In Silico Models Using Simple Molecular Descriptors Predict Placental and Breast Milk Transfer of Cannabinoids from Cannabis sativa.

Despite an increasing interest in the pharmacology of cannabinoids from Cannabis sativa, little is known to date about their ability to cross the placenta and to be secreted into breast milk, and in this study, we sought to fill this gap. In total, 126 phytocannabinoids previously detected in Cannabis sativa were investigated for their transplacental transfer and secretion into breast milk. Placental transport was predicted using novel multiple linear regression (MLR), artificial neural network (ANN), boosted trees (BT), and support vector regression (SVR) models, based on a reference set of 8

Morpho-functional uterine and placental dynamics during establishment and maintenance of canine pregnancy, from early trophoblast invasion to parturition.

The placenta is an important, endocrinologically active organ that forms the interface between the maternal and fetal compartments. Evolutio … t. It also provides, for the first time, new morphological insights into the initiation of invasive placentation in the dog. By linking these mechanisms to clinically relevant disorders of placentation, it emphasizes the translational relevance of canine placentation for studies in other species, including humans.

Marked Third-Trimester Placental Thickening Is Associated with Maternal Infectious, Hormonal, and Metabolic Burden: A Matched Case-Control Study.

Background: Increased placental thickness has been associated with adverse perinatal outcomes and fetal functional and structural abnormalities. However, whether marked third-trimester placental thickening is associated with a distinct maternal clinical profile compared with pregnancies with normal placental thickness remains insufficiently characterized. This study aimed to compare maternal characteristics, comorbidities, medication exposure, infection history, and fetal findings between pregnancies with marked placental thickening, defined as placental thickness ≥70 mm, and gestational-age-m

Development and validation of cell-free RNA-based predictive models for preeclampsia using placental single-cell transcriptomic data.

INTRODUCTION: Preeclampsia (PE) is a serious complication of pregnancy for which reliable early predictive biomarkers are currently lacking. Cell-free RNA (cfRNA) has emerged as a promising non-invasive approach that may enhance clinical management and facilitate earlier diagnosis. METHODS: To investigate PE, we applied single‑cell RNA sequencing (scRNA‑seq) to placental tissues, enabling the identification of differentially expressed genes. Integration of weighted gene co‑expression network analysis (WGCNA) with cfRNA profiles from PE patients revealed two gene modules that showed significan

The Impact of Human Placental Tissue-Derived Xenograft on Bone Healing and Pain Behaviors in a Murine Segmental Bone Defect Model.

INTRODUCTION: Segmental bone defects (SBD), frequently resulting from high-energy trauma like military combat injuries and major accidents, represent a significant clinical challenge because of the loss of a large bone segment and associated soft-tissue injury. These injuries often occur in military settings, where blast and ballistic trauma cause complex injuries requiring advanced reconstructive strategies. Similar injuries also occur in the civilian population in car accidents, falls, and workplace injuries. Existing therapies (autografts, allografts, and biomaterials) have limitations, inc

Exosomes-mediated molecular mechanisms underlying the transformation of ovarian tissue into a functional placental analogue in black rockfish ( Sebastes schlegelii).

Viviparity has independently evolved multiple times in teleosts, leading to diverse modes of maternal nutrient provisioning. In black rockfi … xpression and secretion, which in turn reprograms gene expression and promotes angiogenesis-related placentation. Together, these results elucidate the molecular underpinnings of ovarian placentation in black rockfish and offer valuable perspectives on the oviparity-to-viviparity transition in fish evolution.

Predictive Value of Inflammatory and Placental-Related Markers for Adverse Neonatal Outcomes in Pregnant Women With Gestational Diabetes Mellitus and Preeclampsia.

