Association of polygenic risk scores with low-density lipoprotein cholesterol levels and control: findings from the Hispanic Community Health Study/Study of Latinos (HCHS/SOL).

BACKGROUND: Hypercholesterolemia is a prevalent cardiovascular (CVD) risk factor among Hispanics/Latinos; however, rates of cholesterol awareness and treatment are low in this population. Using a polygenic risk score (PRS) associated with low-density lipoprotein cholesterol (LDL) levels may help identify those who would benefit most from statin therapy. Previous LDL-PRS have been developed but they have not been properly evaluated for use in a highly diverse Hispanic/Latino population. METHODS: The Hispanic Community Health Study/Study of Latinos (HCHS/SOL) is a prospective study that enrolle

Assessment of Nutritional Risk Representations in Publicly Available Data in Saudi Arabia: A Triangulated Scoping Study.

Background: Saudi Arabia maintains official statistics on its food security, in addition to conducting surveys on health parameters and performing national health assessments. Also, several academic papers on dietary patterns are published every year. Many published documents analyze data on food availability, self-reported eating habits, and biomarker-based health risks. However, individually, they represent distinct aspects of nutrition. Hence, this study was undertaken to examine publicly accessible nutrition-related evidence in Saudi Arabia and assess whether existing public food and nutri

Association of Statin Discontinuation in Pregnancy With Maternal Cardiovascular Health and Birth Outcomes: A Nationwide Cohort Study.

BACKGROUND: Discontinuing statin therapy before pregnancy remains challenging, especially in high-risk women. We evaluated the risks of maternal cardiovascular, gestational, and fetal outcomes associated with continuing versus discontinuing statin therapy before the last menstrual period (LMP). METHODS: We conducted a nationwide cohort study using data from the National Health Insurance Database of South Korea collected between 2009 and 2023. Women who used statins for 12 to 24 weeks before their LMP between 2010 and 2022 were stratified by whether they discontinued statins before their LMP.

Rationale and design of the Corrie Lipids Program: Impact of a digital health-enabled implementation initiative to improve lipid management.

RATIONALE: Atherosclerotic cardiovascular disease (ASCVD) is the leading cause of morbidity and mortality worldwide. Lowering low-density lipoprotein cholesterol (LDL-C) with statins and nonstatin therapies has been demonstrated to reduce cardiovascular events. However, real-world evidence consistently reveals large treatment gaps, including underprescribing of lipid-lowering therapy, limited intensification, and poor patient adherence. Observational registries describe these treatment gaps, and randomized trials demonstrate therapy efficacy under controlled conditions, but interventions trans

N -aldehyde modified phosphatidylethanolamines formed by lipid peroxidation inhibit efferocytosis.

BACKGROUND: Impairments in efferocytosis, the phagocytic clearance of apoptotic cells, contribute to the development of chronic inflammation and promote the formation of large necrotic cores within atherosclerotic plaques. A better understanding of the factors that impair efferocytosis could lead to improved interventions. Many chronic inflammatory conditions feature excessive lipid peroxidation, which generates reactive lipid aldehydes that can potentially form N -aldehyde-modified phosphatidylethanolamines (NAMPs). We therefore investigated the extent to which levels of NAMPs are elevated un

Age- and sex-specific reference values for plasma lipids and lipoproteins in thai adults: a national health examination survey analysis.

BACKGROUND: Age-, sex-, and country-specific reference values for lipoprotein levels are essential for identifying lipoprotein disorders, such as familial hypercholesterolemia (FH). However, the Thai-specific data are limited. This study aimed to establish age-and sex-specific reference percentiles for lipid and lipoprotein levels in Thai adults. METHODS: This study analyzed plasma lipid and lipoprotein levels from the 6th National Health Examination Survey (NHES-VI) conducted in Thailand. Data from Thai adults without cardiovascular disease or lipid-lowering medication were examined to deter

Treatment patterns and impact of glucocorticoids on health outcomes in generalized myasthenia gravis: a retrospective observational study based on the Medical Data Vision database in Japan.

