Curcumin nanoparticles attenuate neurochemical and neurobehavioral deficits in experimental model of Huntington's disease.

Till date, an exact causative pathway responsible for neurodegeneration in Huntington's disease (HD) remains elusive; however, mitochondrial dysfunction appears to play an important role in HD pathogenesis. Therefore, strategies to attenuate mitochondrial impairments could provide a potential therapeutic intervention. In the present study, we used curcumin encapsulated solid lipid nanoparticles (C-SLNs) to ameliorate 3-nitropropionic acid (3-NP)-induced HD in rats. Results of MTT (3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide) assay and succinate dehydrogenase (SDH) staining of s

Experimental study on effect of hydroalcoholic extract of Emblica officinalis fruits on glucose homeostasis and metabolic parameters.

Polyphenols from natural source are potential therapeutics that act alone or supplement anti-diabetic drugs in the prevention and treatment of diabetes. The present investigation was undertaken to study the effect of hydroalcoholic extract (HE) of fruits of Emblica officinalis on type 1 diabetic rats. Diabetes was induced by streptozotocin (STZ) (45 mg/kg i.v.). HE (100 mg/kg, p.o.) was administered for 4 weeks and at the end of treatment, blood samples were collected and analyzed for various biochemical parameters. STZ produced a diabetic state exhibiting all the cardinal symptoms such as los

Assessment of oxidative parameters in rat spinal cord after chronic constriction of the sciatic nerve.

Although reactive oxygen species (ROS) are involved in neuropathic pain, the direct relationship between these species and chronic constriction of sciatic nerve (CCI) has not been studied in spinal cord. Thus, this study induced CCI in rats and these animals were sacrificed 3 and 10 days after the surgical procedure to determine the superoxide dismutase (SOD) and catalase activities, as well as ascorbic acid, hydrogen peroxide (H(2)O(2)) and lipid hydroperoxide levels in lumbosacral spinal cord. Von Frey Hair and hot plate tests were performed to assess the degree of mechanical and thermal hyp

Paraoxonase 1 L55M, Q192R and paraoxonase 2 S311C alleles in atherothrombosis.

Increased oxidative stress is known to play a role in the pathogenesis of atherosclerosis, and polymorphisms in genes encoding for enzymes involved in modulation of oxidant stress, such as paraoxonases (PONs), provide a potentially powerful approach to study the risk of disease susceptibility. Aim of our study is to investigate the possible association among PONs polymorphisms, clinical and metabolic factors, and atherothrombotic events in an Italian population. We evaluated in 105 subjects, with or without atherosclerotic risk factors, the presence of PON1 L55M, PON1 Q192R, and PON2 S311C gen

Time-course evaluation of oxidative stress-related biomarkers after renal transplantation.

Patients with chronic renal disease have a high prevalence of oxidative stress (OS), which is associated with the cardiovascular complications occurring in this population. The restoration of kidney function after kidney transplantation (KT) can lead to reduction in the metabolic abnormalities and elimination of the OS. Time-dependent changes in OS-related markers and specific kidney function and metabolic parameters were evaluated in patients (N = 39; 23 males; 16 females; mean age = 57 ± 10 years) before (day 0) and after KT (day 1, 7, 30, 90, and 180) to monitor the graft. In particular, to

Phototoxicity of nano titanium dioxides in HaCaT keratinocytes--generation of reactive oxygen species and cell damage.

Nano-sized titanium dioxide (TiO(2)) is among the top five widely used nanomaterials for various applications. In this study, we determine the phototoxicity of TiO(2) nanoparticles (nano-TiO(2)) with different molecular sizes and crystal forms (anatase and rutile) in human skin keratinocytes under UVA irradiation. Our results show that all nano-TiO(2) particles caused phototoxicity, as determined by the MTS assay and by cell membrane damage measured by the lactate dehydrogenase (LDH) assay, both of which were UVA dose- and nano-TiO(2) dose-dependent. The smaller the particle size of the nano-T

Cadmium chloride-induced oxidative stress and DNA damage in the human Jurkat T cell line is not linked to intracellular trace elements depletion.

Cadmium (Cd) is a widespread environmental contaminant. Cd affects the cellular homeostasis and generates damage via complex mechanisms involving interactions with other metals, induction of oxidative stress and apoptotic or necrotic cell death, depending on the cell type and the concentration. The goal of the present study was to investigate the effect of exposure to CdCl(2) on the intracellular trace elements levels, the antioxidant enzyme activities and on DNA damage in the Jurkat T cell line. Cells were exposed to 5, 25 and 50 μM of CdCl(2) for 24 h. Cd significantly reduced the viability

Maximized PUFA measurements improve insight in changes in fatty acid composition in response to temperature.

A general mechanism underlying the response of ectotherms to environmental changes often involves changes in fatty acid composition. Theory predicts that a decrease in temperature causes an increase in unsaturation of fatty acids, with an important role for long-chain poly-unsaturated fatty acids (PUFAs). However, PUFAs are particularly unstable and susceptible to peroxidation, hence subtle differences in fatty acid composition can be challenging to detect. We determined the fatty acid composition in springtail (Collembola) in response to two temperatures (5 degrees C and 25 degrees C). First,

Oxidative stress in plasma from maple syrup urine disease patients during treatment.

