Overcoming Barriers in Porcine SCNT: A Comprehensive Review of Developmental Challenges and Innovations.

Porcine somatic cell nuclear transfer (SCNT) demonstrates significant potential for application in biomedical research, agricultural breeding, and xenotransplantation. However, compared to in vivo fertilized embryos, SCNT embryos exhibit suboptimal developmental capacity, higher rates of abortion, and neonatal abnormalities, which collectively hinder the large-scale application of this technology. Such developmental anomalies arise from a complex interplay of interconnected factors, primarily incomplete epigenetic reprogramming (the core driver), telomere shortening, mitochondrial dysfunction,

A reconstruction of the mammalian secretory pathway identifies mechanisms regulating antibody production.

The secretory pathway processes >30% of mammalian proteins, orchestrating their synthesis, modification, trafficking, and quality control across multiple organelles via coordinated interactions, making its regulation difficult to decipher. To advance such research, we present secRecon, a reconstruction of the mammalian secretory pathway, comprising 1,127 manually curated genes organized within 77 secretory process terms, annotated with functional roles, subcellular localization, protein interactions, and complexes. Applying secRecon to omics data revealed distinct secretory topologies in antib

Yeast Prions: Discovery, Nature, Cellular Manipulation and Implication.

Prion is simply an infectious protein, and it occurs spontaneously without any well-defined reason. Once prions occurred, they mostly propagated in biophysically very stable amyloid form with self-templating mechanism. Human and mammalian prion results in untreatable and even fatal brain disorders, thus study of prion and its pathology extremely difficult. However, since the first discovery of two prions in model microorganism Saccharomyces cerevisiae, studies about prion biology, host physiology affected by prion or prion disease have facilitated. Moreover, many attempts to understand human a

Cold atmospheric plasma as a novel "drug" for cancer therapy.

Plasma with low temperature generated at atmospheric pressure is known as cold atmospheric plasma (CAP). Owing to its unique characteristics and biological effects of inducing tumor cell death without endangering the surrounding healthy tissues, CAP is regarded as an emerging potent anticancer strategy and has been extensively investigated in preclinical research. In this review, we define the reactive species that play a major role in CAP as a novel "drug" (termed as plasma drug) used in cancer therapy. Various methods of plasma drug use in tumor treatment were summarized, mainly including pl

Glaucoma animal models in rabbits: State of the art and perspectives-A review.

Glaucoma, a visual thief, is characterized by elevated intraocular pressure (IOP) and the loss of retinal ganglion cells (RGCs). Selecting suitable animals for preclinical models is of great significance in research on the prevention, early screening, and effective treatments of glaucoma. Rabbit eyeballs possess similar vascularity and aqueous humor outflow pathways to those of humans. Thus, they are among the earliest in vivo models used in glaucoma research. Over the years, rabbit models have made substantial contributions to understanding glaucomatous pathophysiology, surgical adaptations,

Cryptococcosis, tuberculosis, and a kidney cancer fail to fit the atherosclerosis paradigm for foam cell lipid content.

Foam cells are dysfunctional, lipid-laden macrophages associated with chronic inflammation of diverse origin. The long-standing paradigm that foam cells are cholesterol-laden derives from atherosclerosis research. We previously showed that, in tuberculosis, foam cells surprisingly accumulate triglycerides. Here, we utilized bacterial (Mycobacterium tuberculosis), fungal (Cryptococcus neoformans), and human papillary renal cell carcinoma (pRCC) models to address the need for a new explanation of foam cell biogenesis. We applied mass spectrometry-based imaging to assess the spatial distribution

The therapeutic implications of all-in-one AAV-delivered epigenome-editing platform in neurodegenerative disorders.

Safely and efficiently controlling gene expression is a long-standing goal of biomedical research, and CRISPR/Cas system can be harnessed to create powerful tools for epigenetic editing. Adeno-associated-viruses (AAVs) represent the delivery vehicle of choice for therapeutic platform. However, their small packaging capacity isn't suitable for large constructs including most CRISPR/dCas9-effector vectors. Thus, AAV-based CRISPR/Cas systems have been delivered via two separate viral vectors. Here we develop a compact CRISPR/dCas9-based repressor system packaged in AAV as a single optimized vecto

Exploring the Effects of Various Capping Agents on Zinc Sulfide Quantum Dot Characteristics and In-vitro Fate.

The choice of capping agents used during the synthesis process of quantum dots (QDs) can significantly influence their fate and fundamental properties. Hence, choosing an appropriate capping agent is a critical step in both synthesis and biomedical application of QDs. In this research, ZnS QDs were synthesized via chemical precipitation process and three commonly employed capping agents, namely mercaptoethanol (ME), mercaptoacetic acid (MAA), and cysteamine (CA), were used to stabilize the QDs. This study was aimed to examine how these capping agents impact the physicochemical and optical char

Mini-Review Regarding the Applicability of Genome Editing Techniques Developed for Studying Infertility.

Infertility is a highly debated topic today. It has been long hypothesized that infertility has an idiopathic cause, but recent studies demonstrated the existence of a genetic substrate. Fortunately, the methods of editing the human genome proven to be revolutionary. Following research conducted, we identified a total of 21 relevant studies; 14 were performed on mice, 5 on zebrafish and 2 on rats. We concluded that over forty-four genes in total are dispensable for fertility in both sexes without affecting host homeostasis. However, there are genes whose loss-of-function induces moderate to se

Mirror-Image Oligonucleotides: History and Emerging Applications.

As chiral molecules, naturally occurring d-oligonucleotides have enantiomers, l-DNA and l-RNA, which are comprised of l-(deoxy)ribose sugars. These mirror-image oligonucleotides have the same physical and chemical properties as that of their native d-counterparts, yet are highly orthogonal to the stereospecific environment of biology. Consequently, l-oligonucleotides are resistant to nuclease degradation and many of the off-target interactions that plague traditional d-oligonucleotide-based technologies; thus making them ideal for biomedical applications. Despite a flurry of interest during th

Infestation of research zebra finch colony with 2 novel mite species.

A zebra finch (Taeniopygia guttata) housed in a neuroscience laboratory was observed to have numerous feather mites. Subsequently, similar mites were found on other birds in the animal facility and research space. The most abundant mite was a novel, undescribed species in the genus Neocheyletiella. Whereas known Neocheyletiella mites have previously been characterized as skin parasites of various birds worldwide, the species on the zebra finches is unique because it lives and builds nests in the feathers. Infrequent specimens of a 'true' feather mite, a new species of Megninialges, were presen

Quantitative molecular assessment of chimerism across tissues in marmosets and tamarins.

BACKGROUND: Marmosets are playing an increasingly large and important role in biomedical research. They share genetic, anatomical, and physiological similarities with humans and other primate model species, but their smaller sizes, reproductive efficiency, and amenability to genetic manipulation offer an added practicality. While their unique biology can be exploited to provide insights into disease and function, it is also important that researchers are aware of the differences that exist between marmosets and other species. The New World monkey family Callitrichidae, containing both marmoset

Strategies for modelling the intestinal mucus interface: Focus on drug transport and host-microbiota interactions.

The intestinal mucus barrier is a complex, dynamic hydrogel that plays a central role in host-microbiota interactions, nutrient transport, and protection against pathogens. Understanding its physicochemical properties has become essential for developing predictive in vitro and in silico models for drug delivery, microbiome research, and disease modelling. Over the past decade, advances in polymer chemistry, glycoscience, and microfluidics have enabled the development of increasingly sophisticated mucus models that mimic structural, rheological, and biochemical features of native intestinal muc
