Psoriasis and psoriatic arthritis (PsA) are chronic inflammatory diseases, recognized as one entity, termed psoriatic disease (PsD). CD39 is an ectonucleotidase that hydrolyzes ATP to adenosine and is expressed on a human Treg sub-population capable of exerting interleukin (IL-) 17 suppression. We aimed to investigate the effect of apremilast on CD39 expression in T cells from patients with PsD. Peripheral blood mononuclear cells (PBMCs) were analyzed by multicolor flow cytometry and appropriate monoclonal antibodies. Intracellular cytokine production of PBMCs was measured after in vitro PMA p

We investigated whether plasma cytomegalovirus (CMV) IL-10 (cmvIL-10) levels could serve as a biomarker of active CMV replication in allogeneic hematopoietic transplant recipients (allo-HCT) in the presence or absence of letermovir (LMV) prophylaxis. A total of 189 leftover plasma samples that tested positive for CMV DNA (Alinity m CMV assay), representing 33 episodes of CMV DNAemia were run on a laboratory-developed enzyme-linked immunosorbent assay for cmvIL-10 quantification. Eighteen episodes developed during LMV prophylaxis. Overall, 16 episodes of CMV DNAemia were classified as clinicall

BACKGROUND: Chronic hepatitis B virus (HBV) infection is characterized by selective impairment of virus-specific immune responses. However, progressive liver disease, particularly cirrhosis, is associated with broader immune remodeling that may influence B cell phenotype and function. CD72 is a negative regulator of B cell receptor (BCR) signaling but its role in CD5+B cell regulation under these conditions remains unclear.
METHODS: Peripheral Blood Mononuclear cells (PBMCs) were isolated from 51 HBV-infected patients at differing disease stages and 24 healthy controls by density gradient cen

Infection with the Epstein-Barr virus (EBV) is a major risk factor for the development of cancer and autoimmune disorders. The virus enters the body in the pharynx, but EBV causes disease in distant organs, including the gut and the brain. Here we show, using in vitro culture and mouse infection models, that EBV-infected B cells display features of homing cells. Infected B cells undergo migration following paracrine CCL4 release and CCR1 induction, while CCR1 deficiency inhibits migration and, unexpectedly, proliferation of infected B cells. Furthermore, migrating EBV-infected B cells undergo

To investigate the role of Fuzi polysaccharide(FPS) in combination with lenvatinib(LEN) in regulating tumor-associated macrophage(TAM) polarization to enhance the anti-hepatocellular carcinoma(HCC) efficacy, the non-toxic concentrations of FPS and LEN on RAW264.7 cells were screened by cell counting kit-8(CCK-8). The co-culture system of RAW264.7 and Hepa1-6 cells was established, and flow cytometry was used to detect changes in M1 and M2 macrophage phenotypes. Quantitative real-time polymerase chain reaction(qRT-PCR) was performed to assess the mRNA expression levels of M1-associated factors,

Helicobacter pylori infection is one of the most common factors inducing gastric mucosal inflammation. Upon infecting gastric epithelial cells, H. pylori generates reactive oxygen species (ROS), which act as inducers of matrix metalloproteinases (MMPs). ROS can regulate MMP gene expression and promote their production through the ERK signaling pathway, with MMP-10 being a primary MMP induced during H. pylori infection. By mediating the remodeling of the gastric epithelial and lamina propria layers, MMP-10 enhances H. pylori colonization and its pro-inflammatory effects. As resistance to eradic

INTRODUCTION: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease characterized by dysregulated innate and adaptive immune responses. However, data on immunologic disturbances in SLE flares remains limited. Thus, we aimed to evaluate serum levels of selected cytokines involved in its pathogenesis such as APRIL (a proliferation-inducing ligand), BAFF (B cell activating factor), and interleukin(IL)-10 levels in SLE patients to identify potential biomarkers of disease activity.
PATIENTS AND METHODS: We investigated 52 patients with SLE: 15 (28.8%) with disease exacerbation (SLE di

BACKGROUND: Single nucleotide polymorphisms (SNPs) of cancer-immunity relevant genes may decide the extent of tumor immunosurveillance and have clinical significance. The Immuno ALCL trial was designed to investigate whether genetic variability in 13 cancer-immunity relevant genes correlated with clinical features and outcome of anaplastic lymphoma kinase (ALK)-positive anaplastic large cell lymphoma (ALCL) patients. METHODS: One hundred eighty patients were enrolled and genotyped for 14 SNPs. Age at diagnosis, progression-free survival, histological subtype and anti-ALK antibody titer data we

Chronic infection with the hepatitis B virus (HBV) remains a major public health issue. Its progression depends on several factors, including immunogenetic factors. The aim of this study was to investigate the association between interleukin 10 gene (IL10) polymorphism and the progression of this infection. This retrospective case-control study involved 156 chronic HBV carriers (CHBV-C) and 174 healthy HBV-negative controls (HBsAg-). The analysis of IL10 promoter polymorphism was carried out using the TaqMan allele discrimination technique at two single nucleotide polymorphisms (SNPs), -592A>C

