Alcohol dehydrogenase 3 and risk of esophageal and gastric adenocarcinomas.

OBJECTIVES: Alcohol increases esophageal squamous carcinoma risk but has been less consistently associated with esophageal adenocarcinoma. Alcohol dehydrogenase catalyzes the oxidation of approximately 80% of ethanol to acetaldehyde, a carcinogen. The alcohol dehydrogenase gene has several polymorphisms which may lead to faster conversion of ethanol to acetaldehyde, which may increase cancer risk. METHODS: We undertook a study to examine whether a common polymorphism in the alcohol dehydrogenase 3 gene was associated with a higher risk of esophageal adenocarcinoma using data and biological sa

Probiotics for non-alcoholic fatty liver disease and/or steatohepatitis.

BACKGROUND: Non-alcoholic fatty liver disease comprises a spectrum of diseases ranging from simple steatosis to non-alcoholic steatohepatitis, fibrosis, and cirrhosis. Probiotics have been proposed as a treatment option because of their modulating effect on the gut flora that could influence the gut-liver axis. OBJECTIVES: To evaluate the beneficial and harmful effects of probiotics for non-alcoholic fatty liver disease and/or steatohepatitis. SEARCH STRATEGY: We searched The Cochrane Hepato-Biliary Group Controlled Trials Register (July 2006), the Cochrane Central Register of Controlled Tri

Exposure to the taste of alcohol elicits activation of the mesocorticolimbic neurocircuitry.

A growing number of imaging studies suggest that alcohol cues, mainly visual, elicit activation in mesocorticolimbic structures. Such findings are consistent with the growing recognition that these structures play an important role in the attribution of incentive salience and the pathophysiology of addiction. The present study investigated whether the presentation of alcohol taste cues can activate brain regions putatively involved in the acquisition and expression of incentive salience. Using functional magnetic resonance imaging, we recorded BOLD activity while delivering alcoholic tastes to

Morphometry of dorsal raphe nucleus serotonergic neurons in alcoholism.

BACKGROUND: Reduced serotonergic function is hypothesized in alcohol abuse and dependence. Serotonergic innervation of the cortex arises predominantly from the dorsal raphe nucleus (DRN). We sought to determine the number and morphometric characteristics of DRN serotonergic neurons postmortem in alcoholic individuals (n=9; age: 16-66 years; 8M:1F) compared with psychiatrically normal, nonalcoholic controls (n=6; age: 17-74 years; 4M:2F). METHODS: Brainstems were collected at autopsy, fixed and cryoprotected. Alcohol dependence or abuse was determined by psychological autopsy (DSM-IV), the pre

Estradiol valerate and alcohol intake: dose-response assessments.

BACKGROUND: An injection of estradiol valerate (EV) provides estradiol for a prolonged period. Recent research indicates that a single 2.0 mg injection of EV modifies a female rat's appetite for alcoholic beverages. This research extends the initial research by assessing 8 doses of EV (from .001 to 2.0 mg/female rat), as well assessing the effects of 2.0 mg EV in females with ovariectomies. RESULTS: With the administration of EV, there was a dose-related loss of bodyweight reaching the maximum loss, when it occurred, at about 4 days after injections. Subsequently, rats returned to gaining wei

Interaction of alcoholic extracts of hops with pentobarbital and diazepam in mice.

The interaction of alcoholic extracts of Magnum, Aroma and wild genotype hops with drugs that lower the activity of the central nervous system (CNS) was studied in mice. Hops drying and preparation of extracts were performed according to standard pharmacological procedures for preparing total alcoholic extracts of medicinal plants, i.e. in a ratio of one part dry herbs to two parts of 70% alcohol, with evaporation to dryness so that the extracts no longer contained any alcohol. The mice received four doses intraperitoneally (i.p.) of 0.5% aqueous solutions of the above-mentioned extracts, whic

Alcohol dependence patterns and their impact on New York City.

As a follow-up to a national survey by the Community Anti-Drug Coalitions of America (CADCA), an expert panel was convened to discuss local alcoholism data from New York, NY. The impact of alcoholism on the US economy is estimated at $185 billion annually. About 15% of New Yorkers drink alcohol excessively, with the highest percentage in Manhattan (22%). Over 135,000 New York City residents are admitted for alcohol treatment. Alcohol dependency is highest among whites (21%), followed by Latinos (14.8%) and African Americans (11%). Despite lower levels of excessive drinking, African-American an

Impaired eyeblink conditioning in children with fetal alcohol syndrome.

BACKGROUND: Eyeblink conditioning (EBC) is a Pavlovian paradigm that involves contingent temporal pairing of a conditioned stimulus (e.g., tone) with an unconditioned stimulus (e.g., air puff). Animal studies have shown that binge consumption of alcohol during pregnancy impairs EBC and that this impairment is likely mediated by a loss of neurons in the inferior olive and the cerebellar cortex and deep nuclei, as well as by a reduction in neural plasticity in the cerebellar deep nuclei. METHODS: Short delay EBC was examined in 98 5-year-old children born to women from the Coloured (mixed ances

Novel molecular targets for the prevention of fetal alcohol syndrome.

Alcohol abuse produces damaging effects on the CNS that leads to several types of disorders. When consumed during pregnancy, alcohol may cause craniofacial malformations, growth retardation and brain damage in offspring. These symptoms are grouped by the term fetal alcohol syndrome (FAS). FAS is the most common cause of non-genetic mental retardation in the western world. Substantial efforts to elucidate the molecular basis of these impairments are currently in progress. Whereas FAS is totally preventable by avoiding alcohol intake during pregnancy, efficient therapies to prevent or mitigate t

[Interactions between alcohol and work exposure to chemical substances].

BACKGROUND AND OBJECTIVES: The quite diffused habit of a significant assumption of alcohol drinks, can interfere with the professional exposure to chemical substances. The interaction may result in increasing their toxicity and/or modifying the parameters of the biological monitoring. It may also act as a confounding factor, not only in epidemiologic researches but also at individual level when the assessment of the occupational exposure and/or the diagnosis of an occupational diseases, is under consideration. We review available references in the literature summarizing major scientific eviden
