Targeting Brain Plasticity: Vagal Nerve Stimulation as a Therapy for Autism-Like Symptoms in a Valproic Acid Mouse Model.

Autism spectrum disorder (ASD) is a multifactorial neurodevelopmental condition defined by social deficits, stereotypical or repetitive behaviors, and anxiety. This study evaluates the therapeutic potential of transauricular vagal nerve stimulation (tVNS) in a valproic acid (VPA)-induced mouse model of ASD. The study comprised three groups: the control + sham (saline-treated offsprings receiving sham stimulation), the autistic + sham (VPA-treated offspring receiving sham stimulation), and the autistic + tVNS (VPA-treated offsprings receiving tVNS). Male C57BL/6 mice exposed to VPA on embryonic

Rethinking abortion law reform in Malawi: a multi-method study.

Despite decades of sustained advocacy, Malawi retains one of the world's most restrictive criminal abortion laws. A draft Termination of Pregnancy Bill, developed in 2015 to propose semi-liberal reform to Malawi's abortion law, has yet to be enacted. In contrast, similar or less intensive advocacy efforts have resulted in liberalisation of abortion laws in other low- and middle-income countries (LMICs), including Zambia, South Africa, and Ethiopia. This qualitative case study examines the policy processes adopted by advocates and identifies potential pathways for abortion law reform in Malawi.

Weight stigma from close others may drive reduced adherence to physical activity and nutrition recommendations during pregnancy and postpartum.

PROBLEM: Weight stigma, defined as negative misconceptions and stereotypes associated with weight, is a social issue in pregnancy and postpartum that can impair engagement in healthy behaviours. BACKGROUND: Previous studies have not tested the behavioural implications of weight stigma on integral factors that promote maternal and newborn wellbeing, including physical activity (PA) and nutrition. AIM: To evaluate the relationship between experiencing weight stigma from close others (i.e., family, friends, partners) in pregnancy and postpartum and adherence to PA and nutrition recommendations.

Making Sense of Consent: A Qualitative Analysis of Australian Mock Juror Decision-Making in a Rape Trial.

Jurors in rape trials often rely on stereotypical expectations about how a "real" victim-survivor should behave. Drawing on the Story Model for juror decision-making, this study examined how mock jurors constructed narrative accounts of consent, credibility, and responsibility in an alcohol-related acquaintance rape case. A total of N = 250 jury-eligible Australian community members read a written trial transcript and provided verdicts accompanied by open-ended explanations of their reasoning. These responses were analysed using a hybrid reflexive thematic analysis, combining an initial induct

A species rules syntax model accurately organizes birdsong syllables into songs.

Birdsong is a rare example of learned vocal communication. Like speech, song consists of acoustic units (syllables) that are learned from adults and organized into temporal sequences (syntax) during development. Tutors' and pupils' syllables and syntax are often the same, suggesting that both song features are learned. Syntax is highly species-specific and consistent across individuals and populations of the same species, unlike syllables. Here, we hypothesized that song syntax can be accurately predicted by computational models based on the relationship between syllable acoustics and order th

Maternal anti-collapsin response mediator protein 1 antibody inducing autism-like behaviors in offspring.

Maternal autoantibodies targeting neural proteins represent a major environmental contributor to autism spectrum disorder (ASD). Although anti-collapsin response mediator protein 1 (CRMP1) antibodies (Abs) have been detected in mothers of ASD children, the pathogenic role of anti-CRMP1 Ab in ASD remains unclear. Here, we evaluated the ability of maternal anti-CRMP1 Ab to induce pathology in a mouse model. Using a cell-based assay, we found that maternal anti-CRMP1 Ab persisted throughout gestation and was detected in offspring serum on postnatal day 16. Exposed offspring exhibited delayed deve

Deep learning based approach for Behavior classification in diagnoses of Autism Spectrum Disorder using naturalistic videos.

Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder that is marked by a lack of communication skills in social situations and repetitive and stereotypical Behaviors. The most widespread form of diagnosing ASD among children is based on psychological screening test along with monitoring of the Behavioral pattern, especially repetitive Behaviors. Some of these Behaviors include hand-flapping, head banging and spinning which are common among ASD children. In our research, we examine abnormal Behavioral patterns that may reflect ASD through the videos of children engaged in the everyda

Analysis of intrinsic connectivity in a multifunctional central pacemaker nucleus in vertebrates.

Gymnotiform fish emit electric organ discharges (EODs) for both active electroreception and electrocommunication. In pulse-type species, EODs are generated by a hierarchical electromotor network controlled by a medullary pacemaker nucleus (PN), which comprises intrinsic pacemaker cells (PM-cells) and projecting relay cells (R-cells). Active electroreception requires the emission of stereotyped EODs, an electromotor output that implies a functional PN configuration based on the synchronous rhythmic discharge of PM-cells, a high-safety-factor feedforward transmission to R-cells, and the subseque

Primitive reflexes as candidate quantitative readouts of hierarchical inhibitory control across the lifespan.