AIMS/BACKGROUND: The coexistence of gestational diabetes mellitus (GDM) and preeclampsia (PE) significantly increases the risk of adverse neonatal outcomes. However, effective predictive tools for this high-risk population remain limited. This study aimed to evaluate the predictive value of inflammatory and placental-related biomarkers for adverse neonatal outcomes in pregnant women with GDM and PE. METHODS: This retrospective observational study enrolled 230 pregnant women with GDM and PE between February 2022 and February 2025. Participants were divided into an event group (n = 53) and a no

Maternal Rumen-Protected Leucine Supplementation Enhances Placental Nutrient Transport Capacity and Increases Birth Weight in Hu Sheep.

Leucine, an essential branched-chain amino acid, serves not only as a substrate for protein synthesis but also as a key regulator of placental function and fetal development. This study investigated the effects of dietary supplementation with RP-Leu during late gestation on placental development and offspring performance in Hu sheep. Sixty twin-pregnant ewes at day 80 of pregnancy were randomly assigned to either a control group (fed a basal diet) or an RP-Leu group (fed a basal diet supplemented with 19 g/day RP-Leu). The feeding trial lasted for 60 d. The ewes were slaughtered at day 140 of

Proinflammatory and Antiangiogenic Effects of Gingival Crevicular Fluid on Placental and Immune Cells: Implications for Oxidative Stress and NET Formation in Pregnancy.

PROBLEM: An association between periodontal disease and adverse pregnancy outcomes, with high risk for the mother and the newborn, has been widely documented. So far, the mechanisms linking gingival inflammation to placental dysfunction remain poorly defined. METHODS OF STUDY: In this study, we aimed to investigate whether bioactive inflammatory components in the gingival crevicular fluid are capable of modulating placental and immune cells in vitro and to analyse whether these effects correlate with periodontal parameters. We conducted a cross-sectional study in 37 pregnant women undergoing

Successful perinatal management of a large placental chorio-angioma: a case report demonstrating the effectiveness of a public-private partnership model.

Placental chorio-angiomas are rare benign vascular tumours of the placenta which are usually small and clinically insignificant, but large lesions can cause serious fetal complications such as anaemia, hydrops fetalis and an adverse perinatal outcome. We report the case of a 29-year-old multigravida diagnosed at 28 weeks' gestation with a large placental chorioangioma accompanied by sonographic evidence of fetal anaemia and hydrops fetalis. Despite two intra-uterine transfusions which led to partial improvement, the hydrops persisted, necessitating a preterm caesarean delivery at 31 weeks. The

Ex vivo cytokine responses and metabolic reprogramming are associated with γδ T cell differentiation in Plasmodium falciparum placental malaria.

BACKGROUND: γδ T cells play a key role in modulating immune responses to pregnancy-associated malaria and can enhance vaccine efficacy through their activation and cytotoxic functions. However, the mechanisms guiding γδT cell differentiation in placental malaria (PM) remain poorly understood. We examined ex vivo associations between cytokines, metabolic profiles and γδ T-cell differentiation in women with PM. METHODS: A case-control study including 50 women at delivery (21 PM+, 29 PM-) was carried out. Peripheral, placental intervillous space, and cord blood mononuclear cells were isolated, a

Dissecting placental host-pathogen interactions: Rift Valley fever virus infection in early human trophoblast stem cells.

Rift Valley fever virus (RVFV) is a mosquito-borne Phlebovirus and zoonotic pathogen affecting maternal-fetal health. Vertical transmission is linked to miscarriage and severe fetal outcomes, but mechanisms of placental pathogenesis remain unclear. We used first-trimester human trophoblast stem cells (hTSCs) to model infection at the maternal-fetal interface. Immunofluorescence, qRT-PCR, western blotting, and single-cell transcriptomics showed that hTSCs are highly susceptible to RVFV. Strand-specific viral transcriptomics confirmed the ambisense S segment and revealed preferential transcripti

Placental Dysfunction as a Common Pathway Linking Preeclampsia, Fetal Growth Restriction, and Preterm Birth: Current Evidence and Future Directions.

