Survival and progression of patients with mature B-cell malignancies, including chronic lymphocytic leukemia and non-Hodgkin's lymphoma, depend on signals from the B -cell receptor, and Bruton's tyrosine kinase(BTK) is a critical signaling kinase in this pathway. Ibrutinib is a novel and selective inhibitor of BTK, inactivating the kinase irreversibly. It has shown exciting results in early clinical trials. In this review, the application and molecular mechanism of Ibrutinib in the treatment of B-cell malignancies are discussed.

Rosuvastatin has been marketed for approximately a decade. In this review we critically discuss available evidence on the benefits and risks from its use. In clinical trials using rosuvastatin, 'lowest is best' was relevant for on-treatment low-density lipoprotein cholesterol levels. Imatinib has been the preferred initial therapy for newly diagnosed chronic myeloid leukemia (CML) patients for the last 10 years. Today, other tyrosine kinase inhibitors (TKIs) are licensed for first-line use. In this paper we analyze the pros and cons of the various alternatives to imatinib and try to give some advice on the management of the newly diagnosed patient.

Although recent studies have suggested that cessation of imatinib (IM) in chronic myeloid leukemia patients can be associated with sustained response, further validation is needed to explore predictive factors.
