Effects of gestational exposure to individual and mixed PFASs on maternal liver function: evidence from the Wuxi Birth Cohort, animal studies, and toxicogenomic analyses.

Per- and polyfluoroalkyl substances (PFASs) are persistent environmental contaminants with potential hepatotoxicity. Their impact during pregnancy remains poorly characterized. We investigated 601 pregnant women from the Wuxi Birth Cohort, measuring serum concentrations of 23 PFASs and 11 liver function markers. Associations between individual PFASs and liver function markers were evaluated with generalized linear model (GLM) and generalized additive model (GAM), while mixture effects were assessed using weighted quantile sum (WQS) and Bayesian kernel machine regression (BKMR). Independent val

Transcriptional regulation of ubiquitin specific peptidase 2b by farnesoid X receptor and its implications in the pathogenesis of hepatocellular carcinoma.

Farnesoid X receptor (FXR) signaling plays an important role in liver regeneration and carcinogenesis. Dysregulation of FXR signaling is linked to the pathogenesis of hepatocellular carcinoma (HCC) while FXR knockout mice spontaneously develop HCC as they age. Ubiquitin specific peptidase 2b (USP2b) is a deubiquitinating enzyme regulating protein stability and other activities. In our recent study, we found that USP2b was significantly downregulated in subjects with HCC and exhibited both tumor-promoting and tumor suppressive activity in a context-dependent manner. However, the mechanistic lin

Protective efficacy of Met-Ser-Arg against irradiation-related testicular injury†.

Ionizing radiation poses a significant risk to male fertility by inducing testicular damage, yet effective protective strategies remain limited. The tripeptide Met-Ser-Arg (MSA) has demonstrated potential in alleviating radiation-induced oxidative stress and lipid metabolic disorders, prompting investigation into its protective effects on the male reproductive system. Male BALB/c mice were exposed to X-ray irradiation (0.5 Gy/day for 5 days) to establish a testicular injury model, followed by MSA treatment. Testicular structure and function were evaluated through histology, electron microscopy

Oleic acid ameliorates the disrupted blood-brain barrier and neuroinflammation following intracerebral hemorrhage in mice by regulating PPARγ.

BACKGROUND: Oleic acid (OA) is demonstrated to have neuroprotective effects and may improve the therapy of neurological disorders. However, the mechanism of OA impact on blood-brain barrier (BBB) and neuroinflammation following intracerebral hemorrhage (ICH) remains indistinct. Here, we aimed to affirm the neuroprotective influence of OA in ICH and to explore the latent molecular mechanism. METHODS: The male C57BL/6 mice (n = 124) was used for this study. ICH model was induced by intracerebral injection of collagenase. OA (60 mg/kg) was administrated 2 h after ICH. Neurobehavioral tests were

The Ageing Adipose Paradox: Implications for Metabolic Health.

BACKGROUND: Preadipocyte commitment to the adipogenic lineage declines markedly with advancing age, while triglyceride accumulation in hypertrophied existing adipocytes persists or expands. This creates a dissociation between adipogenic capacity and lipid-buffering demand, progressively weakening depot metabolic competence and contributing to systemic insulin resistance. OBJECTIVE: This review examines the molecular mechanisms linking impaired adipose tissue plasticity during ageing to metabolic decline, and appraises therapeutic strategies that may restore adipose progenitor competence or li

Exercise-Induced Hepatic Mitochondrial Reprogramming Across Muscle-Gut-Thyroid Axes in MASLD/MASH.

Metabolic dysfunction-associated steatotic liver disease (MASLD) and its progressive form, metabolic dysfunction-associated steatohepatitis (MASH), represent a growing global health burden driven by complex interactions among hepatic lipid accumulation, insulin resistance, chronic inflammation, and mitochondrial dysfunction. Exercise remains the cornerstone of lifestyle therapy for MASLD/MASH; however, its therapeutic benefits extend well beyond weight reduction and involve coordinated molecular adaptations across multiple organ systems. In this review, we introduce hepatic mitochondrial repro

Dose-Dependent Biphasic Effect of Palmitic Acid on Oligodendrocyte Function: Impacts on Viability, Differentiation, and Myelination.

Palmitic acid (PA), the most abundant saturated fatty acid (SFA) in humans, plays a key role in energy metabolism, membrane synthesis, and signaling. Oligodendrocyte precursor cells (OPCs), which generate mature oligodendrocytes (OLs) forming the myelin sheath, are responsive to metabolic and redox signals. Despite increasing interest in lipid metabolism and mitochondrial dynamics as regulators of OPC fate, the effects of PA remain unclear. This study investigates the biphasic, dose-dependent effects of PA on OPCs using the oligodendrocyte precursor MO3.13 cell line and employs rat organotypic

Hormones, heat, and health: a comprehensive review of sex-based differences in brown and beige fat biology.

This review takes a close look at the biology of brown and beige fat, not just as thermogenic tissues, but as active metabolic organs influenced by sex, hormones, age, and even environment. Brown adipose tissue (BAT) and beige adipocytes differ in their origins, gene expression, and regulation. These differences are especially relevant when considering how they behave in males and females. Across both animal and human studies, females show higher BAT volume and more efficient thermogenic activity. Estrogen, acting mainly through estrogen receptor alpha (ERα), increases uncoupling protein 1(UCP

Hyodeoxycholic acid relieves neuropathic pain by activating farnesoid X receptor signaling.

BACKGROUND: Neuropathic pain, is a chronic condition stemming from nervous system injuries. The farnesoid X receptor (FXR), primarily expressed in the liver and intestines, plays a crucial role in regulating bile acids and lipids metabolism. Emerging evidence suggests that bile acids might influence neural function through FXR modulation. OBJECTIVE: To investigate the specific role of intestinal FXR in the modulation of neuropathic pain. METHODS: A neuropathic pain model was established in mice through ligation of the lumbar 4 spinal nerve (SNL). FXR expression was detected in the distal ile
