Mucopolysaccharidosis I (MPS I) is a lysosomal storage disease caused by deficiency of alpha-L-iduronidase. In addition to the well-known human forms (Hurler, Hurler/Scheie, and Scheie syndromes), there exists a canine model of the disease. By using previously described canine cDNA encoding alpha-L-iduronidase as a probe, the canine IDUA gene has been cloned and characterized. It contains 14 exons spread over 13 kb. An unusual GC dinucleotide was found at the donor splice site of intron 11. A transcriptional start site was identified by primer extension 177 bp upstream of the initiator AUG cod

Familial neurohypophyseal diabetes insipidus in humans is a rare disease transmitted as an autosomal dominant trait. Affected individuals have very low or undetectable levels of circulating vasopressin and suffer from polydipsia and polyuria. An obvious candidate gene for the disease is the vasopressin-neurophysin (AVP-NP) precursor gene on human chromosome 20. The 2 kb gene with three exons encodes a composite precursor protein consisting of the neuropeptide vasopressin and two associated proteins, neurophysin and a glycopeptide. Cloning and nucleotide sequence analysis of both alleles of the

The persistent Müllerian duct syndrome (PMDS) is a rare form of male pseudohermaphroditism, characterized by the persistence of Müllerian derivatives in otherwise normal males. Two mutations, present in the homozygous state, have been previously described in such patients. The present observation is the first example of compound heterozygosity in this condition. DNA was obtained from a 3-month-old patient with PMDS, in whom no serum anti-Müllerian hormone (AMH) could be detected by enzyme-linked immunosorbent assay. Sequencing of cloned polymerase chain reaction amplified fragments of the AMH

The t(1;19) chromosomal translocation in acute lymphoblastic pre-B cell leukemias involves the gene E2A for helix-loop-helix (HLH) proteins E12 and E47, ubiquitous transcriptional proteins implicated in the regulation of various lymphoid and nonlymphoid genes. To characterize the molecular features of the t(1;19)(q23;p13) translocation, we molecularly cloned breakpoint DNA from t(1;19)-carrying pre-B cell leukemias. In all cases, breakpoints on chromosome 19 occurred within 2 kb of each other in a single intron of the E2A gene. This clustered arrangement resulted in specific truncation of the

A full-length placental alkaline phosphatase (PLAP) cDNA was used to identify and clone the PLAP-like Nagao isozyme gene from human genomic libraries. The entire nucleotide sequence of the gene reveals the existence of 11 exons interrupted by 10 small introns (76-427 base pairs). Putative regulatory sequences have been identified in the promoter regions as well as dispersed in the introns. The deduced amino acid sequence of the Nagao isozyme indicates that the mature molecule is composed of 513 amino acids, of which 12 residues are different from the sequence of PLAP (98% homology). A sequence

The Burkitt lymphoma cell line KK124, which contains a reciprocal t(8;22) translocation, was shown to have rearranged in a region 3' to the c-myc proto-oncogene on chromosome 8 and 5' to the lambda constant region on chromosome 22. The breakpoint was cloned and sequenced, revealing that c-myc and a portion of its 3' region abutted a complete lambda variable gene that had undergone V-J recombination. Since this cell line expresses kappa light chain, this lambda rearrangement violates the previously proposed hierarchy of immunoglobulin gene rearrangement. A novel duplication of normal chromosome

While third-generation EGFR tyrosine kinase inhibitors (TKIs) improve outcomes in EGFR-mutant NSCLC, optimal selection between these agents remains uncertain. This real-world study compares the efficacy and safety of aumolertinib with those of osimertinib in the first-line treatment setting. We retrospectively analyzed 109 EGFR-mutant (exon 19del/L858R) NSCLC patients treated with first-line aumolertinib (n = 53) or osimertinib (n = 56) across four tertiary hospitals (2016-2023). The primary end points included progression-free survival (PFS) and safety; secondary end points encompassed overal

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) non-structural protein 14 (Nsp14) possesses an N-terminal exonuclease (ExoN) domain that provides a proofreading function for the viral RNA-dependent RNA polymerase and a C-terminal N7-methyltransferase (N7-MTase) domain that methylates viral mRNA caps. Nsp14 also modulates host functions. This includes the activation of NF-κB and downregulation of interferon alpha/beta receptor 1 (IFNAR1). Here, we demonstrate that Nsp14 exerts broader effects, activating not only NF-κB responses but also extracellular-signal-regulated kinase (ERK),

MN1::BEND2 fusion is well established in CNS tumors but exceptionally rare in extracranial mesenchymal neoplasms. We report a subcutaneous upper-back tumor in a 41-year-old woman, composed of bland round-to-epithelioid cells with branching capillary vasculature and focal infiltrative growth. The tumor recurred 42 months after excision. Targeted RNA sequencing identified an in-frame MN1 exon 1::BEND2 exon 9 fusion, supported by Sanger sequencing and MN1 break-apart FISH; Nuclear BEND2 immunoreactivity was an ancillary finding. Unlike two previously reported extracranial MN1::BEND2-harboring tum

