Management of Bone Health in Breast Cancer Survivors.

PURPOSE: Breast cancer survivors (BCS) face a significantly higher risk of treatment-induced bone loss compared with the general population. Despite clinical guidelines, baseline bone mineral density (BMD) screening and indicated pharmacologic interventions remain consistently underutilized in this population. DESIGN: This review synthesizes current evidence regarding the pathophysiology and incidence of bone loss in BCS. We evaluate the impact of cytotoxic and endocrine therapies on skeletal health and summarize contemporary guidelines for the screening, prevention, and management of cancer …

DenseUNet for Breast Cancer Segmentation in Histopathological Images.

BACKGROUND: Breast cancer remains one of the leading causes of mortality among women worldwide, highlighting the urgent need for accurate and efficient diagnostic tools. Histopathological image analysis plays a critical role in diagnosis by enabling cellularlevel tissue examination. However, manual assessment is often timeconsuming, subjective, and prone to variability, driving the need for automated solutions. METHODS: This study proposes DenseUNet169, a deep learning framework that integrates established DenseNet and UNet for systematic evaluation in breast cancer segmentation. The model im…

Germline predisposition and the pattern of breast cancer metastasis.

More than three decades have passed since the identification of BRCA1 as the first breast cancer susceptibility gene (BCSG). Since then, substantial progress has been made in pinpointing BCSGs and estimating the associated risk of developing BC. Simultaneously, new challenges and open questions have also emerged, such as the potential associations between BCSGs and metastasis in BC patients. In this review, we discuss available data from clinical and experimental studies assessing the association between germline pathogenic variants (GPVs) in BCSGs and metastasis pattern. To help guide future …

Cardiotoxicity in breast cancer patients treated with chemoradiotherapy.

With national breast cancer screening programs leading to earlier diagnosis and improved treatments, more women are achieving long-term remission. However, this has resulted in rising rates of cardiotoxicity in breast cancer survivors consequent to their cancer therapies. Anthracyclines, a cornerstone treatment in breast cancer, carry a significant risk of short- and long-term cardiotoxicity through inhibition of topoisomerase II, preventing DNA repair and increasing oxidative stress. This most commonly manifests in the form of left ventricular (LV) systolic dysfunction or heart failure, and i…

Lessons Learned From 26 Adenomyoepitheliomas of the Breast.

Adenomyoepithelioma (AME) of the breast is a biphasic neoplasm prone to misclassification. To enhance diagnostic awareness, we evaluated 26 AMEs stratified across a spectrum of benign (54%), atypical (27%), special salivary gland-like (12%), and malignant (8%) subtypes. Size and borders stratified the spectrum: benign, atypical, and special subtypes (median: 12 to 18 mm) exhibited lobulated/multilobulated contours, whereas malignant AMEs were larger (median: 27.5 mm) and infiltrative. Cytologic atypia and mitoses were predominant in atypical and malignant subtypes. Overall, 35% of cases were r…

Human papilloma virus as causal in breast cancer.

The aim of this review is to consider the role of high-risk human papillomaviruses (HPVs) in breast cancer. HPVs have established causal roles in cancers of the cervix, anus, penis, vagina, vulva, and oropharyngeal cancers. High-risk HPVs have been consistently detected in breast cancers in 23 countries. HPVs have been identified in 32.7% of breast cancers as compared to 7.6% of benign and normal breast controls. HPV-positive benign breast lesions can progress to HPV-positive malignancies, suggesting an early role of HPVs in carcinogenesis. HPV biological activity has been demonstrated by the …

Advances in the Research and Development of Breast Cancer Organoids.

Breast cancer ranks first in global cancer incidence. Due to its high heterogeneity, cancer cells often develop drug resistance during metastasis, leading to therapeutic challenges and poor prognosis. Consequently, breast cancer organoid models have emerged, which can effectively recapitulate the tumor microenvironment and serve as important tools for investigating the mechanisms underlying breast cancer initiation and progression. Breast cancer organoids exhibit interactions between cells and the extracellular matrix (ECM) while retaining the heterogeneity of the original tumor cells. Owing t…

Claudin 18 is not a biomarker for breast mucinous tumor.

BACKGROUND: Breast mucinous cystadenocarcinoma (BMCA) is an exceptionally rare malignancy that may resemble mucinous neoplasms of the pancreas, ovary, lung and colorectum. Claudin 18 (CLDN18), particularly its gastric-associated isoform CLDN18.2, is an emerging therapeutic target; however, CLDN18 immunoreactivity has not been systematically evaluated in BMCA. We investigated whether CLDN18 could distinguish BMCA from breast mucinous carcinoma (BMC) and selected extramammary mucinous mimics. METHODS: Immunohistochemical (IHC) analysis for CLDN18, CK7 and CK20 was performed on 42 mucinous neopl…

The effectiveness of sulforaphane in radiosensitizing breast cancer cells.

