T-2 toxin induces chondrocyte extracellular matrix degradation by regulating the METTL3-mediated Ctsk m6A modification.

T-2 toxin is a major cause of Kashin-Beck disease (KBD), which is characterised by cartilage damage. N6-adenosine-methyltransferase-like 3 (METTL3) regulates cartilage injury; however, its role in T-2 toxin-induced cartilage injury remains elusive. Herein, we investigated the involvement of METTL3-mediated m6A modification in T-2 toxin-induced cartilage damage. METTL3-mediated m6A methylation levels were correlated with cartilage extracellular matrix (ECM) degradation, which was exacerbated following METTL3 silencing. Cathepsin K (Ctsk) was identified as a downstream target of METTL3 using m6A…

Autoreactivity against matrilin-1 in a patient with relapsing polychondritis.

Relapsing polychondritis (RP) is a rare inflammatory disease of cartilage. Chondritis of the auricular, nasal, and tracheal cartilages predominates in this disease, suggesting a response to a tissue-specific antigen. One potential antigen is matrilin-1, a cartilage matrix protein found uniquely in the tracheal, auricular, and nasal cartilage of adults. We describe herein a patient with RP who had both a humoral and a cellular immune response directed toward the cartilage matrix protein matrilin-1.

The synovial microenvironment suppresses chondrocyte hypertrophy and promotes articular chondrocyte differentiation.

During the development of the appendicular skeleton, the cartilaginous templates undergo hypertrophic differentiation and remodels into bone, except for the cartilage most adjacent to joint cavities where hypertrophic differentiation and endochondral bone formation are prevented, and chondrocytes instead form articular cartilage. The mechanisms that prevent hypertrophic differentiation and endochondral bone formation of the articular cartilage have not been elucidated. To explore the role of the synovial microenvironment in chondrocyte differentiation, osteochondral allografts consisting of ar…

Alterations in sclerostin protein in lesions of equine osteochondrosis.

Osteochondrosis (OC) is a common and clinically important joint disease that occurs in many species, including humans, pigs, chickens and horses. It has been described as a focal failure of endochondral ossification (EO), but no cellular/molecular mechanisms are fully described that explain the cause of this condition. Recently a Wnt signalling inhibitor, sclerostin, has been described in osteoarthritic cartilage, where it has been proposed to protect damaged cartilage from degradation. Cartilage degradation is a key event in EO, thus, abnormalities of sclerostin in growth cartilage could, pot…

Effects of aging and exercise training on the histological and mechanical properties of articular structures in knee joints of male rat.

The impact of aging on joints can have a profound effect on an individual's functioning. Our objectives were to assess the histological and mechanical properties of the knee joint capsule and articular cartilage with aging, and to examine the effects of exercise on age-related changes in the knee joint. 2-year-old Wistar rats were divided into a sedentary control group and an exercise-trained group. 10-week-old animals were used to investigate the changes with aging. The joint capsule and cartilage were evaluated with histological, histomorphometric, immunohistochemical, and mechanical analyse…

[Experimental study of chondromalacia patellae (author's transl)].

There are many theories concerning the etiology of chondromalacia patellae, as well as many points which have not been fully investigated yet. Using rabbits experimentally, shortening and elongation of the 3 mm patellar ligament were performed, producing the changes of contact surface and pressure on the patellar cartilage, and the patellar cartilage and the subchondral bone were studied sequentially. In the group of shortening, after the course of 16 weeks, no particular changes were recognized in the cartilage and the subchondral bone, compared with the control. In the group with elongation,…

Chondrocyte culture and assay.

Chondrocytes constitute the sole cell type found within cartilage, and control the formation and composition of cartilage. Cellular, biochemical and pharmacological studies of arthritis and other cartilage disorders have increasingly focused on chondrocyte function. Three methods are presented in this unit for culturing chondrocytes, and two assays are described that characterize proteoglycan synthesis, a key measure of chondrocyte function.Chondrocytes constitute the sole cell type found within cartilage, and control the formation and composition of cartilage.

Biomimetic design and fabrication of multilayered osteochondral scaffolds by low-temperature deposition manufacturing and thermal-induced phase-separation techniques.

Integrative osteochondral repair is a useful strategy for cartilage-defect repair. To mimic the microenvironment, it is necessary that scaffolds effectively mimic the extracellular matrix of natural cartilage and subchondral bone. In this study, biomimetic osteochondral scaffolds containing an oriented cartilage layer, a compact layer, and a three-dimensional (3D)-printed core-sheath structured-bone layer were developed. The oriented cartilage layer was designed to mimic the structural and material characteristics of native cartilage tissue and was fabricated with cartilage matrix-chitosan mat…

Differentiation of the osteochondrogenic cells of the periosteum in chondrotrophic environment.

The behaviour of the cells in free periosteal graft was studied in growing rabbits in three chondrotrophic recipient milieus: costal cartilage, ear cartilage and synovial fluid of the knee joint. The periosteal grafts first formed cartilage, which was then quite rapidly transformed into bone on the costal cartilage, quite slowly and in smaller amounts in the ear cartilage, whereas no bone was found in the cartilaginous loose bodies formed in the knee joint. In bone formation vascularization plays a major role, but other factors are also involved.

Morphologic aspects of the injured nasal septum in children.

Biopsies from the septal cartilage in children have been examined histologically to study the influence of trauma, surgery, septal abscess, and transplantation on the growing cartilage. Loss of cartilage, complete, but mostly incomplete regeneration of the defects are the main reactions. A regenerative potential mainly arising from the perichondrium plays an important factor for restoring the septal structure, often in a deforming and excessive way. Even preserved cartilage implanted immediately into the septal abscess, may be transformed into autogenous cartilage if the perichondrium is prese…

Liposomic lubricants suppress acute inflammatory gene regulation in the joint in vivo.

Osteoarthritis (OA) is a widespread, debilitating joint disease associated with articular cartilage degradation. It is driven via mechano-inflammatory pathways, whereby catabolic genes in the cartilage-embedded chondrocytes are presumed up-regulated due to increased shear stress arising from friction at the cartilage surface as joints articulate. The enhanced expression of these cartilage-degrading and inflammatory genes leads to tissue degeneration. However, the nature of the stress, and how the cells within the joint respond to it, are poorly understood. Here we show, in a proof of concept s…
