Firing characteristics of swallowing interneurons in the dorsal medulla during physiologically induced swallowing in perfused brainstem preparation in rats.

Oropharyngeal swallowing is centrally mediated by a swallowing central pattern generator (Sw-CPG) in the medulla oblongata. The activity of the Sw-CPG depends on the sensory inputs determined by physical and chemical bolus properties. Here we investigate the sensory-motor integration during swallowing arising from different sensory sources. To do so we electrically stimulated the superior laryngeal nerve and we triggered swallowing with oral injections of distilled water or capsaicin solution and extracellularly recorded from swallowing interneurons in arterially perfused brainstem preparation

Combined lung and brain ultrasonography for an individualized "brain-protective ventilation strategy" in neurocritical care patients with challenging ventilation needs.

When intracranial hypertension and severe lung damage coexist in the same clinical scenario, their management poses a difficult challenge, especially as concerns mechanical ventilation management. The needs of combined lung and brain protection from secondary damage may conflict, as ventilation strategies commonly used in patients with ARDS are potentially associated with an increased risk of intracranial hypertension. In particular, the use of positive end-expiratory pressure, recruitment maneuvers, prone positioning, and protective lung ventilation can have undesirable effects on cerebral ph

Investigating the effect of carbon source on rabies virus glycoprotein production in Pichia pastoris by a transcriptomic approach.

Several factors affect protein expression in Pichia pastoris, one among them is the carbon source. In this work, we studied the effect of this factor on the expression level of rabies virus glycoprotein (RABV-G) in two recombinant clones harboring seven copies of the gene of interest. The expression was driven either by the constitutive glyceraldehyde-3-phosphate dehydrogenase (GAP) promoter or the inducible alcohol oxidase1 (AOX1) promoter. Clones were compared in terms of cell physiology and carbon source metabolism. The transcription levels of 16 key genes involved in the central metabolic

Synthetic gene network restoring endogenous pituitary-thyroid feedback control in experimental Graves' disease.

Graves' disease is an autoimmune disorder that causes hyperthyroidism because of autoantibodies that bind to the thyroid-stimulating hormone receptor (TSHR) on the thyroid gland, triggering thyroid hormone release. The physiological control of thyroid hormone homeostasis by the feedback loops involving the hypothalamus-pituitary-thyroid axis is disrupted by these stimulating autoantibodies. To reset the endogenous thyrotrophic feedback control, we designed a synthetic mammalian gene circuit that maintains thyroid hormone homeostasis by monitoring thyroid hormone levels and coordinating the exp

Development of physiological resistance and its stage specificity in Culex quinquefasciatus after selection with deltamethrin in Assam, India.

The study investigated the development and stage specificity of physiological resistance to insecticides in a colony of Culex quinquefasciatus Say (Diptera: Culicidae) mosquitoes, which are vectors of bancroftian filariasis in India, after selection with deltamethrin. Resistance was selected by exposing the larvae to the concentration of deltamethrin that caused 50% mortality in the tested population (i.e., LC50). Under continuous selection pressure, the LC50 increased steadily in subsequent generations. The estimated LC50 for the F0 generation was 0.409 microg/L; the LC50 first displayed a su

Evidence that exocytosis is driven by calcium entry through multiple calcium channels in goldfish retinal bipolar cells.

Ribbon-containing neurons represent a subset of neural cells that undergo graded membrane depolarizations rather than Na(+)-channel evoked action potentials. Bipolar cells of the retina are one type of ribbon-containing neuron and extensive research has demonstrated kinetically distinct pools of vesicles that are released and replenished in a calcium-dependent manner. In this study, we look at the properties of the fastest pool of releasable vesicles in these cells, often referred to as the immediately releasable pool (IRP), to investigate the relationships between vesicle release and calcium

A PET study following treatment with a pharmacological stressor, FG7142, in conscious rhesus monkeys.

FG7142 is a benzodiazepine partial inverse agonist, which is known as a pharmacological stressor. Several reports demonstrated that FG7142 produced anxiety in humans, non-human primates, and rodents, and impaired working memory in non-human primates and rodents. In this study, we examined the effect of FG7142 on cerebral blood flow and glucose metabolism using positron emission tomography (PET) in conscious rhesus monkeys. Male rhesus monkeys were intramuscularly treated with FG7142 (0.2 or 1.0 mg/kg, n=5, respectively), and regional cerebral blood flow (rCBF) and regional cerebral metabolic r

Differential influence of the lipid peroxidation product 4-hydroxynonenal on the growth of human lymphatic leukaemia cells and human periopherial blood lymphocytes.

The purpose of this study was to compare the influence of physiological and supraphysiological concentrations of 4-hydroxynonenal (HNE), a peroxidation product of omega-6-polyunsaturated fatty acids, on the proliferation of malignant CEM-NKR lymphatic leukaemia cells and normal human lymphocytes (HPBM). Furthermore the growth modulating effect of phytohemagglutinin (PHA) on both cell lines was examined. The effects of HNE were monitored 18 hours and 3 days after incubation, using two different DNA-synthesis assays ([3H]-thymidine- and BrdU- incorporation) and a mitochondrial dehydrogenases act

Renal response to volume expansion: learning the experimental approach in the context of integrative physiology.