INTRODUCTION: Radiotherapy is a vital therapeutic option in the treatment of breast cancer nowadays. However, a major obstacle to the full effectiveness of radiation therapy is still the radioresistance of cancer cells. Various studies have proven sulforaphane's (SFN) beneficial effects against cancer and its possible utilization as a radiosensitizer in radiotherapy. This study aimed to investigate whether SFN has a radiosensitizing effect on breast cancer cells. MATERIAL AND METHODS: The anticancer efficiency of SFN and radiosensitizing effect in MCF-7 and MDA-MB-231 cell lines were assessed…

Investigating the Anna Karenina principle of the breast microbiome.

The relationship between the microbiome and disease has long been a central focus of research in human microbiome. Inspired by Leo Tolstoy's dictum, the Anna Karenina Principle (AKP) offers a framework for understanding the complex dynamics of microbial communities in response to perturbations, suggesting that dysbiotic individuals exhibit greater variability/heterogeneity in their microbiome compared to healthy counterparts. While some studies have proved the alignment of microbiome responses to disease with the AKP effect, it remains uncertain whether the human breast microbiome responds sim…

[Brain metastases in breast cancer: Diagnosis and management].

Breast cancer is the second most common cause of brain metastases, after lung cancer. The risk of developing brain metastases varies according to the molecular subtype of breast cancer, with a higher incidence for triple-negative or HER2-positive cancers. The discovery of brain metastases, whether synchronous or metachronous, is a turning point in oncology management, and requires discussion at a neuro-oncology multidisciplinary consultation meeting to assess the value and modalities of local treatment by surgery and/or radiotherapy (stereotactic or total brain). Systemic treatments also play …

Imaging Characteristics and Diagnostic Implications of Breast Amyloidoma.

Breast amyloidoma is an extremely rare and potentially underdiagnosed entity with underreported diagnostic implications. Breast amyloidomas typically present as painless masses with varying imaging characteristics ranging from nonspecific asymmetries to mammary carcinoma mimics. Calcifications are visualized in many described cases in the literature with a characteristic histopathologic appearance following Congo red staining under polarized light. While breast amyloidomas are benign lesions, there are proposed associations with systemic autoimmune and hematolymphoid disorders in the literatur…

Recent Advances in Malignant Phyllodes Tumors of the Breast.

Phyllodes tumors (PTs) represent an uncommon category of fibroepithelial neoplasms found in the breast tissue and are classified as benign, borderline, or malignant based on their histopathological features. Histological assessment remains the cornerstone of a PT diagnosis. However, recent genomic advancements have revealed the biological mechanisms underlying PTs, particularly malignant phyllodes tumors (MPTs). This comprehensive review integrates current genomic and molecular investigations that have elucidated the distinguishing features of PTs. Mutations in the MED12 gene represent early t…

Consumption of ultraprocessed foods and breast density in adolescence.

Breast density (BD) is one of the major risk factors for breast cancer (BC) and has the potential to be used as an early marker. Puberty is a critical period for carcinogenesis, and identifying events that modify BD during this stage could aid in understanding future BC risk. The consumption of ultra-processed foods (UPFs) has been associated with BC; however, evidence of their effect on BD, particularly in adolescents, is lacking. To assess the association between the intake of UPFs during the peripubertal period and BD measured at 4 years after menarche onset, absolute fibroglandular volume …

Mapping the BCSC-1 interactome in breast cancer.

BCSC-1 (breast cancer suppressor candidate-1), the official gene symbol VWA5A (von Willebrand factor A domain containing 5 A), is downregulated in a variety of cancer types including breast cancer. The role of BCSC-1 in tumorigenesis and the underlying mechanisms are poorly understood. One of the ways to predict a spectrum of functions of an unknown protein is to identify the protein-protein interaction network involving it. Flag-pLV-Neo-BCSC1 or the empty vector were stably transfected into breast cancer cells MCF-7. The levels of BCSC1 expression were identified by Western blotting. Co-immun…

Advances in Neuroimaging of Breast Cancer Pain: An Overview.

Breast cancer is currently the most common malignant tumor, primarily affecting women, and it frequently leads to chronic pain that significantly impairs physical and mental health. Neuroimaging studies have demonstrated that breast cancer-related pain is associated with specific brain alterations, including changes in activation, connectivity, and structure of pain-processing regions. This review synthesizes findings on functional and structural brain changes related to chronic pain in breast cancer and compares them with non-cancer chronic pain patterns. By integrating recent evidence, it pr…
