Hypoxia Increases Connexin46 and Connexin43 Levels in KNS-42 Glioblastoma Cells.

Glioblastoma multiforme is a devastating brain tumor that frequently progresses or recurs despite therapy. We used the glioblastoma-derived cell line, KNS-42, to study the response of the gap junction proteins, connexin46 and connexin43, to chemotherapeutic agents and to prolonged hypoxia to mimic conditions within the tumor microenvironment. Under standard culture conditions, KNS-42 cells have high levels of connexin43 and very low levels of connexin46. The cells that survived temozolomide treatment had increased connexin46 levels and decreased connexin43 levels. In contrast, prolonged hypoxi

LncRNA-HLX-2-7/HLX axis-dependent metabolic reprogramming drives cancer progression in group 3 medulloblastoma.

Group 3 medulloblastoma (G3 MB) is an aggressive pediatric brain tumor with limited treatment options, necessitating the development of novel therapeutic strategies. The long noncoding RNA (lncRNA) lnc-HLX-2-7 and its host-coding transcription factor, HLX, are highly expressed in G3 MB and act as oncogenic drivers. However, their role in regulating metabolism in G3 MB remains poorly understood. In this study, we investigate the metabolic functions of lnc-HLX-2-7 in D425-Med and MED211 cells and uncover its role in regulating oxidative phosphorylation (OXPHOS) and ATP production through the TCA

A Co-opted Developmental Gene Regulatory Program in Endothelial Progenitors Promotes Tumor Angiogenic Phenotypes.

UNLABELLED: The transition of endothelial cells (EC) to tumor-associated ECs (TEC) is a critical process in tumor progression. The underlying mechanisms driving this angiogenic state could represent potential therapeutic targets. In this study, we revealed that a co-opted gene regulatory program from EC progenitors promotes the angiogenic phenotypes of TECs. Specifically, the TEC-specific gene regulatory program, derived from large-scale single-cell transcriptomic and epigenomic atlases across multiple solid tumors, was projected onto multiomic maps of human embryonic stem cell-derived EC diff

Targeting the Akt-EphA2 axis and cell-cell adhesion enhances anoikis sensitivity in cancer cells.

Rigidity sensing enables cells to respond to extracellular matrix stiffness and governs survival and apoptotic decisions. Tropomyosin 2.1 (Tpm2.1), a key actin-binding protein, is frequently downregulated in cancer. While re-expressing Tpm2.1 restores rigidity sensing and limits anchorage-independent growth, its ability to fully trigger anoikis, apoptosis induced by loss of matrix attachment, remains unclear. Here, we show that although Tpm2.1 overexpression reduces Akt activation and suppresses survival signaling in suspended breast cancer cells, most cells remain viable without matrix attach

Time-Dependent Loss of miR-548c-3p and Activation of E2F3/FOXM1 in Breast Cancer: In Vitro and TCGA-Based Evidence for a Post-Transcriptional Mechanism.

MicroRNAs are key post-transcriptional regulators in breast cancer, but their time-dependent dynamics and downstream oncogenic effects are not fully understood. miR-548c-3p has been proposed as a tumor suppressor, yet its temporal behavior and impact on cell cycle drivers remain unclear. This study investigated the time-dependent expression of miR-548c-3p and its post-transcriptional regulation of E2F3 and FOXM1 in MCF-7 breast cancer cells. Cells were analyzed at multiple time points (2-72 h) by quantitative real-time PCR to assess dynamic changes in miR-548c-3p, E2F3, and FOXM1 mRNA levels.

The Interaction of CircESR1 and HNRNPAB Regulates Cell Cycle Transition of Breast Cancer Cell.

The mechanisms by which circRNAs regulate estrogen receptor (ER)-positive breast progression and therapeutic resistance remain poorly defined. By screening circRNAs involved in ER signaling, circESR1 was identified as a novel circRNA exhibiting high specificity of expression in ER+ breast cancer. CircESR1 interacted with HNRNPAB, which was transcriptionally activated by ER/SP1 signaling. HNRNPAB promoted the back-splicing and expression of circESR1 by binding to the Alu elements of cognate pre-mRNA; and circESR1 transcripts increased the stability and expression of HNRNPAB, ensuring an efficie

SOX4-Mediated Post-transcriptional Suppression of PTEN via miR-106b∼25 Cluster Contributes to Prostate Cancer Aggressiveness.

