Vascular Contributions to Healthy Aging and Dementia.

Vascular pathologies are among the most common contributors to neurodegenerative changes across the spectrum of normal aging to dementia. Cerebral small vessel disease (SVD) encompasses a wide range of conditions affecting capillaries, small arteries, and arterioles, as well as perivascular spaces and fluid dynamics in the brain, playing a significant role in vascular contributions to cognitive impairment and dementia (VCID). These factors can accelerate the progression of SVD and neuronal degeneration. Since aging is the primary risk factor for Alzheimer's disease (AD) and AD-related dementia

Interaction of oat bran and exercise training improved exercise adaptability via alleviating oxidative stress and promoting energy homeostasis.

Skeletal muscle performance is influenced by both diet and the mode of exercise, with diet playing a crucial role in individuals' adaptation to exercise training. Our study investigated the interaction of oat bran (OB) diet and moderate intensity exercise training (MIET) on skeletal muscle function and athletic performance. Studies have reinforced the positive association of high-fat diet (HFD) with chronic systemic inflammation and corresponding peripheral skeletal muscle dysfunction during exercise training. OB could alleviate the inflammation, oxidative stress, and energy homeostasis disord

Multi-timescale neural dynamics for multisensory integration.

Carrying out any everyday task, be it driving in traffic, conversing with friends or playing basketball, requires rapid selection, integration and segregation of stimuli from different sensory modalities. At present, even the most advanced artificial intelligence-based systems are unable to replicate the multisensory processes that the human brain routinely performs, but how neural circuits in the brain carry out these processes is still not well understood. In this Perspective, we discuss recent findings that shed fresh light on the oscillatory neural mechanisms that mediate multisensory inte

Glycosylation: mechanisms, biological functions and clinical implications.

Protein post-translational modification (PTM) is a covalent process that occurs in proteins during or after translation through the addition or removal of one or more functional groups, and has a profound effect on protein function. Glycosylation is one of the most common PTMs, in which polysaccharides are transferred to specific amino acid residues in proteins by glycosyltransferases. A growing body of evidence suggests that glycosylation is essential for the unfolding of various functional activities in organisms, such as playing a key role in the regulation of protein function, cell adhesio

The spermatheca ultrastructure of the ground beetle Clinidium canaliculatum (Costa) (Carabidae, Rhysodinae).

The ground beetle Clinidium canaliculatum is a member of Rhysodinae, a taxon with still discussed systematic position. The spermatheca of this species is a small cylindrical structure connected to the common oviduct by a thin duct. The ultrastructure of the organ has revealed that the apical receptacle is provided with an epithelium lined by a thick cuticle from the deeper region of which several finger-like cuticular structures extend into the cytoplasm. On these structures adhere microtubule bundles that cross the whole cytoplasm to anchor on short densities along the basal plasma membrane.

Myo-Inositol and Its Derivatives: Their Roles in the Challenges of Infertility.

Myo-inositol (MYO) and D-chiro-inositol (DCI) are the two most significant isomeric forms of inositol, playing a critical role in intracellular signaling. MYO is the most abundant form of inositol in nature; DCI is produced from MYO through epimerization by an insulin-dependent enzyme. Recently, it has been demonstrated that inositol may influence oocyte maturation and improve intracellular Ca2+ oscillation in the oocytes, and it has been proposed as a potential intervention for restoring spontaneous ovulation. The MYO concentration in human follicular fluid is considered a bioindicator of ooc

Discovery of Triketone-Indazolones as Novel 4-Hydroxyphenylpyruvate Dioxygenase Inhibiting-Based Herbicides.

4-Hydroxyphenylpyruvate dioxygenase (HPPD) is a crucial herbicide target in current research, playing an important role in the comprehensive management of resistant weeds. However, the limited crop selectivity and less effectiveness against grass weeds of many existing HPPD inhibitors, limit their further application. To address these issues, a series of novel HPPD inhibitors with fused ring structures were designed and synthesized by introducing an electron-rich indazolone ring and combining it with the classical triketone pharmacophore structure. The cocrystal structure of representative com

An invariant C-terminal tryptophan in McdB mediates its interaction and positioning function with carboxysomes.

