of pregnancies worldwide and is often associated with placental insufficiency. Iron is essential for maternal haematopoietic adaptations and placental processes such as mitochondrial iron-sulphur (Fe-S) cluster assembly, haem synthesis and erythropoiesis. This study aimed to characterise iron transport and downstream utilisation in FGR. Placental tissues from term uncomplicated (n = 19) and FGR (n = 18) pregnancies were analysed. Maternal iron status was retrospectively assessed from clinical records. Placental mRNA and protein expression of iron-dep

Altered placental expression of neurotransmitter and stress-related molecules in first-episode psychosis during pregnancy: Implications for cytoarchitecture and function.

BACKGROUND: First-episode psychosis during pregnancy (FEP-PW) is a rare but serious condition that intersects maternal mental health and placental-fetal biology. While psychosis is characterized by disruptions in perception and cognition, its impact on placental neuroendocrine signaling and fetal development remains poorly understood. The placenta, as a key mediator of intrauterine environment, expresses receptors involved in neurotransmission and stress response, potentially linking maternal psychopathology to fetal neurodevelopmental risk. OBJECTIVE: This study investigates the expression o

Chronic histiocytic intervillositis of the placenta: clinical outcomes and relationships with circulating placental growth factor and uterine artery Doppler waveforms.

BACKGROUND: Chronic histiocytic intervillositis is a rare placental disorder characterized by maternal macrophage accumulation within the intervillous space of the placenta. Local cytokine release by these immune cells mediates injury to the developing placental villi, resulting in a spectrum of early pregnancy loss, severe fetal growth restriction, or stillbirth. Despite good evidence that chronic histiocytic intervillositis begins in early pregnancy, no effective risk assessment tools exist to predict pregnancy outcomes and, thus, appropriate fetal surveillance. OBJECTIVE: This study aimed

CRISPRi Screening Identifies Essential E. coli Virulence Factors for Placental Barrier Breach in a Maternal-Fetal Infection Model.

Early-onset neonatal sepsis caused by Escherichia coli (E. coli) threatens neonates' lives due to the pathogen's high virulence and multidrug resistance. The mechanisms that enable its placental barrier breach are poorly understood. Using a clinically isolated ST95 ExPEC strain from a neonatal sepsis case, along with a pregnant rat model and an in vitro placental barrier model, we performed CRISPR interference screening. This screen targeted 264 virulence factor genes and identified virulence factors for motility, iron acquisition, hemolysin secretion, and adherence/invasion as critical. We de

Transcriptional Control by FOXO4 Regulates Trophoblast Differentiation and Safeguards Placental Development Through MITD1- and PCBP2-Mediated Redox Homeostasis.

Forkhead box O4 (FOXO4) is highly expressed in the placenta, but its molecular role in trophoblast biology remains unclear. Transcriptomic analysis of human placenta revealed FOXO4 abundance, prompting investigation of its function in trophoblast differentiation and placental development. We found that FOXO4 expression is induced during mouse trophoblast stem (TS) cell differentiation. Gain- and loss-of-function studies demonstrated that FOXO4 promotes differentiation of TS cells towards the trophoblast giant cell (TGC) lineage. Chromatin immunoprecipitation sequencing (ChIP-seq) identified FO

Machine learning identification of maternal inflammatory response and systemic inflammatory response from placental membrane whole slide images.

INTRODUCTION: The placenta forms a critical barrier to infection through pregnancy, labor and, delivery. Acute placental inflammation in the membranes, diagnosed as Maternal Inflammatory Response (MIR), has short-term, and long-term consequences for offspring health. Digital pathology and machine learning can play an important role in understanding placental inflammation. The goal of this work is to develop machine learning models to predict pathologist-classified MIR stages from based on whole slide images (WSI) and establish early benchmarks. METHODS: We used digitized histopathology whole

Co-trimoxazole-induced reproductive toxicity and placental-barrier disruption: impact on cell-cell junctions and ERK signaling pathway.

Co-trimoxazole (CTX), a combination of sulfamethoxazole (SMZ) and trimethoprim (TMP), is used during pregnancies complicated by infections like Urinary Tract Infections (UTI) and Human Immunodeficiency Virus (HIV), despite potential fetal safety concerns. This study examines CTX's effects on the placenta using both in vivo and in vitro models. Pregnant mice received CTX for 4 days during both early and late gestation. On gestational day 19, tissues were collected for analysis. Furthermore, in vitro, human BeWo placental cells were exposed to non-cytotoxic concentrations of CTX for 24 h. Gene e

Decursinol Protects Against Lipopolysaccharide-Induced Placental Inflammation and Trophoblast Dysfunction via Mitochondrial Preservation and NLRP3 Inflammasome Inhibition.

Inflammation-induced placental dysfunction is a major contributor to pregnancy complications. Activation of NF-κB and NLRP3 inflammasome pathways in the placenta is a key driver of this pathology. Decursinol, a natural coumarin derivative from Angelica gigas, possesses anti-inflammatory properties; however, its effect on placental inflammation remains unclear. Therefore, in this study, we investigated the protective effects of decursinol against lipopolysaccharide (LPS)-induced placental inflammation and trophoblast dysfunction and explored the underlying molecular mechanisms. Decursinol signi
