Lysine hydroxylase 1 (PLOD1) regulates glucose metabolism and promotes Th17 cell differentiation in psoriasis.

Psoriasis is a common and chronic inflammatory skin disease. To treat psoriasis, more effective molecular targets still need to be developed and the mechanisms elucidated. PLOD1, as one of the genes related to glycolysis, can promote the glycolysis of cells. However, it is not clear whether upregulation of PLOD1 plays a causal role or is directly involved in psoriasis. Herein, we have examined the PLOD1 expression in patients with psoriasis and analyzed its effect on the progression of psoriasis. We found that PLOD1 is highly expressed in psoriasis. Also, the role of PLOD1 in promoting Th17 ce…

Cathelicidin-HG alleviates psoriasis by targeting glycoprotein VI to inhibit platelet-neutrophil complexes formation.

Psoriasis, a chronic inflammatory skin disorder, is characterized by a complex interplay among immune cells. Although the specific mechanisms of platelet involvement in psoriasis are not fully elucidated, platelet activation is considered a major pathogenic factor. In this study, we investigated the role of Cathelicidin-HG (Cath-HG), a peptide derived from Hylarana guentheri skin that exhibits potent inhibitory activity against platelet glycoprotein VI (GPVI), in an imiquimod-induced psoriasis mouse model. Our experimental findings demonstrated that Cath-HG significantly alleviates psoriasis s…

Interleukin-17A-Related Inflammation Mediates Cardiac Injury in Chronic Relapsing Psoriasis-Like Mouse Model.

Psoriasis is an inflammatory disease characterised by chronic recurrent relapses. Previous observational studies have shown that patients with psoriasis are predisposed to cardiovascular comorbidities, but few studies have investigated the impact of psoriasis-related chronic inflammation on cardiac function. In this study, we used imiquimod (IMQ) to establish psoriasis-like mouse models with short-term inflammation (IMQ-ST) or long-term repeated inflammation (IMQ-LT), to mimic acute or chronic recurrent pathophysiology of psoriasis inflammation. The inflammatory pattern in the hearts of IMQ-ST…

Elevated Cutaneous Interleukin-21 Links Eczematous Eruption to Interleukin-17A Inhibitor Treatment in Psoriasis.

BACKGROUND: Eczematous eruption (EE) is an adverse effect observed in psoriasis patients undergoing interleukin (IL)-17A inhibitor therapy, with reported incidence rates ranging from 2.2% to 12.1%. In some cases, this reaction leads to discontinuation of treatment. However, the underlying mechanism of EE development remains unclear. Therefore, we aimed to elucidate the pathogenesis of EE associated with anti-IL-17A treatment and identify pathogenic molecules involved. METHODS: Skin samples were collected from psoriasis patients both before and after anti-IL-17A treatment, and the treated skin…

Leukocytes have a heparan sulfate glycocalyx that regulates recruitment during psoriasis-like skin inflammation.

The glycocalyx is a proteoglycan-rich layer present on the surface of all mammalian cells and is particularly prevalent on endothelial cells lining the vasculature. The glycocalyx is thought to affect leukocyte migration by masking adhesion molecules and reducing leukocyte adhesion to the endothelium. Leukocyte recruitment is a key driver of inflammatory diseases, including psoriasis. Here, we found that leukocytes had the glycocalyx component heparan sulfate on their cell surface and that it was lost in response to psoriasis-like skin inflammation. In contrast, endothelial heparan sulfate was…

Light-emitting diode red light attenuates epidermal thickening and keratinocyte proliferation in psoriasis models.

Psoriasis is an immune-mediated skin condition that impacts approximately 3% of the United States population, with limited adjunctive therapy options. This study investigates the effects of light emitting diode red light (LED-RL) as a potential adjunct. Using in vitro keratinocytes, three-dimensional (3D) recapitulated skin models, and a mouse model, LED-RL's impact on keratinocyte proliferation and epidermal thickness was assessed. Our findings show that LED-RL significantly reduced keratinocyte proliferation without inducing apoptosis, likely through cell cycle modulation and decreased phosp…

Inflammation mediation of the association between brominated flame retardants and psoriasis among U.S. adults.

BACKGROUND: Brominated flame retardants (BFRs), widely applied in fire prevention, are persistent environmental pollutants. Accumulating evidence indicates that BFRs can trigger inflammatory responses in humans, potentially contributing to various diseases. Given the central role of inflammation in psoriasis pathogenesis, investigating the association between BFR exposure and psoriasis risk is of considerable importance. METHODS: We analyzed data from the 2005-2006 and 2009-2014 cycles of the National Health and Nutrition Examination Survey (NHANES). concentrations of BFRs were natural logari…

Eco-conscious TLC assay for dual psoriasis drugs in plasma: a green analytical approach.

Psoriasis is one of the dermatological autoimmune diseases that involve cracking, redness, bleeding and inflammation on the surface of the skin. Sulfasalazine (SUL) is an immunity suppressing and anti-phlogistic drug. Pentoxifylline (PTN) is an immunosuppressant and vasodilator. So, the two drugs are co-administered together in the treatment protocol for psoriasis. No chromatographic analytical method was developed in the literature for the quantitative determination of SUL and PTN in their binary mixture and spiked human plasma. So, this work's goal is to establish an environmentally friendly…

Mortality, prevalence, comorbidity, and flare patterns in generalised pustular psoriasis: a comprehensive literature review.

Generalised pustular psoriasis (GPP) is a rare, chronic, inflammatory skin disease that may manifest as a relapsing disease with recurrent flares or a persistent disease with intermittent flares. GPP is associated with a significant clinical burden, including pain, fatigue, and comorbidities, leading to severe, life-threatening complications. The frequency and severity of GPP flares have not been characterised comprehensively in epidemiological studies. Comprehensive literature searches were conducted to identify studies evaluating mortality, prevalence, comorbidities, and flares in the GPP po…

Elevated SNHG15 empowers keratinocytes hyperproliferation through activation of STAT3/Cyclin D1 axis in psoriasis.

Psoriasis is a common inflammatory skin disease with characterization of epidermal hyperplasia and sustained skin inflammation. Long noncoding RNAs (lncRNAs), which contain more than 200 nucleotide-long transcripts, are emerging as the crux of epigenetic regulators in multiple biological processes and diseases. However, how lncRNAs contribute to the etiology of psoriasis remains to be elucidated. For the first time, this study revealed that SNHG15, which was elevated in cytokines-stimulated keratinocytes and psoriasis lesions, promoted keratinocytes hyperproliferation. Mechanistically, SNHG15 …