Bacterial microcompartments (BMCs) are widespread, protein-based organelles that regulate metabolism. The model for studying BMCs is the carboxysome, which facilitates carbon fixation in several autotrophic bacteria. Carboxysomes can be distinguished as type α or β, which are structurally and phyletically distinct. We recently characterized the maintenance of carboxysome distribution (Mcd) systems responsible for spatially regulating α- and β-carboxysomes, consisting of the proteins McdA and McdB. McdA is an ATPase that drives carboxysome positioning, and McdB is the adaptor protein that direc

Acute and Longitudinal Effects of Sport-related Concussion on Reactive Balance.

Postural instability is a common observation after concussions, with balance assessments playing a crucial role in clinical evaluations. Widely used post-concussion balance tests focus primarily on static and dynamic balance, excluding the critical aspect of reactive balance. This study investigated the acute and longitudinal effects of concussion on reactive balance in collegiate athletes. The assessments were conducted at pre-season baseline and 4 post-concussion timepoints: acute, pre-return-to-play, post-return-to-play, and six months post-concussion. The instrumented-modified Push and Rel

Usnic Acid Derivatives as Multi-Target Anti-Alzheimer's Disease Agents: Design, Synthesis, X-Ray Single Crystal Structure of Zn(II) Complex and Biological Activities.

Alzheimer's disease (AD) is multifactorial, which makes the design of multi-target-directed ligands an attractive strategy for the development of anti-AD drugs. In order to enhance the anti-AD effects and reduce the toxicity, two usnic acid (UA) derivatives (1-2) were designed, synthesized and fully characterized by introducing dimethylamine Schiff base moiety into the toxic "triketone" portion. Ellman's method and molecular docking were used to test the cholinesterase inhibitory activities. Antioxidant activities were studied with Fenton reaction, cyclic voltammetry and C. elegans. The result

Epigenetic Alteration in Colorectal Cancer: Potential Diagnostic and Prognostic Implications.

Colorectal cancer (CRC), a prevalent malignant tumor of the digestive system, ranks as the third and second in global incidence and mortality, respectively, in 2020, with 1.93 million new cases (≈10% of all cancers). There are 940,000 deaths (≈9.4% of all cancers), and the incidence of CRC in younger patients (under 50 years of age) has become a new trend. The pathogenesis of CRC is primarily attributed to a series of genetic and epigenetic abnormalities within normal colonic epithelial cells, coupled with the reshaping of the tumor microenvironment in the surrounding stroma. This process lead

Probing methylmercury photodegradation by different fractions of natural organic matter in water: Degradation kinetics and mercury isotope fractionation characteristics.

Recent advancements in mercury (Hg) isotopic fractionation research have evolved from conceptual demonstrations to practical applications. However, few studies have focused on revealing fractionation fingerprinting for aqueous methylmercury (MeHg) photodegradation due to its sensitivity to natural organic matter (NOM). Here, the impact of NOM fractions with varying chemical properties on MeHg photodegradation kinetics and Hg isotope fractionation characteristics was investigated. Findings reveal that reduced NOM, containing alcohol/phenol groups, slows the degradation rate compared to the oxid

Light Pollution and Oxidative Stress: Effects on Retina and Human Health.

Visible light refers to the frequencies within the electromagnetic spectrum that humans can see, encompassing radiation with wavelengths falling between 380 nm to 760 nm. The energy of a single photon increases with its frequency. In the retina, photoreceptor cells contain light-sensitive pigments that absorb light and convert it into electrical stimuli through a process known as phototransduction. However, since the absorption spectrum of photoreceptors closely aligns with blue light (ranging from 400 to 500 nm), exposure to high light intensities or continuous illumination can result in oxid

Remyelination-oriented clemastine treatment attenuates neuropathies of optic nerve and retina in glaucoma.

As one of the top causes of blindness worldwide, glaucoma leads to diverse optic neuropathies such as degeneration of retinal ganglion cells (RGCs). It is widely accepted that the level of intraocular pressure (IOP) is a major risk factor in human glaucoma, and reduction of IOP level is the principally most well-known method to prevent cell death of RGCs. However, clinical studies show that lowering IOP fails to prevent RGC degeneration in the progression of glaucoma. Thus, a comprehensive understanding of glaucoma pathological process is required for developing new therapeutic strategies. In