UNLABELLED: Molecular-based risk stratification of prostate cancer holds significant potential for guiding precision therapeutic strategies. Previous studies revealed that SOX4 activation drives prostate cancer progression in PTEN-deficient tumors through the PI3K-AKT signaling pathway. However, the mechanistic interplay between SOX4 and PTEN, as well as their clinical utility for prognostic stratification, remains to be elucidated. In this study, we revealed that SOX4 expression is increased in patients with prostate cancer with low PTEN levels, and the expression of SOX4 and PTEN is inversel

VCAN Is Essential for ERK5-Driven Tumorigenesis in Soft Tissue Sarcoma.

The ERK5 signaling pathway has recently emerged as a critical regulator of soft tissue sarcoma (STS) biology, contributing to tumor initiation, progression, and maintenance. In this study, we identify VCAN, a chondroitin sulfate proteoglycan, as a novel transcriptional target of ERK5 and a central mediator of ERK5-related oncogenesis. Through a combination of genetic (silencing, overexpression) and pharmacological approaches, applied in both a chemically induced murine sarcoma model and several human STS cell lines, we demonstrate that ERK5 positively regulates VCAN expression. Functionally, V

Tumour cell-intrinsic complement C1r and C1s regulate cancer cell fitness and shape the immune microenvironment in triple-negative breast cancer.

Emerging evidence positions tumour cell-intrinsic complement proteins as regulators of cancer cell behaviour and immune crosstalk, yet their role in triple-negative breast cancer (TNBC) remains undefined. Here, transcriptomic profiling across breast cancer molecular subtypes reveals that C1R, C1S and C3 are preferentially expressed in TNBC cells, both in patient tumours and in vitro, and that their expression is upregulated by pro-inflammatory cytokines. In situ mRNA hybridization and hyperplex SeqIF immunofluorescence demonstrate that C1R and C1S share more closely matched expression patterns

DNAJC9 promotes cervical cancer cell proliferation by regulating GLI1 expression.

DNAJC9, an HSP40 family member with histone chaperone function, exhibits unclear roles in cervical cancer. DNAJC9 is specifically overexpressed in malignant cervical cancer cells, and downregulation of DNAJC9 inhibits proliferation, induces G1/S arrest, and suppresses tumorigenicity. GLI1 has been identified as a key downstream effector of DNAJC9, and GLI1 rescue reverses proliferation defects. Mechanistically, DNAJC9 promotes the p300-H3 interaction to sustain H3K27ac at the GLI1 enhancer and facilitate GLI1 transcription, driving proliferation. Furthermore, DNAJC9 expression correlates posit

ORC6 Exerts Oncogenic Functions in Kidney Renal Clear Cell Carcinoma by Regulating Cell Proliferation and Apoptosis: A Prognostic and Immunological Indicator for Future Clinical Interventions.

With the application of precise clinical interventions in tumor treatment, it is of great value to explore novel biomarkers and their underlying molecular mechanisms. In this paper, we firstly explored the prognostic, immunological features, functions, and potential mechanisms of ORC6 in kidney renal clear cell carcinoma (KIRC) via single-cell and bulk RNA sequencing. Single-cell and bulk RNA sequencing data related to ORC6 data in KIRC were acquired from shared online databases. Spearman correlation analyses were carried out to investigate the relationships between ORC6 expression and interes

CAFs and Endocrine Therapy Resistance in Hormone Receptor-Positive Breast Cancer.

The development of endocrine resistance represents a major obstacle when treating hormone receptor-positive breast cancer. The tumor microenvironment (TME), represented by cancer-associated fibroblasts (CAFs) in this context, has recently been proposed as a key mediator significantly contributing to resistance against currently available endocrine therapies. The exact mechanisms behind this interaction are not fully understood; specific breast CAF subtypes have been linked to it, such as CAFs lacking the expression of the glycoprotein CD146 or maintaining the expression of CD63. Other proposed

Bioinformatics Profiling of a Paeoniflorin-Associated "Macrophage-Lactylation" Axis in Hepatocellular Carcinoma: An LDHA-Based Prognostic Model and Structural Computational Evidence.

Hepatocellular carcinoma (HCC) progression is shaped by crosstalk between the tumor immune microenvironment (TME) and metabolic reprogramming. This study aims to characterize a macrophage-lactylation molecular axis in HCC and to develop a quantitative prognostic stratification model. Using the TCGA-LIHC cohort, differentially expressed genes were intersected with Paeoniflorin (PF)-related targets, HCC disease targets, and macrophage-/lactylation-related genes to identify candidate genes. Prognostic genes were selected through Cox and LASSO-Cox analyses to construct a risk score model, followed

lnc-ALX1-2:10 is a novel regulator that enhances proliferation, migration and invasion in prostate cancer cells.