INTRODUCTION: Osimertinib is the standard of care in first-line therapy for advanced EGFR-mutant NSCLC, but resistance invariably develops through EGFR-dependent or independent mechanisms.
CASE DESCRIPTION: Here, we report the case of a stage IVA adenocarcinoma patient with several metastases harboring an EGFR exon 19 deletion, which progressed locally after five years on osimertinib. Sequencing of the re-biopsy revealed the emergence of an acquired exon 20 mutation, p.(Cys797Ala), not previously characterized and absent from variant databases. Immunohistochemical analysis (IHC) showed phosph

Effective precision oncology demands integration of pharmacokinetics/pharmacodynamics (PK/PD) profiling with tumor-specific genomic features. Here, we present a personalized treatment model using a patient-derived Networking Organoid Culture System (NOCS) composed of intestinal, liver, and kidney organoids differentiated from induced pluripotent stem cells (iPSCs) of an NF1-mutant breast cancer patient. This multi-organoid system enabled individualized assessment of drug absorption, distribution, metabolism, and excretion. Integrative genomic and pathway analyses uncovered therapeutic vulnerab

Genes affecting DNA methylation (DNAme) are frequently comutated with splicing factors in acute myeloid leukemia (AML) and associate with more aggressive phenotypes. To elucidate the underlying molecular mechanisms, we deeply profiled wild-type and IDH2R140Q/SRSF2P95 single- or double-mutant AMLs. We find a unique set of mis-spliced genes and differentially methylated CpGs in double mutants. Mis-spliced exons are enriched in CCNG splicing enhancers and in the corresponding DNAme changes. Using a machine learning model, we can accurately predict exon inclusion levels from proximal CpGs. These C

Pulmonary Alveolar Microlithiasis (PAM) is a rare hereditary lung disorder characterized by the intra-alveolar deposition of calcium phosphate microliths. It is primarily familial and follows an autosomal recessive inheritance pattern, with no significant gender disparity in incidence. In its early stages, PAM is often asymptomatic, and most cases are detected incidentally through abnormal imaging findings during routine health examinations. We report a case of a male patient in his mid-50 s with a 10-year history of exertional dyspnea and cough unresponsive to conventional therapy. Initially

A cardinal sign of myotonic dystrophy type 1 (DM1) is myotonia, slow muscle relaxation after voluntary contraction. Myotonia results from mis-regulated splicing of chloride channel 1 (ClC-1), leading to loss of channel function and runs of involuntary action potentials in muscle fibers. Preceding the onset of weakness, myotonia is often the first symptom of DM1, and thus this raises the possibility that muscle hyperexcitability contributes to the subsequent weakness and myopathy. Here, we show that genomic deletion of ClC-1 exon 7a (E7a), a cryptic exon abnormally regulated in DM1, completely

Inherited retinal degenerations (IRDs) are a group of clinically and genetically heterogeneous blinding disorders. In this study, we describe five families clearly or which were presumed to be diagnosed with autosomal recessive non-syndromic IRD and one with mild syndromic IRD, in which affected probands carried rare bi-allelic variants in SCLT1, a gene previously associated with multiple autosomal recessive ciliopathies. Eight of the ten variants identified were novel; five variants affected splicing, including the known missense p.(Lys544Arg), detected in compound heterozygosity in three Eas

Mutations in the kinase domain of the epidermal growth factor receptor (EGFR) are a frequent cause of non-small cell lung cancer (NSCLC). Osimertinib, a third-generation tyrosine kinase inhibitor (TKI) that is selective for mutant EGFR, is standard of care for patients with the classical exon 19 deletion variants and the L858R point mutation. Although osimertinib and a recently developed therapy that combines TKI lazertinib with the antibody amivantamab improve outcomes for these patients, resistance limits the durability of response to these agents. Additionally, patients with the L858R mutat

CD45 is the most abundant glycoprotein on the surface of all nucleated hematopoietic-lineage cells, comprising multiple isoforms generated by alternative splicing of three exons ("A," "B," and "C"), which are exquisitely restricted across hematopoietic development. Despite the ubiquitous expression of CD45 on hematopoietic cells, its function(s) remain rather obscure. Here, we report that discrete CD45 isoforms expressed uniquely by immature human hematopoietic cells are distinguished as functional glycoforms ("isoglycoforms") that bind E-selectin. Moreover, our studies indicate that "CD45RA,"

Lung adenocarcinoma (LUAD) progression involves alterations in oncogenes and tumor suppressor genes, collectively shaping tumorigenic landscape. However, the precise interactions within this landscape remain inadequately understood. Here, we present a functional characterization of a novel long non-coding RNA (lncRNA), SPAT (splice associated transcript). SPAT is downregulated in LUAD and its expression positively correlates with favorable prognosis. In vitro and in vivo experiments demonstrated that SPAT inhibits the migration of LUAD cells. This inhibitory effect is mediated by SPAT's intera