Preeclampsia, fetal growth restriction (FGR), and preterm birth remain major contributors to maternal and perinatal morbidity and mortality worldwide. Although these conditions are traditionally approached as distinct clinical entities, they frequently coexist and share overlapping risk factors, suggesting a common underlying pathophysiology. Increasing evidence indicates that placental dysfunction represents a central biological substrate linking these adverse pregnancy outcomes. Impaired placentation, inadequate spiral artery remodeling, and resultant uteroplacental hypoperfusion initiate a …

Hypoxia-induced metabolic shifting and AHR/BCRP axis impairment drive placental dysfunction in early-onset preeclampsia and FGR.

BACKGROUND: Placental hypoxia and vascular dysfunction are hallmarks of preeclampsia (PE). The Breast Cancer Resistance Protein (BCRP) is critical for maintaining the placental barrier against oxidative stress. We investigated whether the hypoxic microenvironment drives BCRP dysregulation and how this correlates with fetal growth restriction (FGR) in severe preeclampsia. METHODS: This case-control study enrolled 59 normotensive and 72 preeclamptic pregnancies to compare clinical phenotypes and placental BCRP expression across distinct gestational age subgroups. Mechanistic investigations were

Placental hexokinase activity is related to fetal blood flow distribution in a cross-sectional study of healthy human pregnancies at term.

OBJECTIVE: Given that placental hexokinase is a key enzyme in glucose metabolism, we hypothesized that placental hexokinase activity is related to fetal blood flow distribution in healthy pregnancies with scheduled cesarean delivery at term. METHODS: We included sixty-seven healthy pregnant women at term. Blood flow in the maternal uterine artery, umbilical vein, and ductus venosus was measured by Doppler ultrasound. Blood samples from the maternal radial artery and uterine vein, and umbilical artery and vein were collected at scheduled cesarean section, and glucose and insulin concentrations

Limited passage and functional effects of polystyrene micro- and nanoplastics in a physiologically-relevant in vitro human placental co-culture model.

The placenta plays a crucial role during pregnancy, yet effective in vitro models for placental toxicity testing are limited. In this study, a Transwell co-culture model combining BeWo b30 and HUVEC cells was developed and characterized, and used to study the transport and effects of polystyrene micro- and nanoplastics (PS-MNPs). After 72 h, 8.7 % of 50 nm and 1.2 % of 200 nm of fluorescent (F)PS-MNPs were detected on the basolateral side, while 1000 nm FPS-MNPs were undetectable. Confocal microscopy showed the uptake of 50 and 200 nm FPS-MNPs by the BeWo b30 and HUVEC cell layer, whereas the

Sex-specific DNA methylation changes in placental thyroid hormone regulatory genes following prenatal exposure to flame retardants.

Prenatal exposure to organophosphate esters (OPEs) and polybrominated diphenyl ethers (PBDEs) has been linked to disrupted fetal thyroid hormone (TH), though the underlying mechanisms remain unclear. Based on the S-MBCS cohort, we analyzed OPE metabolite concentrations in maternal urine during early pregnancy and PBDE levels in cord plasma. Methylation of five TH regulatory genes, namely deiodinase type 3 (DIO3), solute carrier family 16 member 2 (SLC16A2), solute carrier organic anion transporter family member 1C1 (SLCO1C1), thyrotropin-releasing hormone (TRH), and transthyretin (TTR), was qu

Rat-Specific Expression of Placental MATE1 Contributes to Species Difference in Fetal Transfer of Metformin and 1-Methyl-4-Phenylpyridinium.

Metformin readily crosses the human placenta. However, the fetal-to-maternal plasma concentration ratio (F/M ratio) of metformin has been reported to be as low as 0.057 in rats. This study investigated the underlying mechanisms of these interspecies differences by examining the expression and function of candidate transporters, organic cation transporter 3 (OCT3/SLC22A3) and multidrug and toxin extrusion protein 1 (MATE1/SLC47A1), in the placenta across species. Expression analysis at gene and protein levels using RNA-sequencing and absolute quantification by LC-MS/MS revealed minimal species