We describe a laboratory experience for upper-level science students that provides a hands-on approach to understanding the basics of experimental physiology. A pre-lab, interactive tutorial develops the rationale for this experiment by reviewing the renal and cardiovascular mechanisms involved in the response to extracellular fluid volume expansion. After a hypothesis is stated, an experiment is designed to determine the relative importance of dilution of plasma proteins to the overall renal excretory response following volume expansion with intravenous saline. In the lab, students collect da

Fetal hypothalamic transplants in the third ventricle of the adult rat brain. Correlative scanning and transmission electron microscopy.

Blocks of anterior hypothalamus were transplanted from 19 day-old fetuses of Wistar/Lewis rats into the third ventricle of adult male Brattleboro rats. Physiological changes in graft recipients and in sham-operated animals were monitored daily. Twenty days after surgery, the graft recipients and sham-operated animals were killed and their brains examined by correlative scanning and transmission electron microscopy. Host animals that exhibited both decreased polydipsia and increased urine concentration were found to have viable grafts within the third ventricle. The observed physiological chang

Distinguishing Common Digital Phenotyping and Self-Report Parameters for Monitoring and Predicting Depression: Scoping Review.

BACKGROUND: Digital health interventions incorporating self-management strategies are increasingly used to support individuals in managing depression. These interventions often leverage self-monitoring and passive sensor-based data collection to provide personalized feedback, guiding behavioral change. With the proliferation of smartphones and wearable devices, there is growing potential to continuously collect behavioral and physiological data. However, a major limitation in the field is the lack of consolidated evidence on which specific parameters are most useful for monitoring and predicti

Prediction of acute mountain sickness from overnight pulse oximetry during ascent to Everest Base Camp: a field-based machine learning study.

BACKGROUND: Acute mountain sickness (AMS) affects a large proportion of individuals ascending to high altitude every year. While AMS is not life threatening, if left untreated it can progress to serious illness (e.g., high altitude cerebral oedema). Current assessment tools rely on subjective symptom reporting and do not enable prospective prediction. Objective, field-deployable methods for early risk stratification remain limited. This study evaluated whether physiological biomarkers derived from overnight pulse oximetry can predict subsequent AMS during ascent to Everest Base Camp (EBC). ME

Hierarchical physiological sensitivity of the coral, Orbicella faveolata, to suspended sediment and elevated temperature.

Coral reefs are increasingly exposed to multiple environmental stressors, including ocean warming and elevated turbidity from coastal development, dredging, and sediment resuspension. Understanding how these stressors affect coral physiology is essential for improving sediment management and protecting reef ecosystems. We investigated the short-term physiological responses of the Caribbean reef-building coral, Orbicella faveolata, to suspended sediment and elevated temperature during controlled 72-h laboratory experiments. Coral fragments were first exposed in independent chamber-sump systems

Sex-biased expression of enteroendocrine cell-derived hormones contributes to higher fat storage in Drosophila females.

Enteroendocrine (EE) cells in the Drosophila gut produce and release multiple factors, including Allatostatin A (AstA), Allatostatin C (AstC), neuropeptide F (NPF), tachykinin (Tk), Diuretic hormone 31 (Dh31), Bursicon, CCHamide 1, CCHamide 2, and short neuropeptide F. Collectively, these peptides ensure that physiology (e.g., fat storage, fluid balance) and behavior (e.g., feeding, sleep) are coordinated with environmental factors such as nutrient quantity and quality. Despite notable sex differences in physiology and behavior, it remains unclear whether the regulation and function of these E

Hemin Recapitulates the Labile Iron Pool in the Cellular Fenton Reaction with DNA.

The iron-Fenton reaction in biology is influenced by diverse cellular molecules that coordinate with redox-active ferrous ion. Contemporary research proposed that hydroxyl radical or a ferryl species (HO•/Fe=O2+) are the primary oxidants; however, this was challenged by the observation that physiological bicarbonate redirects the reaction to form carbonate radical anion (CO3•-). Questions remained regarding the roles of O2 concentration, ascorbate, and iron speciation in CO3•- formation. Accordingly, in cellulo studies were conducted under physiological O2 (∼25 μM) with ascorbate replenishment

Salinity strengthens plant-pollinator mutualism through leaf physiological status in grafted tomatoes.

The interaction between rootstock-mediated leaf Na and nutrient concentrations, and the subsequent impact on Fv/Fm and unbalanced C/N are major traits associated with plant growth, yield, and bumblebees' pollinating activity under salinity. Soil and water salinization compromises crop yield by reducing plant growth and reproductive fitness through altering source-sink activities, which in turn, affects pollination ecosystem services. However, while pollination services are barely considered as crop breeding targets, little is known about how stress-induced changes in plants affect pollinators'

Beyond blood gases: diaphragm-guided sequential non-invasive ventilation for COPD with type II respiratory failure.

Chronic obstructive pulmonary disease (COPD) with type II respiratory failure is driven by diaphragm load-capacity imbalance and abnormal central respiratory drive, yet clinical decision-making for sequential non-invasive ventilation (NIV) remains largely dependent on blood gas normalization. This narrative review, informed by a structured literature search of PubMed, Embase, and Cochrane Library (January 2000 to June 2026), synthesizes physiological evidence supporting a shift from blood gas-guided to diaphragm-guided sequential NIV management. We propose a testable, three-tier conceptual fra