Although long noncoding RNAs (lncRNAs) have been implicated in the progression of prostate cancer (PCa), the functional roles of many of these molecules, especially in metastasis, remain poorly understood. To identify novel lncRNAs linked to PCa malignant phenotypes, we analyzed the differences in lncRNA expression between the highly metastatic PCa cell line (PC-3 M-1E8) and the poorly metastatic PCa cell line (PC-3 M-2B4) using transcriptome sequencing. This differential expression was confirmed by RT-qPCR, which showed that the lnc-ALX1-2 gene cluster (including lnc-ALX1-2:5, lnc-ALX1-2:7, l

Inhibition of histone demethylase LSD1 suppresses CD47 expression and enhances efficacy of CD47 blockade in breast cancer.

BACKGROUND: CD47 functions as a "don't eat me" checkpoint, inhibiting macrophage-mediated phagocytosis in triple-negative breast cancer (TNBC). While anti-CD47 therapies can restore immune surveillance, their efficacy in TNBC is often limited by immune evasion and drug development challenges. METHODS: We investigated the crosstalk between the histone demethylase lysine-specific demethylase 1 (LSD1) and CD47 signaling in TNBC using in silico datasets, isogenic cell lines, conditional BRCA1 knockout models, and syngeneic mouse models. Techniques such as immunohistochemistry, multiplex immunoflu

Flavonol-mediated modulation of angiogenesis-related microRNAs in breast cancer: a narrative synthesis of current evidence and knowledge gaps.

Angiogenesis is essential for breast cancer progression and metastasis; however, the clinical impact of vascular endothelial growth factor (VEGF)-targeted therapies remains limited due to adaptive resistance and activation of compensatory angiogenic pathways. Angiogenesis-regulating microRNAs (angiomiRs) act as upstream modulators of both VEGF-dependent and VEGF-independent signaling networks contributing to vascular plasticity, immune evasion, and therapeutic escape. Dietary flavonols such as kaempferol, myricetin, and quercetin have demonstrated anti-angiogenic activity in preclinical models

A multi-gene urinary extracellular vesicle mRNA panel as a non-invasive biomarkers for prostate cancer detection.

Prostate cancer (PCa) remains one of the most common malignancies in men older than 50 globally and in India. Although prostate-specific antigen (PSA) remains the standard diagnostic tool, its low specificity leads to overdiagnosis and unnecessary biopsies, highlighting the need for improved non-invasive biomarkers. Urinary extracellular vesicles (UEVs) have emerged as a promising, non-invasive source of molecular biomarkers, particularly RNA, for cancer detection. This study evaluated the diagnostic potential of six PCa associated genes (AGR2, ERG, PTEN, PCA3, AMACR, SPDEF) in UEVs.UEVs were

Small Molecule Liver X Receptor Modulator GAC0001E5 Targets Mechanisms of Endocrine Resistance in Estrogen Receptor-Positive Breast Cancer Cells.

Endocrine therapy is an effective and common treatment strategy for estrogen receptor (ER)-positive breast cancers. However, the development of endocrine resistance, through genetic mutations and epigenetic alterations, in about 40% of treated patients remains a significant therapeutic challenge. Liver X receptors (LXRs) are nuclear receptors that regulate lipid metabolism and cholesterol homeostasis and have been implicated in metabolic reprogramming in breast cancers and other malignancies. We previously identified a novel LXR ligand GAC0001E5 (1E5), with potent antiproliferative activity ac

SGO1 and SGO2 are Associated With Disease Progression and an Immunosuppressive Microenvironment in Papillary Renal Cell Carcinoma.

BACKGROUND: Shugoshin proteins SGO1 and SGO2 regulate chromosome cohesion and segregation during mitosis and meiosis, and their aberrant expression is implicated in several cancers. However, the mechanistic involvement of these two proteins in papillary renal cell carcinoma (KIRP) and their contribution to immune evasion remains unclear. Therefore, in the present study, we aimed to determine SGO1 and SGO2 expression patterns and prognostic significance, clarify their functional involvement in driving tumor progression, and evaluate their impact on the tumor immune microenvironment.