Primitive reflexes are typically inhibited during normal neurodevelopment as cortical and subcortical inhibitory systems mature. However, persistent or incompletely integrated primitive reflexes have been observed in neurodevelopmental conditions including attention-deficit/hyperactivity disorder (ADHD), dyslexia, developmental coordination difficulties, sensory-processing-related presentations, and delayed speech and language development. Re-emergence of primitive reflexes has also been reported in aging and neurological disorders such as Alzheimer's disease and Parkinson's disease. Based on

Time to reframe osteoporosis: a position statement to characterize the osteoporosis care gap.

Osteoporosis, a condition marked by increased fracture risk, remains under-diagnosed and under-treated worldwide, resulting in a substantial "treatment gap"-the difference between those eligible for osteoporosis treatment and those who actually receive it. While the concept of closing the treatment gap is commendable, and has galvanized clinical and policy efforts, this position statement argues that the prevailing narrative is in danger of becoming disease-focused and parentalistic, neglecting person-centered care. An international consensus group, including public contributors with lived exp

DPP-4 inhibitor alleviates gut-brain axis pathology in Parkinson's disease.

BACKGROUND: Dipeptidyl peptidase-4 inhibitors (DPP-4is) have been reported to exhibit therapeutic effects in Parkinson's disease (PD), increasing their potential for drug repurposing. One aspect of PD pathogenesis is thought to be associated with the gut-brain axis, where α-synuclein from the gut is transmitted to the brain via the vagus nerve (VN). OBJECTIVE: We explored whether sitagliptin, a DPP-4i, exhibits a protective effect in a low-dose rotenone-treated gut-brain axis-associated PD model. DESIGN: To explore the effect of sitagliptin, we used the oral rotenone-treated mouse model, whi

Pervasive chromosomal instability drives the karyotypic evolution of hypodiploid tumours.

BACKGROUND: Tumours frequently exhibit extreme levels of aneuploidy. While increases in ploidy are well-characterised, the opposite phenomenon-extensive chromosome loss leading to hypodiploidy-remains underexplored. METHODS: Here, we analyse over 17,000 cancer genomes from 34 cancer types and perform a pan-cancer analysis of karyotypic evolution in hypodiploid tumours. We develop methods to identify current and former hypodiploid tumours, analyse predictors of chromosome loss patterns across tissues, and characterise the relationship between hypodiploidy and other forms of chromosomal instabi

Examining the Effect of Volitionally Altering Fundamental Frequency on Measures Associated With Vocal Hyperfunction.

Speech-language pathologists who work with transgender and nonbinary individuals are often concerned about introducing vocal hyperfunction (VH) while supporting the use of a more gender-congruent voice. The current study examined the effects of volitionally altering fundamental frequency (fo) on measures associated with VH. Forty (20 female, 20 male) speakers with no vocal complaints and who did not identify as transgender were recruited. Speakers were recorded in two conditions: habitual fo and altered fo. In the altered fo condition, speakers volitionally altered their fo by two semitones in

Ophthalmological Findings and Their Clinical Correlations in Children With Autism Spectrum Disorder: A Comparative Study.

ObjectiveAutism spectrum disorder (ASD) can affect vision both directly through central nervous system involvement and indirectly by complicating ophthalmological examinations due to limited eye contact and poor cooperation. Visual impairments may exacerbate social interaction difficulties and affect motor development in individuals with ASD. Previous studies reported ophthalmological involvement in 27-71% of children with ASD, including refractive errors, strabismus, and amblyopia. The purpose of this study was to investigate visual impairments in children with ASD and examine their relations

Unacknowledged Burdens and Clinical Assets of BIPOC Genetic Counseling Students: Qualitative Evidence to Inform Supervision.

As the genetic counseling profession works to diversify its predominantly white workforce, understanding the experiences of Black, Indigenous, and People of Color (BIPOC) students is central to equity efforts. While BIPOC students bring invaluable cultural and linguistic diversity that improves patient care, they often navigate clinical training environments that lack diversity and psychological safety. This article draws on data from a longitudinal constructivist qualitative study to examine how racial and ethnic concordance (or lack thereof) with patients and clinical supervisors influenced

Non-allometric expansion and enhanced compartmentalization of Purkinje cell dendrites in the human cerebellum.

Purkinje cell (PC) dendrites are optimized to integrate the vast cerebellar input array and drive the sole cortical output. PCs are classically seen as stereotypical computational units, yet mouse PCs are morphologically diverse and those with multi-branched structure can receive non-canonical climbing fiber (CF) multi-innervation that confers independent compartment-specific signaling. While otherwise uncharacterized, human PCs are universally multi-branched. Do they exceed allometry to achieve enhanced integrative capacities relative to mouse PCs? To answer this, we used several comparative

Shear Force Cropping Organic Molecular Crystals Based on Adaptive Hydrogen Bonding Network Reconstructions.

Organic molecular crystals are stereotypically considered brittle, fragile, and prone to disintegration, especially when under anisotropic shear forces. However, robust intermolecular interactions and subtle molecular packing motifs may enable a crystal lattice with extraordinary mechanical properties. Here, we present an organic molecular crystal composed of a chiral dithiolane derivative (R-TAE) that can be cropped and cut into arbitrary shapes by external shear forces. Macroscopic R-TAE crystals prepared through a three-solvent diffusion method can be mechanically torn apart by tweezers or