INTRODUCTION: Generalized myasthenia gravis (gMG) is a rare, chronic, autoimmune disorder with a substantial disease burden. Oral glucocorticoids (GCs) are one of the common early-line treatments used globally, but there are growing concerns regarding risks associated with GC adverse effects, which can have serious medical consequences. In Japan, an important gMG treatment goal is minimal disease manifestation with a low target GC dose (≤5 mg/day) to minimize GC exposure. This study aimed to assess gMG treatment patterns in Japan, with a focus on GC exposure and incident comorbidities associat

Analysis of early effects of human APOE isoforms on Alzheimer's disease and type III hyperlipoproteinemia pathways using knock-in rat models with humanized APP and APOE.

APOE is a major genetic factor in late-onset Alzheimer's disease (LOAD), with APOE4 increasing risk, APOE3 acting as neutral, and APOE2 offering protection. APOE also plays key role in lipid metabolism, affecting both peripheral and central systems, particularly in lipoprotein metabolism in triglyceride and cholesterol regulation. APOE2 is linked to Hyperlipoproteinemia type III (HLP), characterized by mixed hypercholesterolemia and hypertriglyceridemia due to impaired binding to Low-Density Lipoproteins receptors. To explore the impact of human APOE isoforms on LOAD and lipid metabolism, we d

Coronary Computed Tomography Angiography-Derived Modified Duke Index Is Associated with Peri-Coronary Fat Attenuation Index and Predicts Severity of Coronary Inflammation.

Background and Objectives: The modified Duke index derived from coronary computed tomography angiography (CCTA) was designed to predict cardiovascular outcomes based on the severity of coronary stenosis. However, it does not take into consideration the presence or severity of peri-coronary inflammation. The peri-coronary fat attenuation index (FAI) is a novel imaging marker determined by CCTA which reflects the degree of inflammation in the coronary tree in patients with coronary artery disease. To assess the association between the modified Duke index assessed by CCTA, cardiovascular risk fac

Obesity-Associated Breast Cancer: Analysis of Risk Factors and Current Clinical Evaluation.

Several studies show that a significantly stronger association is obvious between increased body mass index (BMI) and higher breast cancer incidence. Additionally, obese and postmenopausal women are at higher risk of all-cause and breast cancer-specific mortality compared with non-obese women with breast cancer. In this context, increased levels of estrogens, excessive aromatization activity of the adipose tissue, overexpression of pro-inflammatory cytokines, insulin resistance, adipocyte-derived adipokines, hypercholesterolemia, and excessive oxidative stress contribute to the development of

Risk Factors for Surgical Treatment of Lumbar Degenerative Disc Disease in Middle-aged and Older Women: A Prospective Case-Control Study of 2370 Subjects.

OBJECTIVE: Given the distinct physiological and societal traits between women and men, we propose that there are distinct risk factors for lumbar degenerative disc disease surgeries, including lumbar disc herniation (LDH) and lumbar spinal stenosis (LSS), in middle-aged and older populations. However, few studies have focused on middle-aged and older women. This study aims to identify these risk factors specifically in this population. METHODS: In this case-control study, the study group comprised 1202 women aged ≥ 45 years who underwent operative treatment of lumbar degenerative disc disease

Retinal microvascular alterations in patients with active rheumatoid arthritis without cardiovascular risk factors: the potential effects of T cell co-stimulation blockade.

BACKGROUND: The evaluation of microvascular alterations might provide clinically useful information for patients with an increased cardiovascular (CV) risk, such as those with rheumatoid arthritis (RA), being the small artery remodeling the earliest form of target organ damage in primary CV diseases, such as arterial hypertension. The evaluation of retinal arterioles is a non-invasive technique aimed to identify an early microvascular damage, represented by the increase of the wall-to-lumen ratio (WLR) index. Abatacept (ABA), a T-cell co-stimulator blocker, is used to treat RA. A CV protective

The effect of cardiovascular risk on disease progression in de novo Parkinson's disease patients: An observational analysis.

BACKGROUND: Currently available treatment options for Parkinson's disease are symptomatic and do not alter the course of the disease. Recent studies have raised the possibility that cardiovascular risk management may slow the progression of the disease. OBJECTIVES: We estimated the effect of baseline cardiovascular risk factors on the progression of Parkinson's disease, using measures for PD-specific motor signs and cognitive functions. METHODS: We used data from 424 de novo Parkinson's disease patients and 199 age-matched controls from the observational, multicenter Parkinson's Progression
