Bayesian prior elicitation on the efficacy of medical therapies in perianal fistulizing Crohn's disease.

BACKGROUND & AIMS: Robust evidence for most licensed Crohn's disease therapies is lacking for perianal fistula outcomes due to a lack of dedicated clinical trials. This study aimed to use a Bayesian framework to determine the efficacy of medical therapies for perianal fistulizing Crohn's disease (PFCD). METHODS: A formal prior elicitation exercise was conducted by a group of 11 gastroenterologists and 5 statisticians. Consensus priors were developed leveraging both existing published data and clinical expertise, to determine one-year fistula remission rates for medical treatments with 5 diffe

Thyroid-stimulating hormone enhances platelet activation and arterial thrombosis.

AIMS: Thyroid-stimulating hormone (TSH), elevated in conditions like subclinical hypothyroidism, has been clinically linked to a higher risk of morbidity and mortality in patients with coronary artery disease (CAD). Here, we aim to investigate the effects of TSH on CAD-associated platelet activation and arterial thrombosis, along with the underlying mechanisms. METHODS AND RESULTS: Clinical study demonstrated that higher TSH levels are associated with an increased risk of major adverse cardiovascular events in CAD patients. In vitro human and mouse platelet function studies found that TSH pot

Polymerizing laminins: Assembly, functions and disorders.

Laminins initiate basement membrane (BM) assembly by adhering to cells, establishing cytoskeletal links through integrin and dystroglycan receptors, and by polymerizing to form a sheet-like provisional matrix that recruits other BM components to complete assembly. Laminin polymerization is mediated by the LN domain tips of the three short arms that bind together to form "polymer nodes" connecting adjacent laminins. The first step of this assembly is the low affinity binding of a β-LN domain to a γ-LN domain, followed by a higher affinity binding of an α-LN domain to the β-γ complex. Negative s

FAPI Dimerization for Theranostic Applications: Molecular Design, Preclinical Validation, and Clinical Translation.

Fibroblast activation protein (FAP), a serine protease overexpressed in cancer-associated fibroblasts of >90% epithelial malignancies, has emerged as a highly promising pan-cancer target for theranostic radiopharmaceuticals. However, the rapid clearance of monomeric FAP inhibitors (FAPIs) limits their therapeutic efficacy despite excellent diagnostic performance. Multimerization strategies-particularly dimeric and heterodimeric constructs-have been developed to overcome this limitation through enhanced binding avidity and prolonged tumor retention. This review provides a comprehensive analysis

Platelet-erythrocyte complex production is exacerbated by histones and protected by small polyanion histone neutralizing compounds.

Platelet-erythrocyte (PE) complexes, composed of single erythrocytes bound to platelets, are a physiological component of the bloodstream and contribute to erythrocyte turnover. Conditions in which PE complex production is increased or their clearance from the circulation is impeded are associated with hypercoagulable states and thrombocytopenia. Sepsis is a serious acute inflammatory condition that is often complicated by major hemostatic derangements, including thrombosis, anemia and thrombocytopenia, and an important driver of sepsis pathology are extracellular histones, which are released

Δ9-Tetrahydrocannabinol (THC) and Cannabidiol (CBD) Diminish CD16+ Monocyte-Induced Astrocyte Inflammation, while THC Uniquely Inhibits Monocyte Chemotaxis Independent of HIV Status.

CD16+ monocytes are a minor subset of the total monocyte population that play a disproportionate role in contributing to neuroinflammation in human immunodeficiency virus (HIV)-associated neurocognitive disorders (HAND). This has been evidenced by the enhanced transmigration of CD16+ monocytes into the brain compared to their CD16- counterpart. CD16+ monocytes can be activated by HIV ssRNAs through toll-like receptors (TLR) 7 and TLR8, and subsequently interact with brain-resident cells, including astrocytes. Previous studies from our laboratory identified monocyte-derived IL-1ß as an inducing

Molecular insights and therapeutic innovations in low-risk human papillomavirus-associated cutaneous wart.

Human papillomavirus (HPV) is a DNA virus that belongs to the Papillomaviridae family. Among the various types, high-risk strains are associated to malignancy, whereas low-risk types cause benign skin warts due to persistent infection. Unlike high-risk HPVs, low-risk HPV genomes remain in an episomal state while expressing E6/E7 proteins. These proteins exhibit a reduced ability to degrade pRb and p53, which finally leads to controlled epithelial hyperplasia instead of developing malignancy. Infection with low-risk HPV activates distinct host signaling pathways, ultimately promoting the prolif

Analysis of the Short- and Long-Term Immune Response in BALB/c Mice Immunized with Total Naegleria fowleri Extract Co-Administered with Cholera Toxin.