BACKGROUND: The role of B cells in antitumor immunity remains controversial, with studies suggesting the protumor and antitumor activity. This controversy may be due to the heterogeneity in B cell populations, as the balance among the subtypes may impact tumor progression. The immunosuppressive regulatory B cells (Breg) release interleukin 10 (IL-10) but only represent a minor population. Additionally, tumor-specific antibodies (Abs) also exhibit antitumor and protumor functions dependent on the Ab isotype. Transcription factor c-Maf has been suggested to contribute to the regulation of IL-10

Better understanding of the host responses to Mycobacterium tuberculosis infections is required to prevent tuberculosis and develop new therapeutic interventions. The host transcription factor BHLHE40 is essential for controlling M. tuberculosis infection, in part by repressing Il10 expression, where excess IL-10 contributes to the early susceptibility of Bhlhe40-/- mice to M. tuberculosis infection. Deletion of Bhlhe40 in lung macrophages and dendritic cells is sufficient to increase the susceptibility of mice to M. tuberculosis infection, but how BHLHE40 impacts macrophage and dendritic cell

At present, the characteristics of double-hit multiple myeloma (DHMM) are unknown. We retrospectively analyzed the clinical data from 433 new diagnosed MM patients and found that DHMM have a higher β2-MG level and percentage of bone marrow plasma cell. Cox regression analysis showed that the prognosis of DHMM was not limited by clinical indicators. The abnormal proliferation of bone marrow in DHMM is obvious, and the proportion of poorly differentiated plasma cell is high. By collecting specimens from our center and performing flow cytometry to analyze the immunophenotypic and functional chara

BACKGROUND: Hepatocellular carcinoma (HCC) is the fifth most diagnosed cancer and the second leading cause of cancer-related deaths worldwide. Sorafenib is the standard treatment used in the advanced stages of HCC. Cell therapy with mesenchymal stem cells (MSCs)-based cell therapy has proven effective in immune regulation and tumour growth inhibition.
OBJECTIVES: In this study, we investigated the anti-inflammatory effect of MSCs on HCC xenografts.
METHODS: Human HepG2 cell lines were subcutaneously implanted into the flank of 12 nude mice, divided into three groups: the control group, the I

Adaptive immune components are thought to exert non-overlapping roles in antimicrobial host defence, with antibodies targeting pathogens in the extracellular environment and T cells eliminating infection inside cells1,2. Reliance on antibodies for vertically transferred immunity from mothers to babies may explain neonatal susceptibility to intracellular infections3,4. Here we show that pregnancy-induced post-translational antibody modification enables protection against the prototypical intracellular pathogen Listeria monocytogenes. Infection susceptibility was reversed in neonatal mice born t

In the present study, we evaluated the anti-inflammatory properties of Lactiplantibacillus plantarum 22A-3 (LP22A3) and attempted to elucidate the underlying molecular mechanism. The oral administration of LP22A3 significantly inhibited body weight reduction and decreased colon shortening and colitis score in mice with dextran sulfate sodium (DSS)-induced colitis. It was demonstrated that the production of the active-form of TGF-β tended to increase in both the intestinal epithelial cells (IECs) of the ileum and serum but not in the colon of non-DSS-treated mice by LP22A3. IL-10 level in serum

Spinal cord injury results in irreversible tissue damage followed by a very limited recovery of function. In this study we investigated whether transplantation of undifferentiated human induced pluripotent stem cells (hiPSCs) into the injured rat spinal cord is able to induce morphological and functional improvement. hiPSCs were grafted intraspinally or intravenously one week after a thoracic (T11) spinal cord contusion injury performed in Fischer 344 rats. Grafted animals showed significantly better functional recovery than the control rats which received only contusion injury. Morphologicall

The chemokine CXCL13 controls the normal organization of secondary lymphoid tissues and the neogenesis of ectopic lymphoid structures in nonlymphoid organs, particularly the lungs. The progression and severity of idiopathic pulmonary fibrosis (IPF), a fatal and irreversible interstitial lung disease, is predicted by the circulating blood concentrations of CXCL13. Although CXCL13 is produced by pulmonary tissues, it has not been determined which cells are involved. This study examines CXCL13 production by lung tissue macrophages from patients with IPF and the signaling pathways controlling CXCL

Bryostatin-1 (Bry-1) has been proven to be effective and safe in clinical trials of a variety of immune-related diseases. However, little is known about its effect on Crohn's disease (CD). We aimed to investigate the impact of Bry-1 on CD-like colitis and determine the mechanism underlying this effect. In the present study, 15-week-old male Il-10-/- mice with spontaneous colitis were divided into positive control and Bry-1-treated (Bry-1, 30 μg/kg every other day, injected intraperitoneally for 4 weeks) groups. Age-matched, male wild-type (WT) mice were used as a negative control. The effects
