Chaihu-Guizhi Decoction Attenuates Airway Remodelling in COPD Mice via the TGF-β1/Smads Signalling Pathway: An Integrated Proteomic and Metabolomic Analysis.

Chronic obstructive pulmonary disease (COPD) is characterised by persistent airway injury and remodelling, and the mechanisms by which Chai-Hu-Gui-Zhi Decoction (CGD) acts in COPD remain unclear. This study investigated whether CGD attenuates airway remodelling through the TGF-β1/Smads-EMT-ECM signalling axis. A mouse COPD model was established by cigarette-smoke exposure plus intranasal lipopolysaccharide. After model induction, mice received dexamethasone acetate or low-, medium- or high-dose CGD for 28 days. UHPLC-MS/MS, network pharmacology, molecular docking, molecular dynamics, proteomic

Microwave pulsed vacuum drying improves drying efficiency and quality attributes of Cistanche: comparative evaluation of structural, bioactive, volatile, digestive, and metabolomic characteristics.

This study evaluated the effects of microwave pulsed vacuum drying (MPVD) on the quality attributes and bioactive profile of Cistanche, in comparison with freeze drying (FD) and hot air drying (HAD). Four MPVD vacuum pulse ratios, namely 1:1, 3:1, 5:1, and 7:1, were comparatively investigated. MPVD4 achieved the shortest drying time (130 min), whereas among the tested MPVD conditions, MPVD3 (vacuum pulse ratio 5:1) showed a favorable overall balance among drying efficiency, structural preservation, color attributes, and selected bioactive compounds. MPVD3 showed the highest echinacoside conten

High-fat diet differentially alters hippocampal lipidomic and metabolomic profiles and cognition-related behavior in wild-type and LDL receptor-deficient mice.

Obesity and dyslipidemia are linked to cognitive decline, yet the role of LDL receptor (LDLr) deficiency in cognitive susceptibility to high-fat diet (HFD) remains unclear. This study examined the effects of HFD on cognitive function and hippocampal lipidomic and metabolomic profiles in wild-type (WT) and LDLr⁻/⁻ mice. Male mice of both genotypes were fed a control diet or HFD for 12 weeks and assessed for body weight, serum lipid profiles, anxiety-like behavior, spatial and recognition memory, and hippocampal lipidomic/metabolomic alterations using untargeted LC-MS/MS. HFD increased body weig

Gut microbiota and metabolome signatures in preterm infants with high versus low risk for neurodevelopmental impairment: a prospective, matched, longitudinal multi-omics study.

Preterm birth is a leading global cause of neurodevelopmental impairment (NDI), yet early predictive biomarkers remain elusive. The gut microbiome, developing in parallel with the brain and communicating via the microbiota-gut-brain axis, holds potential as a source of such biomarkers. However, specific longitudinal multi-omics signatures predictive of NDI risk in preterm infants are poorly defined. We conducted a prospective, matched, longitudinal study of 60 preterm infants, classified at 3 months corrected age (CA) into high-risk (HR, n=30) or low-risk (LR, n=30) groups for NDI based on com

Dietary Chlorogenic Acid Attenuates Hepatic Lipid Accumulation and Reprograms Lipid Metabolism in Heat-Stressed Laying Hens: Integrated Transcriptomic and Metabolomic Analyses.

Heat stress leads to excessive hepatic lipid deposition and oxidative imbalance in laying hens, especially during peak laying period. Chlorogenic acid (CGA), a dietary polyphenol with antioxidant and lipid-modulating properties, may improve hepatic lipid homeostasis, yet its effects under heat-stress conditions remain unclear. In this study, 240 Hy-Line Brown laying hens at 36 weeks of age were randomly assigned to one of two treatments (120 hens per treatment, with six replicates of 20 hens each): a basal diet or a basal diet supplemented with 300 mg/kg CGA and subjected to heat-stress condit

Maternal-prenatal gut microbiome-systemic metabolome perturbations and TH2-skewed immunity link to offspring gut microbiome disruption and atopic dermatitis susceptibility.

BACKGROUND: Emerging evidence suggests that maternal-prenatal gut microbiome disturbances shape offspring allergic outcomes through modulation of the in utero immune environment. Yet, no comprehensive clinical studies in human mother–offspring dyads have deconvoluted the maternal-prenatal gut microbiome and systemic immune-metabolome signatures underlying offspring allergic predisposition. METHODS: We performed a longitudinal nested case–control study involving 128 well-characterized mother–offspring dyads from defined cases (offspring with atopic dermatitis (AD); n = 64) and controls (offspri

Unraveling the role of glucosamine: Fructose-6-phosphate aminotransferase (GFAT) in Entamoeba histolytica: A comprehensive exploration through biochemical, reverse genetic, and Metabolomic analyses.

Entamoeba histolytica is a protozoan parasite that causes amebiasis in humans. Its life cycle consisting of two stages: the proliferative trophozoite, which is responsible for clinical symptoms, and the dormant cyst, which mediates disease transmission. The cyst wall is primarily composed of chitin, a polymer of β-1,4-linked N-acetylglucosamine. The hexosamine biosynthetic pathway (HBP) is crucial for producing uridine 5'-diphospho-N-acetyl-D-glucosamine (UDP-GlcNAc), an essential metabolite for chitin and glycoprotein synthesis. Glucosamine:fructose-6-phosphate aminotransferase (GFAT) catalyz

Transcriptomic and Metabolomic Profiling Identifies a Core Gene-Metabolite Axis Driving African Swine Fever Virus Replication in the Soft Tick Ornithodoros lahorensis.

African swine fever virus (ASFV) causes an incurable swine disease with nearly 100% mortality, posing a catastrophic threat to global pig production. The soft tick Ornithodoros lahorensis acts as a critical biological vector that sustains persistent ASFV replication and mediates long-distance viral transmission, yet the molecular mechanisms governing ASFV-tick interplay remain poorly understood. Here, we integrated transcriptomics and metabolomics to systematically dissect molecular changes in O. lahorensis across three infection stages: Uninfected control, early infection (7 days post-infecti