BACKGROUND: Naegleria fowleri is a free-living amoeba that inhabits warm freshwater and causes primary amoebic meningoencephalitis (PAM), a rapidly fatal infection with >95% mortality. Due to the lack of early diagnosis and effective therapy, preventive vaccination represents a promising strategy. METHODS: This study evaluated short- and long-term immune protection in BALB/c mice (20 mice per group) immunized intranasally with total N. fowleri extract co-administered with cholera toxin (CT). Mice were challenged with a lethal dose of trophozoites either 24 h (short-term) or three months (long

An intricate relationship between fibrosis and autoimmune diseases: a specific focus on unraveling its molecular, immunological, and epigenetic drivers.

Fibrosis is a pathological process of wound healing, characterized by excessive deposition of extracellular matrix (ECM) and chronic activation of fibroblasts, leading to organ scarring and a decline in function. Fibrogenesis is predominantly initiated by tissue injury and sustained inflammation, further driven by the complex interplay of growth factors, cytokines, metabolic alterations, and epigenetic reprogramming. Activated myofibroblasts and immune cells function as primary profibrotic mediators. The central molecular pathways implicated include TGF-β/SMAD signaling, non-canonical cascades

Effects of Cell Seeding Density, Extracellular Matrix Composition, and Geometry on Yes-Associated Protein Translocation in Corneal Fibroblasts.

Corneal fibroblasts are central to normal and abnormal wound healing in the cornea. During the wound healing process, several biochemical and biophysical signals that are present in the extracellular matrix (ECM) play critical roles in regulating corneal fibroblast behavior. The translocation and activation of Yes-associated protein (YAP)-a main transcriptional factor in the Hippo signaling pathway-is one example of mechanotransduction involving these signals. However, how corneal fibroblasts integrate these simultaneous cues is unknown. In this study, we utilized well-defined micropatterns of

Identifying conserved metastatic pathways across cancers through integrated transcriptomic and network analysis.

Metastasis remains the leading cause of cancer-related mortality, yet shared molecular mechanisms across cancer types are poorly understood. Identifying conserved metastatic pathways could offer new therapeutic targets. We performed an integrative cancer analysis across five cancer types (breast, lung, endometrial, prostate, colorectal) using RNA-Seq datasets from the GEO database. Differentially expressed genes (DEGs) between primary and metastatic tumors were identified using limma. Overlapping DEGs across cancers underwent pathway enrichment and protein-protein interaction (PPI) network ana

LncRNA NR_045147 modulates osteogenic differentiation and migration in PDLSCs via ITGB3BP degradation and mitochondrial dysfunction.

Periodontitis is an inflammation of the alveolar bone and soft tissue surrounding the teeth. Although mesenchymal stem cells (MSCs) have been implicated in periodontal regeneration, the mechanisms by which they promote osteogenesis remain unclear. We examined whether epigenetic modifications mediated by the long-noncoding RNA (lncRNA) NR_045147, which plays a crucial role in cancer, influence the osteogenic differentiation of periodontal ligament stem cells (PDLSCs). Alkaline phosphatase staining, alizarin red staining, and western blotting were used to detect the effects of NR_045147 on PDLSC

miR-758-3p Interferes with Neuronal Apoptosis in Cerebral Ischemia-Reperfusion by Inhibiting ILK.

This study investigated the role of integrin-linked kinase (ILK) in neuronal apoptosis induced by cerebral ischemia‒reperfusion injury (CIRI) and its interaction with a circRNA (0000964) and miR-758-3p. Using in vivo and in vitro rat models, we clarified how ILK regulates neuronal apoptosis during CIRI. Our findings revealed that ILK expression is upregulated in response to CIRI and is modulated by the circRNA (0000964)/miR-758-3p axis. This study provides new insights into the molecular mechanisms of CIRI and suggests potential therapeutic targets to reduce neuronal apoptosis. A CIRI rat mode

Platelets display immunophenotypic alterations and dysregulated transcriptomic signature in Philadelphia-negative myeloproliferative neoplasms.

INTRODUCTION & OBJECTIVES: Polycythemia vera (PV), essential thrombocythemia (ET), and primary myelofibrosis (MF) are Philadelphia-negative myeloproliferative neoplasms (MPN) associated with gain-of-function mutations in JAK2, CALR, and MPL genes. Chronic inflammation is a central hallmark of MPN, significantly contributing to disease pathogenesis and progression and severe complications such as thrombosis. Alterations in platelet immunophenotype and gene expression may influence the thromboinflammatory state observed in MPN patients. We aimed to characterize platelet immunophenotype, ex vivo