Maple Syrup Urine Disease (MSUD) is an autosomal recessive metabolic disorder caused by a deficiency of branched-chain alpha-keto acid dehydrogenase complex activity leading to accumulation of the branched-chain amino acids leucine, isoleucine and valine and their corresponding branched-chain alpha-keto acids. Affected patients usually present hypoglycemia, ketoacidosis, convulsions, poor feeding, coma, psychomotor delay and mental retardation. Considering that the pathophysiology of MSUD is still poorly understood, in this study we evaluated some parameters of oxidative stress, namely thiobar

Carbon disulphide induced activation of liver UDP glucuronosyltransferase in rats pretreated with phenobarbitone.

Carbon disulphide (CS2) exposure has been shown to activate the UDP glucuronosyltransferase of liver microsomes in rats pretreated with phenobarbitone. Now the nature of CS2 induced activation of the enzymes has been studied further. Phenobarbitone pretreated rats were exposed to 0.15% CS2 for 2 hrs on two successive days. The activity of UDP glucuronosyltransferase was measured from the liver microsomes after the enzymes was activated by incubation of the microsomes with various concentrations of the detergents Triton X-100, digitonin and cetylpyridinium chloride. The exposed animals showed a

The emerging role of CETP inhibition in the prevention of Alzheimer's disease.

We recently showed that patients with atherosclerotic cardiovascular disease (ASCVD) carry a substantial but largely unrecognized burden of early Alzheimer's disease (AD) pathology. In the BROADWAY pivotal phase 3 lipid-lowering trial, nearly half of participants with high-risk ASCVD had plasma p-tau217 concentrations above thresholds associated with preclinical AD, yet none had undergone evaluation for cognitive impairment. In this population, apolipoprotein E ε4 (APOE4) carriers were disproportionately represented among those with the highest p-tau217 levels. These findings expose a critical

Targeting glutathione with the triterpenoid CDDO-Im protects against benzo-a-pyrene-1,6-quinone-induced cytotoxicity in endothelial cells.

Epidemiological studies have exhibited a strong correlation between exposure to air pollution and deaths due to vascular diseases such as atherosclerosis. Benzo-a-pyrene-1,6-quinone (BP-1,6-Q) is one of the components of air pollution. This study was to examine the role of GSH in BP-1,6-Q mediated cytotoxicity in human EA.hy96 endothelial cells and demonstrated that induction of cellular glutathione by a potent triterpenoid, CDDO-Im (1-[2-cyano-3-,12-dioxooleana-1,9(11)-dien-28-oyl]imidazole), protects cells against BP-1,6-Q induced protein and lipid damage. Incubation of EA.hy926 endothelial

ESR evidence for in vivo formation of free radicals in tissue of mice exposed to single-walled carbon nanotubes.

Nanomaterials are being utilized in an increasing variety of manufactured goods. Because of their unique physicochemical, electrical, mechanical, and thermal properties, single-walled carbon nanotubes (SWCNTs) have found numerous applications in the electronics, aerospace, chemical, polymer, and pharmaceutical industries. Previously, we have reported that pharyngeal exposure of C57BL/6 mice to SWCNTs caused dose-dependent formation of granulomatous bronchial interstitial pneumonia, fibrosis, oxidative stress, acute inflammatory/cytokine responses, and a decrease in pulmonary function. Morphology-based ovarian stratification reflects coordinated redox imbalance and apoptosis across ovarian tissue, follicular fluid, and granulosa cells in cattle.

Physiological heterogeneity in slaughterhouse-derived bovine ovaries is a major source of variability in assisted reproductive technologies (ART). It remains unclear whether morphology-based ovarian stratification reflects coordinated functional changes across ovarian compartments. Bovine ovaries were stratified into high-quality (HQO) and low-quality (LQO) groups based on five morphological criteria. Oxidative stress and apoptosis were then evaluated in ovarian tissue, follicular fluid (FF), and granulosa cells (GCs). Across ovarian compartments, antioxidant capacity decreased by 30-60% and l

Effects of environmental enrichment on lymph node mitochondrial-redox status and immune response in adult male C57BL/6 mice.

Environmental enrichment (EE), an experimental paradigm promoting cognitive, sensory, social, and motor stimulation, has been associated with neuroprotective effects. Although EE modulates immune function, its influence on the basal redox and metabolic environment of lymph nodes remains unclear. This study investigated whether EE induces physiological immunometabolic and redox adaptations in lymph nodes under non-pathological conditions. Adult male C57BL/6 mice were housed for six weeks in either a standard environment (SE) or EE, followed by evaluation of mitochondrial markers, redox balance,

Neurotoxicogenomic impact of 4-nonylphenol on Heteropneustes fossilis via molecular, histopathological and bioinformatic analysis.

Exposure to 4-Nonylphenol at concentrations of 64 µg/L (low) and 160 µg/L (high) for 30 to 60 days, spanning the pre-spawning to spawning phases in H. fossilis, induces significant adverse effects in the brain, particularly within the telencephalon and cerebellum regions. Additionally, protein content in brain decreases in treated group of 4-Nonylphenol (4-NP), brain acetylcholinesterase (AChE) activity decreases in a manner contingent upon the amount of dose and exposure period. In the brain tissue of H. fossilis, exposure to 4-Nonylphenol (4-NP) results in a dose-dependent decrement of antio
