Exposure to indoor wood smoke, rather than hypoxemia, is a risk factor for cognitive impairment in COPD patients living at high altitude.

BackgroundIn the general population without COPD, indoor air pollution from biomass is a causal factor of cognitive impairment (CI). CI is also common in patients with tobacco smoke COPD and has been associated with hypoxemia and cardiovascular comorbidities. CI and its risk factors have not been evaluated in COPD patients living at high altitude, who experience greater hypoxemia and frequent exposure to wood smoke.MethodsPatients with COPD residing at high altitude with FEV1/FVC <0,70 and COPD risk factors (wood smoke, Tobacco smoke or combined exposure). Carbohydrate dynamics and transcriptomic changes during floral development in heterodichogamous Ziziphus jujuba.

Flower opening is driven by petal cell elongation, a process that depends on water influx into cells and the maintenance of turgor pressure. Consequently, carbohydrate metabolism is fundamental to flower development, yet its role during floral development in heterodichogamous species remains poorly understood. In this study, we investigated biomass accumulation across the unopened flower bud, male, and female floral phases in the Early and Late morphs of Ziziphus jujuba. We further characterized non-structural carbohydrate dynamics and transcriptomic changes in flowers and corresponding leaf t…

Increased low-molecular-weight mucins in muco-obstructive airway disease limit Staphylococcus aureus growth.

Muco-obstructive airway diseases result in an increase in mucus accumulation and a decrease in mucus clearance. MUC5B is the most abundant s … e. We found that low-molecular-weight (LMW) mucin negatively impacts S. aureus survival and biofilm biomass compared to high-molecular-weight (HMW) mucin. Adding extracellular DNA to SCFM with LMW mucin was not sufficient to restore growth. LMW mucin had a broad negative impact on S. aureus laboratory strains and CF clinical isolates. We next tested other …

Antimicrobial peptide SAAP-148 inhibits Helicobacter pylori and is associated with membrane disruption, biofilm suppression, and reduced cell-associated urease activity.

AIMS: To investigate the in vitro antibacterial activity and related phenotypic effects of the novel antimicrobial peptide SAAP-148 against … th bacterial morphological damage, increased inner and outer membrane permeability, reduced biofilm biomass and biofilm-associated protein content, decreased cell-associated urease activity, and reduced ureA/ureB mRNA expression. In the dedicated MDR model, SAAP-148 also reduced viable counts of one four-drug-resistant H. pylori clinical isolate. CONCLUS …

N-nonyloxypentyl-L-deoxynojirimycin reduces Stenotrophomonas maltophilia virulence in vitro and in Galleria mellonella infection model.

INTRODUCTION: Stenotrophomonas maltophilia is an opportunistic pathogen able to resist multiple classes of antibiotics and form robust biofi … nd to human lung epithelial cells by over 90%, and disrupted preformed biofilms, decreasing biofilm biomass and metabolic activity by approximately 70%. In addition, L-NPDNJ has been shown to enhance macrophage phagocytosis and reduce intracellular bacterial survival, while improving epithelial cell viability. Notably, pretreatment of S. maltophilia cells …

Stigmurin derivatives as potent-biofilm eradicating agents against the major human opportunistic pathogen Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a versatile opportunistic pathogen whose capacity to form biofilms contributes to persistent and difficult-to-trea … re able to disrupt mature biofilms on polystyrene pegs. Stigmurin (24 h exposition) reduced biofilm biomass by nearly 47%, while StigA31 achieved dispersal levels of up to 35%, even at nanomolar concentrations. Under dynamic flow conditions, both peptides strongly reduced biofilm biovolume and thickness within 2 h and altered matrix exopolysaccharide dete …

The Small Noncoding RNA, RsaC, Is Essential for Staphylococcus aureus Virulence.

BACKGROUND: Bacterial small noncoding RNAs (sRNAs) play critical roles in virulence, stress adaptation, and host-pathogen interactions. Transcriptomic analyses during infection can help reveal pathogen-derived sRNAs required for pathogenesis, providing valuable insights for the development of novel therapeutic strategies. However, the low abundance of pathogen biomass within the host tissues poses a significant challenge for such analyses. METHODS: We employed 2-step cell disruption to enrich Staphylococcus aureus cells from infected mouse organs and conducted RNA sequencing (RNA-seq) analysi…

Antibacterial and Immunomodulatory Effects of Withania somnifera Ethanolic Root Extract Against Colistin-Resistant Acinetobacter baumannii in Leukopenic Mice.

Acinetobacter baumannii, particularly colistin-resistant strains, represents a major therapeutic challenge because of multidrug resistance, … e-kill analysis demonstrated bactericidal activity at 250 μg/mL, and WSEE significantly reduced the biomass of preformed biofilms. In cyclophosphamide-induced leukopenic mice, oral administration of WSEE (100 and 200 mg/kg) restored leukocyte counts and significantly reduced pulmonary bacterial burden following infection with both isolates. A Narrative Review of Airway and Gut Microbiota in Chronic Obstructive Pulmonary Disease: Clinical Associations, Methodological Heterogeneity, and Translational Priorities.

Chronic obstructive pulmonary disease (COPD) is a heterogeneous disorder in which exacerbation susceptibility, persistent inflammation, disease progression, and treatment response are not fully explained by spirometry. Culture-independent studies have associated airway and gut microbial features with clinically relevant COPD phenotypes, but findings are highly sensitive to sampling site, low biomass, contamination control, sequencing depth and platform, bioinformatic workflow, microbial-load quantification, medication exposure, disease state, and host or environmental confounding. Methodologic…

The intrauterine microbiome-neurodevelopment axis: decoding the prenatal microbial imprint on lifelong mental health.

INTRODUCTION: The traditional view of a sterile intrauterine environment has been challenged by sequencing studies detecting low-biomass microbial DNA in placenta, amniotic fluid, and fetal tissues. These findings suggest that maternal microbiota-derived signals may contribute to fetal brain development and influence long-term neuropsychiatric outcomes. CONTENT: This narrative review synthesizes evidence from over 90 preclinical and clinical studies examining maternal microbiota-fetal brain interactions. Maternal immune activation - characterized by elevated cytokines such as interleukin (IL)…
